Summary: This study developed and validated two methods for the determination of clozapine and norclozapine in dried blood spots and dried plasma spots. The results showed a strong correlation between the concentrations in dried samples and serum levels, with higher accuracy in dried plasma spots. This method can be used as an alternative sampling strategy in clozapine therapeutic drug monitoring.

Summary: Indirubin has been identified as an active component in a traditional Chinese herbal mixture and has shown anticancer activity in patients with chronic leukemia. Multiple studies have focused on the mechanisms of its antitumor action, particularly the derivative indirubin-3'-monoxime (I3M). Additionally, synthetic indirubin N-glycosides have demonstrated antiproliferative and cytotoxic properties in various cancer cells. A sensitive LC-MS/MS method was developed and validated for quantification of indirubin N-glycosides KD87, KD85, and I3M in plasma and cell culture medium. The applicability of the method was demonstrated in a mouse plasma study.

Summary: This study investigated the valorization of asparagus leafy by-products as a potential source of rutin through selected extraction and purification protocols. The protocol resulting in the highest extraction yield was chosen, followed by purification via multiple liquid-liquid back extraction using ethanol, methanol, or water. The proposed purification protocol yielded yellow-color crystals, confirmed as rutin through characterization techniques. The purity of the rutin was determined to be 97.6% by ultra-performance liquid chromatography, with a yield of 78.2%. The remaining rutin (21.8%) was found in the collected liquids, which can be recycled using the same purification process. These protocols offer a simple and effective method for rutin extraction and purification from asparagus leafy by-products.

Recent investigations have focused on studying the metabolism of AP-238 to identify useful markers for drug consumption. Through in vitro and in vivo assays, a total of 32 metabolites of AP-238 have been identified, with hydroxylation being the main metabolic pathway. The detection of these metabolites has proven to be effective in confirming drug intake, especially in cases where small traces of the parent drug are present.

Summary: Although the use of synthetic cannabinoids (SCs) is increasing, they are rarely analyzed in abstinence control programs for driver's license regranting. This study analyzed hair samples from various regions in Germany to determine the prevalence of SCs. The analysis detected 181 SCs, with the most prevalent compounds being 5F-MDMB-PICA and MDMB-4en-PINACA. Hair sample analysis proved to be a valuable tool for monitoring SC use, and an up-to-date analytical method is essential to keep pace with newly emerging SC analogs.
