The influence of TNF alpha and IL-8 on secretory action of neutrophils isolated from heifers in the course of bovine respiratory disease

The goal of this study was to give insight into the lesser known action of neutrophils in the course of bovine respiratory disease (BRD). Neutrophils are principally involved in resistance to infection but under certain conditions these cells participate in lung injury. TNF alpha and IL-8, primary cytokines which play an essential role in neutrophil activation during the inflammatory process, may lead not only to host defence reaction but also to destructive responses. Neutrophil constituents, such as elastase, myeloperoxidase (MPO), alkaline phosphatase (ALKP) and superoxide among others, contribute to tissue destruction. Also, 5-oxo-eicosatetraenoic acid (5-oxo-ETE), the potent activator of neutrophil chemotaxis and degranulation, can cause lung injury. This study revealed that neutrophils under the influence of TNF alpha exacerbated the release of elastase, MPO, ALKP and superoxide. IL-8, in turn, triggered those neutrophils which were isolated from heifers suffering from BRD, and released elastase, ALKP and 5-oxo-ETE, but not MPO or superoxide (O-2(-)). The most pronounced degranulative response was observed in the acute form of BRD and a lesser response was seen in the chronic form. The secretory action of neutrophils varied not only depending on the form and phase of the disease but it also diminished successively during recovery.

The Effect of Neutrophil-Derived Products on the Function of Leukocytes Obtained after Titanium Implantation in the Ovine Model

Our study on sheep models found that insertion of Ti implants did not cause adverse effects and had no negative impact on neutrophils and MDM. The immune system plays a pivotal role in the pathogenesis of OA, with both humoral and cellular mediators contributing to cartilage destruction and joint inflammation. Animal models are widely used to study the disease and potential therapies. This review provides an overview of OA signs, pathogenesis, and classification, as well as discussing the advantages and disadvantages of different animal models. Treatment options include mesenchymal stem cells, gene therapies, and blood-derived products, along with tissue engineering techniques and biomaterials.

Native Pig Neutrophil Products: Insights into Their Antimicrobial Activity

Summary: This study aimed to explore the antimicrobial activity and mechanisms of action of a porcine neutrophil cathelicidin mixture (MPPN). It was found that MPPN exhibited bactericidal activity against E. coli at high concentrations and bacteriostatic activity at lower concentrations. The interaction of MPPN with lipid bilayers caused alterations in the structure.

Analysis of In Vitro Leukocyte Responses to Biomaterials in the Presence of Antimicrobial Porcine Neutrophil Extract (AMPNE)

Summary: Implant insertion may cause excessive inflammation and lead to complications. Neutrophils and macrophages play important roles in tissue repair and their responses to biomaterials were examined. Porcine neutrophils exposed to hydroxyapatite (HA) showed increased enzyme release and ROS/RNS generation. Human neutrophils produced more NO after contact with HA, while HAP or FAP did not elicit a significant response. Human macrophages produced more superoxide and NO when cultured with HA, and curdlan reduced ROS/RNS generation. AMPNE reduced neutrophil activity with all biomaterials except curdlan.
