Summary: NR supplementation reduced adipocyte cell size in BAT of HFD-fed mice, and enhanced in vitro norepinephrine-stimulated mitochondrial uncoupling in adipocytes derived from human BAT. However, NR supplementation in human volunteers did not alter BAT activity or cold-induced thermogenesis. Further research needs to be done to understand the differences in NAD+ metabolism between mice and humans.

Summary: GDF15 plays a role in preventing glucotoxicity-mediated altered glucose-stimulated insulin secretion and connexin-36 downregulation. Inhibition of endogenous GDF15 reduces glucose-stimulated insulin secretion in normal conditions while exacerbating glucolipotoxicity-reduced cell survival. This indicates that endogenous GDF15 is necessary for cell survival but not glucose-stimulated insulin secretion in the context of glucolipotoxicity.

New Insight in Hyperinsulinism/Hyperammonemia Syndrome by Magnetic Resonance Imaging and Spectroscopy

Summary: Hyperinsulinism/hyperammonemia syndrome (HI/HA) is caused by activating mutations in the GLUD1 gene, leading to insulin overproduction and chronic asymptomatic hyperammonemia. Brain magnetic resonance imaging in HI/HA patients shows differences in brain biochemistry compared to other hyperammonemia disorders, with normal levels of combined glutamate and glutamine.

NADPH oxidase enzymes (NOX-es) play a crucial role in regulating cellular events related to metabolism, receptor signal transmission, proliferation, and apoptosis. This review evaluates the potential relevance of NOX enzymes as pharmacological targets to alleviate fatty liver disease.

Summary: This systematic review and meta-analysis found an association between epicardial fat thickness (EFT) and nonalcoholic fatty liver disease (NAFLD). EFT was significantly higher in patients with NAFLD compared to controls, and further increased in patients with severe liver steatosis. Higher blood levels of aspartate aminotransferase were associated with a deeper association between EFT and NAFLD.
