Summary: Ti3C2-Fe3O4 composites with exposed non-modified Ti3C2 MXene nanosheets were designed and prepared for the enrichment of phosphopeptides. The composites interacted with the phosphopeptides through metal oxide affinity chromatography and hydrophilic interaction chromatography mechanisms. The magnetic nanomaterial showed high sensitivity and selectivity, and satisfactory results were obtained in the enrichment of phosphopeptides from real samples and traditional Chinese medicine injection.

Summary: Phosphorylation is a key component of cellular signaling cascades, controlling important cellular functions. However, phosphoproteomic analysis is typically limited by the amount of sample required for phosphopeptide enrichment. In this study, a new strategy was developed to increase phosphoproteomic signals from a small amount of sample without phosphoenrichment.

Summary: Posttraumatic infections in orthopedic trauma patients, especially in open fractures, require rapid and accurate identification of pathogens for clinical diagnosis and antimicrobial treatment. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) is effective for identifying pathogens, but pre-purification of liquid clinical specimens is necessary for direct analysis. A study showed success in coupling Fc-MBL@Fe3O4 enrichment with MALDI-TOF MS profiling for direct identification of pathogens in liquid-cultured samples, which has great potential for clinical application.

Summary: Researchers developed a green strategy to prepare carbonaceous spheres rich in phosphoric acid groups with D-Glucose 6-phosphate sodium salt as a sole carbon source. The carbonaceous spheres were further functionalized with Ti4+ to obtain excellent performance in selective capture of phosphopeptides. The CS-Ti4+ showed great potential in mass spectrometry-based phosphoproteome studies by capturing low-abundance phosphopeptides from human saliva.

Summary: We developed a bioinertized nanoflow LC/MS/MS system for sensitive analysis of phosphopeptides. Metal ion depletion from the mobile phase and prevention of indirect contact with metal components significantly improved the recovery of phosphopeptides. By inserting an online metal ion removal device, the identified phosphopeptides showed an average increase in peak areas of 9.9-fold overall and 77-fold for multiply phosphorylated peptides.

Summary: This study demonstrates that using chemically functionalized nanoporous silicon surfaces can address the limitations of MALDI-ToF MS analysis. The physicochemical properties of metabolites and surfaces play a significant role in MS analysis. Porous substrates are more suitable for hydrophobic metabolites, while plane substrates are better for hydrophilic metabolites.
