A label-free MALDI TOF MS-based method for studying the kinetics and inhibitor screening of the Alzheimer's disease drug target beta-secretase

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI TOF MS) is a well-established method with a unique set of qualities including sensitivity, minute sample consumption, and label-free detection, all of which are highly desired in enzyme assays. On the other hand, the application of MALDI TOF MS is usually limited by high concentrations of MS-incompatible compounds in the reaction mixture such as salts or organic solvents. Here, we introduce kinetic and inhibition studies of -secretase (BACE1), a key enzyme of the progression of Alzheimer's disease. Compatibility of the enzyme assay with MALDI TOF MS was achieved, providing both a complex protocol including a desalting step designed for rigorous kinetic studies and a simple mix-and-measure protocol designed for high-throughput inhibitor screening. In comparison with fluorescent or colorimetric assays, MALDI TOF MS represents a sensitive, fast, and label-free technique with minimal sample preparation. In contrast to other MS-based methodological approaches typically used in drug discovery processes, such as a direct injection MS or MS-coupled liquid chromatography or capillary electrophoresis, MALDI TOF MS enables direct analysis and is a highly suitable approach for high-throughput screening. The method's applicability is strongly supported by the high correlation of the acquired kinetic and inhibition parameters with data from the literature as well as from our previous research.

Summary: A novel approach for VOCs analysis based on Au+ ion chemical ionization has been proposed, utilizing a commercially available mass spectrometer with ultrahigh resolving power and sub-ppm mass accuracy. The technique demonstrated excellent performance in analyzing various classes of organic molecules, expanding the available methods for VOCs analysis.

Structure elucidation of multicolor emissive graphene quantum dots towards cell guidance

Summary: GQDs synthesized using a fast microwave-assisted hydrothermal protocol exhibited emission from NUV-Vis to NIR range, with their structure and chemical composition elucidated using advanced NMR and spectroscopy techniques. The observed multicolor red-shifted fluorescence emission in cell imaging demonstrates the potential application of GQDs as bioimaging carriers, along with their non-cytotoxicity confirmed through ATP assay.

Summary: In this study, a novel combination of metal oxide laser ionization mass spectrometry imaging (MOLI MSI) technique with off-line lipid derivatization by ozone was presented for the detection of fatty acids (FA) and their carbon-carbon double bond (C=C) positional isomers in biological tissues. The developed technique allows for the visualization of FA distribution and provides additional information about positional isomers.

Infrared Laser Desorption of Intact Nanoparticles for Digital Tissue Imaging

Summary: This study presents a new technique for digital mapping of biomarkers in tissues using infrared laser ablation and counting intact gold nanoparticle tags. The technique demonstrated successful mapping of a proliferation marker in three-dimensional aggregates of colorectal carcinoma cells, generating sharp distribution maps of the specific biomarker. The developed methodology shows high accuracy and potential applications with multielemental mass spectrometers.

Spontaneous formation of carbon dots helps to distinguish molecular fluorophores species

Summary: Carbon dots (CDs) exhibit similar photophysical features despite different synthesis protocols. Understanding the structure is crucial for achieving desired properties. This study provides insight into the structure and formation of CDs, identifying the presence of molecular fluorophore species. Density functional theory calculations support the spontaneous formation of a compact network of molecular species. Additionally, the study comprehensively investigates the photoluminescence properties, paving the way for CD-based logic gate fluorescent sensors.

Detection of Single Ag Nanoparticles Using Laser Desorption/Ionization Mass Spectrometry

Summary: In this study, the detection of single silver nanoparticles was achieved using subatmospheric pressure laser desorption/ionization mass spectrometry (LDI MS). The authors discussed the sample preparation, measurement conditions, generated ions, and limiting experimental factors. The results showed that 84 to 95% of the deposited 80 nm silver nanoparticles were detected. The LDI MS platform presented in this study provides an alternative method for imaging the distribution of individual nanoparticles and has the potential for multiplexed mapping of low-abundance biomarkers in tissues.
