Summary: Esophageal cancer is a highly lethal disease with rapid progression and poor prognosis. The two major subtypes, esophageal squamous cell carcinoma (ESCC) and esophageal adenocarcinoma (EAC), have distinct risk factors and molecular characteristics. Although surgical and chemoradiotherapy approaches have been applied, molecularly targeted therapy for esophageal cancer is still in its early stages. Advances in large-scale next-generation sequencing have revealed genomic alterations in ESCC and EAC, providing insights into their roles in the development and progression of esophageal cancer. Potential therapeutic targets have been identified, and novel strategies are under development to combat this disease.

Summary: This study analyzed the CNVs, expression, and clinical outcomes of PRMT5 and APE1 genes in oral squamous cell carcinoma (OSCC) patients. The findings showed that PRMT5 and APE1 share almost identical CNV values in cancer tissues, and their high expression levels are associated with poor survival outcomes after radiation therapy. Simultaneous inhibition of PRMT5 and APE1 increases the sensitivity of OSCC cells to radiation therapy.

Summary: This study aimed to identify prognostic markers for esophageal squamous cell carcinoma (ESCC) and build an effective prognostic nomogram. The researchers constructed and verified a nomogram model based on multiple clinicopathologic characteristics and gene expression. They found that the nomogram had higher predictive accuracy for overall survival compared to the American Joint Committee on Cancer seventh edition. Additionally, they identified two marker genes directly related to the prognosis of ESCC.

Summary: Vulvar squamous cell carcinoma (VSCC) is the most common form of vulvar malignancy, and a molecular characterization of VSCC is crucial for diagnosis and treatment. Through genome-wide methylation sequencing, this study identified 199 genes with differential methylation in VSCC, most of which are hyper-methylated and involved in transcription regulator activity, suggesting their importance in VSCC development. Further exploration of the prognostic relevance of these genes in larger cohorts is warranted.

Summary: Oral squamous cell carcinoma (OSCC) has a high incidence in the presence of oral submucous fibrosis (OSF) due to unknown causes. This study aims to identify key genes and explore the molecular mechanisms involved in the conversion of OSF into OSCC. Through analysis of differentially expressed genes (DEGs) between OSCC and OSF, a total of 170 DEGs were identified. Further analyses revealed the functional associations, protein-protein interactions, and potential biomarkers. The overexpression of IRF4 was found to be associated with the progression from OSF to OSCC, and it may serve as a diagnostic marker and contribute to increased immune infiltration in OSCC.
