Multicentric Castleman's disease (MCD), a relatively rare lymphoproliferative disorder that presents with heterogenous symptoms including fevers, anemia, and multifocal lymphadenopathy, is today most commonly observed in individuals infected with human immunodeficiency virus type-1 (HIV). In such individuals, a lymph node biopsy typically identifies cells that stain for Kaposi's sarcoma-associated herpesvirus proteins, and most HIV-associated MCD features can be attributed to the presence of this gamma-herpesvirus. Surgery and antiviral therapies including highly active antiretroviral therapy, interferon-alpha, foscarnet, ganciclovir, and antibodies to interleukin-6 have proved largely ineffective, and chemotherapy in HIV positive individuals is complicated by limited efficacy and pronounced toxicity. While no randomized trials have been performed, more recently the use of the anti-CD20 monoclonal antibody rituximab in large single center cohorts has been associated with prolonged remissions, radiologic responses, as well as hematologic and serum chemistry normalization of the inflammatory picture observed, at the expense of B cell depletion and flare of Kaposi's sarcoma. MCD represents a model of disease at the interplay between tumor biology, infection, and immunology.

Analysis of delay in adjuvant chemotherapy in locally advanced rectal cancer

Summary: The timing of adjuvant chemotherapy (AC) for rectal cancer after neoadjuvant chemoradiation and surgical resection has a significant impact on patients' overall survival. Delayed AC initiation is associated with decreased 5-year survival, and patients in the delayed AC group have an increased risk of death compared to those receiving early or intermediate AC, despite having lower proportions of pathological lymph-node metastasis. Additionally, patients with higher socioeconomic and education status are more likely to receive early chemotherapy.

The Inhibitory Properties of a Novel, Selective LMTK3 Kinase Inhibitor

Summary: Recently, the oncogenic role of LMTK3 has been established in different tumors, making it a potential target for cancer therapy. In this study, a highly specific small molecule inhibitor for LMTK3, called C36, was identified and shown to have significant therapeutic effects in cancer cell lines. The binding affinity between LMTK3 and C36 was demonstrated through microscale thermophoresis, and C36 was found to inhibit the activity of LMTK3 by binding to both the ATP- and substrate-binding sites. Treatment with C36 led to decreased proliferation and increased apoptosis in breast cancer cells, highlighting the potential of LMTK3 inhibitors for cancer therapy.

Summary: This study aimed to investigate the long-term effects of previous immunization on evolving variants of SARS-CoV-2. The study found that patients who received the vaccine had fewer sequelae after COVID-19, as well as a lower incidence of respiratory complications and prolonged fatigue. This finding is important for the treatment and survival of cancer patients.

Exosomes as a Source of Biomarkers for Gastrointestinal Cancers

Summary: Exosomes are small extracellular vesicles that carry important information for intercellular communication. They are produced abundantly by tumor cells and carry tumor-specific content, such as DNA, RNA, and proteins, which play significant roles in cancer development and progression. Due to their presence in various biofluids, exosomes can be developed as liquid biopsy tools for early diagnosis, molecular profiling, disease surveillance, and treatment response monitoring. This review focuses on the diagnostic, prognostic, and predictive potential of exosomal biomarkers in colorectal and gastric cancers, and discusses the challenges and limitations of their clinical application.

Summary: The aim of this study was to evaluate the impact of HIV infection on the functional characteristics of T cell infiltrate in hepatocellular carcinoma (HCC). Results showed that HIV infection was associated with increased immune exhaustion and higher expression of PD-L1 in tumor tissue. Therefore, prospective testing of immunotherapy in HIV-associated HCC is warranted.

Summary: Mucinous adenocarcinoma of the appendix is a rare and highly aggressive form of lower gastrointestinal cancer. Cytoreductive surgery (CRS) and hyperthermic intraperitoneal chemotherapy (HIPEC) have emerged as effective treatment options, particularly for patients with extensive peritoneal carcinomatosis. Repeat CRS can achieve durable response and prolonged survival, even in cases with initial incomplete cytoreduction and high-volume disease.
