Effect of culture conditions, cytokinins, methyl jasmonate and salicylic acid on the biomass accumulation and production of withanolides in multiple shoot culture of Withania somnifera (L.) Dunal using liquid culture

The influence of cytokinins and culture conditions including medium volume, harvest time and elicitation with abiotic elicitors (SA/MeJ) have been studied for the optimal production of biomass and withanolides in the multiple shoot culture of Withania somnifera. Elicitation of shoot inoculum mass (2 g l(-l) FW) with SA at 100 mu M in the presence of 0.6 mg l(-l) BA and 20 mg l(-l) spermidine for 4 h exposure time at the 4th week in 20 ml liquid medium recorded higher withanolides production (withanolides A [8.48 mg g(-l) DW], withanolides B [15.47 mg g(-l) DW], withaferin A [29.55 mg g(-l) DW] and withanone [23.44 mg g(-l) DW]), which were 1.14 to 1.18-fold higher than elicitation with MeJ at 100 mu M after 5 weeks of culture. SA-elicited cultures did not exhibit much variation in biomass accumulation when compared to control. This cytokinin induces and SA-elicited multiple shoot culture protocol provides a potential alternative for the optimal production of biomass and withanolides utilizing liquid culture.

An insight into in vitro strategies for bioproduction of isoflavones

Summary: Isoflavones are potent secondary metabolites widely distributed among Fabaceae family, with beneficial functions such as lowering cardiovascular diseases and reducing menopausal symptoms. In vitro studies have successfully synthesized value-added metabolites with industrial applications, while plant organ cultures are recommended for synthesizing plant-based pharmaceutically active isoflavones. Biotechnological approaches can promote the yield and production of these metabolites.

Influence of Endophytic Bacterium, Cellulosimicrobium sp. FRR2 on Plant Growth of Amaranthus campestris L. and Bacterial Survival at Adverse Environmental Conditions

Summary: This study found that the endophytic bacterium FRR2 isolated from the roots of Ficus religiosa has beneficial effects on the growth of Amaranthus campestris, and has tolerance to salinity and heavy metals, possibly through nitrogen fixation, indole acetic acid production, amylase and protease activities. The results suggest that Cellulosimicrobium sp. strain FRR2 could be an effective endophyte to promote the growth of green leafy vegetables.

Summary: Green leafy vegetables are rich in antioxidants and minerals that prevent food-borne pathogen infections. The endophytic bacterium APL3 was found to significantly enhance the growth of Amaranthus polygonoides L. and increase its antimicrobial potential. Utilization of P. dendritiformis strain APL3 can improve the plants' antimicrobial properties against food-borne pathogens.

Summary: An effective protocol for callus-dependent indirect organogenesis in Indian pea cv. Ageta 6 was standardized using cotyledonary node explant. The study highlighted the positive effects of polyamines and plant growth regulators on pea regeneration, and demonstrated that polyamine-assisted regenerated pea plants exhibited improved physiological and antioxidant profiles.

Biotechnology Advances in Bioremediation of Arsenic: A Review

Summary: Arsenic is a highly toxic metalloid that is widely distributed in the Earth's crust, and its contamination caused by human activities is becoming an emerging environmental issue. Sustainable and effective remediation methods, such as microbial remediation and phytoremediation, are needed to prevent and protect against arsenic exposure. Recent advances in genetic modifications and biotechnology have allowed for the development of more efficient transgenic microbes and plants, as well as novel organo-mineral materials for arsenic remediation. This review discusses the latest insights and relevant physiological and molecular mechanisms in arsenic bio-/phytoremediation, providing valuable information for the development of arsenic-tolerant microbes and plants.

Endophytes: A vital source of medicine-A review

Summary: Soil and endosymbiotic microorganisms have symbiotic and non-symbiotic relationships with plants, and certain bacteria and fungi have been identified as bio-fertilizers to improve crop productivity. However, there have been limited studies on the beneficial effects of endophytic microbes on drug production in plants. This review focuses on medicinal plants and their associated endophytic bacteria and fungi, exploring their potential in developing drugs against human pathogens and illnesses.

Exogenous application of stevioside enhances root growth promotion in soybean (Glycine max (L.) Merrill)

Summary: This study investigated the effects of externally applied stevioside on soybean root growth. Treatment with 24.5μM stevioside significantly increased root length, root numbers, root biomass, shoot length, and shoot biomass. It also enhanced photosynthetic pigments, leaf relative water content, and antioxidant enzymes. However, a higher concentration of stevioside (40.5μM) increased various biochemical parameters such as total polyphenolic content and proline content. Gene expression analysis revealed that stevioside treatment influenced the expression of root growth development-related genes in soybean. Overall, stevioside has the potential to improve morpho-physiological traits, biochemical status, and root development gene expression in soybean.

Mitigation of Salt Stress in Soybean (Glycine max (L.) Merrill) Using Exogenous Application of Onion Extract

Summary: Plant growth and development under salt stress can be improved by exogenous application of onion extract. Onion extract enhances morpho-physiological traits, enzymatic antioxidant activities, and salt tolerance in soybean. It also shows potential as an eco-friendly and cost-effective solution to mitigate abiotic stress in crops.

A simple and efficient genetic transformation system for soybean (Glycine max (L.) Merrill) targeting apical meristem of modified half-seed explant

Summary: The study aimed to establish a fast and highly reproducible transformation method for soybean targeting the apical meristem. Modified half-seed explants were treated with different durations of sonication and vacuum infiltration in the presence of Agrobacterium tumefaciens. The optimized protocol achieved a high transformation efficiency of up to 38.0% and could be applied to other Indian soybean cultivars.

Isolation and Characterization of Erianthus arundinaceus Phosphate Transporter 1 (PHT1) Gene Promoter and 5′ Deletion Analysis of Transcriptional Regulation Regions under Phosphate Stress in Transgenic Tobacco

Summary: Phosphorus deficiency has a significant impact on plant growth and development. The use of promoters from phosphate transporter genes can enhance phosphate uptake abilities in genetically modified plants, leading to improved crop yields in phosphate-deficient soils. In this study, the EaPHT1;2 gene from Erianthus arundinaceus was isolated and characterized. The promoter region of EaPHT1;2 showed higher expression than the commonly used CaMV35S promoter. The short promoter sequence of EaPHT1;2 showed significant expression under low-Pi-stress conditions and holds promise for developing stress-tolerant transgenic crops.

Meta-topolin enhances regeneration and Agrobacterium-mediated genetic transformation in radish (Raphanus sativus L.)

Summary: In this study, a novel plant hormone called Meta-topolin (mT) was found to enhance regeneration and genetic transformation efficiency in radish. By adding mT in optimal concentrations to the culture medium, shoot induction, elongation, and rooting abilities of radish were improved. Using Agrobacterium-mediated transformation, mT also increased the efficiency of genetic transformation in radish.
