Summary: This study developed a miniaturized library preparation protocol called Mini-XT, which is more cost-effective for sequencing SARS-CoV-2 genomes. The protocol reduces library preparation reagent volumes and overall tip usage while maintaining sequencing quality. Validation through sequencing a large number of samples demonstrated that Mini-XT has comparable sequencing quality and lineage calling results to standard protocols.

Genome-wide transcriptome profiling of human trabecular meshwork cells treated with TGF-β2

Summary: This study used RNA-Seq technology to explore the transcriptomic changes of TGF-beta 2 in normal human trabecular meshwork cells, identifying differentially expressed genes and associated pathways that contribute to the pathogenesis of primary open angle glaucoma (POAG). The study revealed the involvement of ECM regulation, TGF-beta signaling, proliferation/apoptosis, inflammation/wound healing, MAPK signaling, oxidative stress, and RHO signaling pathways in POAG. Additionally, the study identified novel genes and pathways affected by TGF-beta 2 treatment, suggesting potential new therapeutic targets.

In vivo single-cell transcriptomics reveal Klebsiella pneumoniae skews lung macrophages to promote infection

Summary: This study investigates a novel mechanism by which the human pathogen Klebsiella pneumoniae counteracts host defense by skewing lung macrophage polarization. The authors found that Klebsiella hijacks a TLR-type I IFN-IL10-STAT6 axis to rewire macrophages and promote its intracellular survival. Additionally, glycolysis and capsule polysaccharide were identified as key factors governing the macrophage polarization induced by Klebsiella. This research provides important insights into the understanding of bacterial infections and the development of therapeutic strategies.

Summary: Pseudoexfoliation glaucoma is an aggressive disease that affects about 30 million people worldwide. TGF-beta 1 plays a role in its pathogenesis. Through RNA sequencing, we identified several differentially expressed genes and determined important pathways and molecular functions. This cellular model provides insights into the disease mechanism and supports the development of novel therapies.

Summary: This study used single-cell RNA sequencing to investigate the transcriptomic changes induced by zinc supplementation in human primary retinal pigment epithelial (RPE) cells. The results showed that zinc supplementation had a multitude of effects on the RPE transcriptome, including modulation of genes involved in pigmentation, complement regulation, mineralization, and cholesterol metabolism processes associated with age-related macular degeneration (AMD).
