Summary: Insecticide resistance is a severe global problem for vector control. A recent study analysed the molecular metabolite changes in deltamethrin-resistant An. sinensis mosquitoes using liquid chromatography tandem mass spectrometry (LC-MS/MS) and found that the differential metabolites were mainly associated with glycerophospholipid metabolism. These findings provide further mechanistic understanding of insecticide resistance in An. sinensis.

Summary: Female adult anautogenous mosquitoes rely on diverse chemical stimuli to locate and discriminate hosts for feeding. Odorant binding protein AsOBP1 plays a crucial role in host-seeking behavior of Anopheles sinensis mosquitoes. The study identifies human odor compounds, such as 1-tetradecanol and heptanal, that enhance the attraction of mosquitoes, and provides a potential target for malaria control.

Summary: Recent discoveries have revealed the potential regulatory functions of reversible N6-methyladenosine (m6A) methylation on messenger RNA (mRNA). This study profiles the transcriptome-wide m6A in Anopheles sinensis, a mosquito species, and identifies its conserved nature. The distribution pattern of m6A in this species is associated with sex-specific pathways and positively correlated with mRNA abundance, indicating a regulatory role in gene expression. The study also highlights the involvement of m6A methylation in spermatogenesis-related genes, providing insights for controlling disease transmission by this mosquito vector.

Summary: ISVs are considered ancestors to arboviruses. This study characterized the RNA viromes in different tissues of two mosquito species using a metatranscriptomics approach. Summary: Miliusanes, belonging to meroterpenoids, are anticancer lead molecules isolated from Miliusa plants. In this study, 20 compounds, including 12 new ones, were isolated and identified from M. sinensis. These compounds showed potent inhibition of cell growth in various human derived cancer cell lines. Compound 11 demonstrated stronger cytotoxic activity and contained a new unconjugated 1,4-diketone system.

Summary: This study successfully applied CRISPR/Cas9 mediated knock-in technology to achieve highly efficient, site-specific mutagenesis in Anopheles sinensis. The integration of the EGFP marker gene into the odorant receptor co-receptor (Orco) confirmed Orco as a key mediator of multiple olfactory-driven behaviors in the An. sinensis life cycle, emphasizing its importance as a key molecular target for malaria control. The results also demonstrated that CRISPR/Cas9 is a simple and highly efficient genome editing technique for An. sinensis and could be utilized in developing genetic control tools for this vector.

Summary: The study identified 32 putative cytosolic and three putative microsomal GST genes in An. sinensis. Transcriptome analysis showed high expression of GSTs in larvae and in specific tissues of adults. GSTd2 and GSTe2 were significantly upregulated in pyrethroid-resistant populations, and silencing these genes increased susceptibility to deltamethrin. Recombinant GSTd2 and GSTe2 showed high enzymatic activity in metabolizing certain chemicals.

Summary: Haldane's rule of speciation states that hybrid male sterility or inviability is more likely to affect the heterogametic sex of inter-species hybrids. Darwin's corollary suggests that there are asymmetric phenotypes in inter-species hybrids from reciprocal crosses. By studying the phenotypes of F1 hybrids among closely related species of malaria mosquitoes, researchers have identified different genetic factors and molecular mechanisms responsible for reproductive isolation in reciprocal crosses among species of the An. gambiae complex.

Summary: We present a case of a patient with common variable immunodeficiency (CVID) who exhibited short stature and was treated with recombinant human growth hormone (rhGH). Whole exome sequencing identified a novel pathogenic single-nucleotide duplication in the NFKB1 gene, leading to NFKB1 haploinsufficiency. This is the first reported case of CVID caused by an NFKB1 mutation presenting with short stature. The study also suggests a potential shared mechanism involving NFKB1 and GH-IGF-1 pathways for both CVID and short stature in this patient.
