A high-fat meal promotes lipid-load and apolipoprotein B-48 receptor transcriptional activity in circulating monocytes

Background: The postprandial metabolism of dietary fats results in the production of apolipoprotein B-48 (apoB48)-containing triglyceride-rich lipoproteins (TRLs), which cause rapid receptor-mediated macrophage lipid engorgement via the apoB48 cell surface receptor (apoB48R). Monocytes circulate together with apoB48-containing TRLs in the postprandial bloodstream and may start accumulating lipids even before their migration to tissues and differentiation to macrophages. Objective: We sought to determine whether circulating monocytes are equipped with apoB48R and whether, in the postprandial state, circulating monocytes accumulate lipids and modulate apoB48R transcriptional activity after intake of a high-fat meal. Design: In a crossover design, we studied the effect of a high-fat meal on fasting and postprandial concentrations of triglycerides, free fatty acids, cholesterol, and insulin in 12 healthy men. TRLs and monocytes were freshly isolated at fasting, hourly until the postprandial peak, and at the late postprandial phase. TRLs were subjected to triglycerides, apoB48, and apolipoprotein B-100 analyses; and lipid accumulation and apoB48R mRNA expression levels were measured in monocytes. Results: Monocytes showed a time-dependent lipid accumulation in response to the high-fat meal, which was paralleled by an increase in apoB48R mRNA expression levels. These effects were coincident only with an increase in apoB48-containing TRLs in the postprandial phase and were also observed ex vivo in freshly isolated monocytes incubated with apoB48-containing TRLs. Conclusion: In a setting of abundant plasma apoB48-containing TRLs, these findings highlight the role of dietary fat in inducing lipid accumulation and apoB48R gene transcription in circulating monocytes. Summary: This study investigates the role of galectin-3 (Gal3) in the development of endotoxemia and sepsis. The absence of Gal3 reduces mortality and tissue damage in a murine model of endotoxemia. Gal3 levels decrease in immune cells, particularly monocytes, after LPS injection, and Gal3 is up-regulated in peripheral organs. These findings suggest that Gal3 plays a significant role in systemic inflammation and may provide potential therapeutic options for sepsis.

Summary: This study analyzed the impact of sex, obesity, and their interactions on the gene expression of two lipid flippases in visceral adipose tissue (VAT). The results showed that obesity had a significant influence on the gene expression of these flippases, with a stronger effect in men for one of the flippases. The study also found associations between these flippases and microRNAs, which were also affected by sex.

Summary: Aneurysmal subarachnoid hemorrhage (aSAH) is a neurovascular disease caused by blood leakage into the subarachnoid space after rupture of cerebral vessels. This study analyzed the changes in peripheral blood mononuclear cells (PBMCs) and their interaction with endothelium in aSAH patients, with a focus on adhesion and expression of adhesion molecules. The study found increased adhesion of PBMCs, particularly monocytes, in patients with aSAH, especially those who developed vasospasm. The expression of several adhesion molecules in T lymphocytes and monocytes was altered, providing insights for predicting vasospasm and improving treatment.

Summary: Older people have lower antibody titers and declining humoral immunity over time after Covid-19 vaccination due to immune system aging. However, the factors that predict the waning immune response have been poorly explored. This study measured antibody levels and various markers in a cohort of nursing home residents and healthcare workers who received the BNT162b2 vaccine. Thymosin-alpha 1 levels were found to be associated with the durability of the vaccine response, suggesting its potential as a biomarker for predicting vaccine response in older individuals.

Summary: Despite receiving three doses of the mRNA COVID-19 vaccine, kidney transplant recipients have a weak humoral response, indicating the need for new approaches to enhance their protective immunity. A longitudinal study found that immunosuppressive therapy, worse kidney function, higher inflammatory status, and older age are associated with a reduced response, while immune cell counts, thymosin-a1 plasma concentration, and thymic output are associated with a stronger humoral response. Further research on thymosin-a1 as a potential adjuvant for vaccine boosters is warranted.

Summary: Aneurysmal subarachnoid hemorrhage (aSAH) is associated with high morbidity and mortality rates. Patients with aSAH exhibit structural alterations in their high-density lipoproteins (HDL), resulting in loss of anti-inflammatory function. This study highlights the importance of investigating HDL in the pathophysiology of aSAH and suggests that functional HDL supplementation could be a novel therapeutic approach for the inflammatory response following aSAH.

Effects of exercise timing on metabolic health

This article reviews the role of exercise timing in metabolic health and discusses the potential mechanisms behind its benefits.

New evidence for dietary fatty acids in the neutrophil traffic between the bone marrow and the peripheral blood

Summary: This study investigated the effects of different fatty acids on the migration of neutrophils from bone marrow to blood in mice. The results showed that a high-fat diet enriched in saturated fatty acids promoted neutrophil migration and was associated with neutrophil apoptosis and bone marrow inflammation. In contrast, monounsaturated fatty acids were found to preserve bone marrow health and ensure proper neutrophil migration.
