Summary: A sensitive and specific heterogeneous assay for quantitation of miRNA-39 was developed using ICSDPR, polyperoxidase conjugated with streptavidin, and enhanced chemiluminescence. The assay demonstrated a low detection limit, low coefficient of variation, high specificity, and negligible matrix effect, making it useful for estimating miRNA purification yield from cells.

Summary: miRNA is a promising biomarker for cancer diagnosis, and the development of new detection methods for it has been a hot topic recently. In this study, we identified miR-625-5p as a potential biomarker for colorectal cancer through expression analysis and phenotypic experiments. We also developed a novel detection method based on strand displacement amplification (SDA) to monitor miR-625-5p levels, which showed good response and high specificity.

Summary: A novel simple multiple strand displacement reaction (MSDR) coupled hybridization strand reaction (HCR) was proposed for label-free and ultrasensitive Type b3a2 assay. The method achieves target recycling and amplification by releasing multiple messenger DNAs (mDNA) and forms a linear duplex-DNA structure for in-situ growth of silver nanoparticles (AgNPs) to generate electrochemical response. Under optimal conditions, the biosensor achieved a low detection limit of 0.56 fM and a wide linear range from 10 fM to 10 nM for sensitive detection of Type b3a2 with good selectivity and stability.

Summary: DNA methyltransferase (MTase) is considered a promising biomarker and therapeutic target, playing a crucial regulatory role in various biological processes. A cascade strand displacement amplification strategy was developed for highly sensitive and label-free fluorescent detection of DNA MTase activity, showing improved detection sensitivity. The strategy also effectively evaluated the inhibitory effects of 5-fluorouracil, benzylpenicillin, and gentamycin on DNA MTase activity, suggesting great potential for new method exploration and early disease diagnosis.
