Nodal segments obtained from in vitro proliferated shoots of Eclipta alba (L.) Hassk, were encapsulated in calcium alginate beads for large-scale clonal propagation, short-term conservation and germplasm exchange and distribution. The best gel complexation was achieved using 3% sodium alginate and 100 mM CaCl(2)center dot 2H(2)O. Maximum percent response (100%) for conversion of encapsulated nodal segments into plantlets was obtained on 0.7% agar-solidified full-strength MS medium containing 0.88 mu M BAP. Encapsulated nodal segments could be stored at low temperature (4A degrees C) up to 60 days with a survival frequency of 51.2%. The well-developed plantlets regenerated from encapsulated nodal segments were hardened-off successfully with 90% survival frequency.

Base editing in rice: current progress, advances, limitations, and future perspectives

Rajesh Yarra, Lingaraj Sahoo

Summary: Base editing is a promising genome editing tool for generating single-nucleotide changes in the rice genome, which is essential for developing new rice varieties with desirable agronomic traits to sustain global food security. Base editing technology utilizes adenosine or cytidine base editors for precise editing at the target region, making it an efficient and reliable tool for rice crop improvement. This review discusses different adenine and cytosine base editors developed for precise genome editing of rice, addressing the current progress, advances, limitations, and future perspectives of base editing technology for rice crop improvement.

Occurrence of Cotton leaf curl Multan virus and associated betasatellites with leaf curl disease of Bhut-Jolokia chillies (Capsicum chinense Jacq.) in India

Sneha Yogindran, Manish Kumar, Lingaraj Sahoo, Keithellakpam Sanatombi, Supriya Chakraborty

Summary: This study identified begomoviruses in infected Bhut-Jolokia chili leaf samples from the North-East region of India. The presence of recombination sites on both viral genome and betsatellite may have aided the virus in successfully establishing infection in chili plants. The findings suggest a potential spread of CLCuMuV to non-host crops in the North-East region of India.

Efficient Agrobacterium tumefaciens-mediated stable genetic transformation of green microalgae, Chlorella sorokiniana

Prabin Kumar Sharma, Vaibhab V. Goud, Y. Yamamoto, Lingaraj Sahoo

Summary: A stable and efficient Agrobacterium tumefaciens-mediated transformation system was established for the first time in Chlorella sorokiniana, resulting in significantly higher transformation efficiency. The integration of hptII gene into the genome of C. sorokiniana was confirmed, and the expression of hptII and GUS genes in the transgenic cell lines was detected. The growth and metabolic processes of transformed cells were not compromised by transgene expression, indicating the potential for future production of biofuels, carotenoids, nutraceuticals, and therapeutic proteins.

Balancing yield trade-off in legumes during multiple stress tolerance via strategic crosstalk by native NAC transcription factors

Richa Srivastava, Lingaraj Sahoo

Summary: NAC transcription factors play a crucial role in plant growth and stress adaptation, but their overexpression can lead to growth inhibition and the functional unpredictability of non-native members limits their application in legume crops. It is essential to understand the crosstalk between stress-responsive genes and growth to overcome the trade-off between growth and stress adaptation.

Deep Recurrent Neural Network Architecture of High Order Indirect Integration Method

Zhengrong Xie, Yibo Ai, Jia Chen, Fan Wang, Weidong Zhang

Summary: The indirect integration method (IIM) is a numerical method used to solve initial value problems of ordinary differential equations. It optimizes the parameters of the highest order derivative function layer through residual iteration and then integrates step by step to the solution function layer using a complex trapezoid quadrature formula. The author designs a single-layer recurrent neural network to simulate the first-order IIM and constructs a deep recurrent neural network by stacking single-layer recurrent networks to implement any order IIM algorithm. The recursive relationship between the gradients of different order IIM is obtained using the backpropagation through time (BPTT) algorithm.

Start codon targeted (SCoT) polymorphism marker in plant genome analysis: current status and prospects

Manoj K. K. Rai

Summary: This review provides a comprehensive overview of the start codon targeted (SCoT) polymorphism marker and its applications in various genetic and genomic studies. The SCoT marker is a targeted fingerprinting technique that has gained importance in plant genetics, genomics, and molecular breeding. It targets the highly conserved region flanking the start codon and can distinguish genetic variations in specific genes related to traits. The marker is simple, cost-effective, highly polymorphic, and reproducible, making it valuable for many applications in plant species.

Pooja Asthana, Manoj K. K. Rai, Uma Jaiswal

Summary: The study demonstrates the selection and regeneration of NaCl tolerant plants of Sapindus trifoliatus via somatic embryogenesis. NaCl tolerant callus lines were selected from callus induced from leaf disc explants and were further characterized for their growth, tolerance index, and ion and organic content. Somatic embryos developed from NaCl tolerant callus showed germination on medium with and without NaCl, and the tolerant plants were able to survive in a potting mixture containing NaCl. The physiological and biochemical parameters of the tolerant plants were analyzed, indicating the effectiveness of in vitro selection in developing NaCl tolerant plants of S. trifoliatus.

Jatan K. Shekhawat, Manoj K. Rai, N. S. Shekhawat, Vinod Kataria

Summary: The synergistic effect of meta-topolin (mT) with auxin and cytokinin effectively stimulates large-scale shoot regeneration in Maytenus emarginata. A combination of 0.5 mg L(-1)mT, 0.25 mg L-1 Kin, and 0.1 mg L-1 IAA was found to be the most effective for shoot proliferation. This study highlights the importance of the interaction between mT, Kin, and IAA in promoting shoot proliferation in medicinal tree species.
