ICAM-1 Deficiency Exacerbates Sarcoid-Like Granulomatosis Induced by Propionibacterium acnes through Impaired IL-10 Production by Regulatory T Cells

Propionibacterium acnes has been implicated as one of the suggested causative antigens for sarcoidosis, a systemic granulomatous disease. By injecting heat-killed P. acnes into the dorsal skin of C57BL/6J mice on days 1, 3, 5, and 14, sarcoid-like granulomatosis was induced in skin and lungs of these mice on day 28. To clarify the role of cell adhesion molecules in cutaneous sarcoidosis, we induced sarcoid-like granulomatosis in mice deficient of intercellular adhesion molecule (ICAM)-1, L-selectin, P-selectin, or E-selectin via repeated P. acnes injection. Histopathologic analysis revealed that granuloma formation was aggravated in the skin and lungs of ICAM-1-deficient mice compared with wild-type mice. Within skin granulomas of ICAM-1-deficient mice, P. acnes immunization up-regulated mRNA expression of tumor necrosis factor-a, although it failed to induce IL-10 mRNA expression in contrast to wild-type mice. Infiltration of regulatory T cells into skin granuloma was similar between wild-type mice and ICAM-1-deficient mice. P. acnes immunization induced IL-10 production by CD4(+)CD25(+)Foxp3(+) regulatory T cells in Lymph nodes of wild-type mice in vivo, which was absent in regulatory T cells of ICAM-1-deficient mice. Our results indicate that ICAM-1 is imperative for inducing regulatory T cells to produce IL-10 in vivo, which would prevent granuloma formation.

Local chromatin dynamics, supported by an interplay between epigenetic regulators and transcription factors, underlie transcriptional responses upon stress exposure. Epigenetic regulators play a leading role in chromatin remodeling during stress adaptation, and the timely release and restoration of these factors are indispensable for appropriate skin repair.

Serum cell-free DNA as a new biomarker in cutaneous T-cell lymphoma

Summary: In recent years, circulating cell-free DNA (cfDNA) has gained attention as a biomarker for cancer. This study found that serum cfDNA levels in patients with cutaneous T-cell lymphoma (CTCL) were significantly higher than healthy controls and increased with disease progression. There were positive correlations between cfDNA levels and certain blood markers and clinical findings, and higher cfDNA levels were associated with a worse prognosis in CTCL patients.

Summary: Bullous pemphigoid (BP) is an autoimmune disease characterized by the activation of immune cells and blister formation through excess extracellular DNA production. This study demonstrates that interleukin (IL)-26 binds to extracellular DNA, contributing to DNA sensing in BP. Patients with BP showed elevated levels of circulating IL-26, forming IL-26-DNA complexes that play a dual role in regulating local immunity and blister formation.

Impact of Pretreatment Systemic Inflammatory Markers on Treatment Persistence with Biologics and Conventional Systemic Therapy: A Retrospective Study of Patients with Psoriasis Vulgaris and Psoriatic Arthritis

Summary: This study examined accessible systemic inflammatory markers in patients with psoriasis vulgaris and psoriatic arthritis, and evaluated their association with psoriasis severity, the presence of arthritis, and treatment continuation rates. The findings showed positive correlations between various inflammatory markers and Psoriasis Area and Severity Index scores. Higher platelet/lymphocyte ratio (PLR) or CRP values were associated with psoriatic arthritis diagnosis. Higher pretreatment neutrophil or platelet count, PLR, and SII were associated with lower treatment continuation rates of conventional systemic agents. These findings suggest that accessible inflammatory markers can effectively assess systemic inflammation and guide therapeutic approaches for psoriasis vulgaris and psoriatic arthritis.
