Alterations of vasoactive intestinal polypeptide receptors in allergic rhinitis

Background: Vasoactive intestinal peptide (VIP) is one of the parasympathetic neurotransmitters involved in the regulation of airway mucus secretion. The biological functions of VIP are mediated through two receptors (vasoactive intestinal peptide receptor type 1 [VPAC1R] and type 2 [VPAC2R]). The purpose of this study was to determine the distribution of VIP receptors and to compare the level of VIP receptor expression in the nasal mucosa of patients with allergic rhinitis and normal controls. Methods: Inferior turbinate mucosal samples were obtained from 20 normal subjects and 20 patients with allergic rhinitis. VPAC1R and VPAC2R mRNA was extracted from the nasal mucosa, and then a reverse-transcription-polymerase chain reaction was performed. Sections were immunostained using specific antibodies for VIP receptors. Western blot analysis was used to analyze differences in the level of expression of VPAC1R and VPAC2R protein between patients with allergic rhinitis and normal controls. Results: The level of expression of VIP receptor mRNA and protein in patients with allergic rhinitis was significantly increased compared with normal nasal mucosa. VIP receptor immunoreactivity was detected on the nasal epithelium and submucosal glands in nasal specimens from both normal controls and patients with allergic rhinitis. In the epithelium from patients with allergic rhinitis, VIP receptor immunoreactivity was strong, whereas in the nasal epithelium from normal subjects it was faint. Conclusion: These results suggest that an increased expression level of VIP receptors is one possible explanation for nasal hyperresponsiveness in patients with allergic rhinitis. DNMTs Are Involved in TGF-β1-Induced Epithelial-Mesenchymal Transitions in Airway Epithelial Cells

Summary: The pathogenesis of chronic rhinosinusitis is closely related to tissue remodeling, particularly epithelial-mesenchymal transition. Epigenetic mechanisms, such as DNA methylation, mediated by DNA methyltransferases, play key roles in this process. This study aimed to investigate the involvement of DNA methyltransferases in TGF-beta 1-induced epithelial-mesenchymal transition and explore the underlying mechanisms. The results showed that DNA methyltransferases were upregulated in chronic rhinosinusitis tissues and their expression was positively correlated with disease severity. Further experiments revealed that TGF-beta 1 induced the expression of DNA methyltransferases, which led to DNA methylation and epithelial-mesenchymal transition via the p38, JNK, Snail, and Slug signaling pathways. Inhibition of DNA methyltransferases suppressed the epithelial-mesenchymal transition process, suggesting its potential as a therapeutic strategy for chronic rhinosinusitis.

Recently, small extracellular vesicles derived from human umbilical cord mesenchymal stem cells (U-exo) have gained attention as a cell-free therapeutic strategy due to their safety and efficacy potential. This study found that U-exo exhibits antiviral activities against influenza A/B virus (IFV) and human seasonal coronavirus (HCoV) infections, potentially through the suppression of viral replication and the enhancement of IFN signaling and ISGs production.

Glycolytic reprogramming is involved in tissue remodeling on chronic rhinosinusitis

Summary: This study aimed to determine whether glycolytic reprogramming contributes to myofibroblast differentiation and ECM production in nasal fibroblasts. Hypoxia-inducing factor (HIF)-1 alpha was a high-level regulator of glycolysis, and increased HIF-1 alpha expression promoted glycolysis of nasal fibroblasts, and inhibition of HIF-1 alpha down-regulated myofibroblasts differentiation and ECM production. This study suggests that inhibition of the glycolytic enzyme and HIF-1 alpha in nasal fibroblasts regulates myofibroblast differentiation and ECM generation associated with nasal mucosa remodeling.
