α-Mangostin suppresses human gastric adenocarcinoma cells in vitro via blockade of Stat3 signaling pathway

Aim: To investigate the anti-tumor effects of a-mangostin, a major xanthone identified in the pericarp of mangosteen (Garcinia mangostana Linn), against human gastric adenocarcinoma cells in vitro, and the mechanisms of the effects. Methods: Human gastric adenocarcinoma cell lines BGC-823 and SGC-7901 were treated with alpha-mangostin. The cell viability was measured with MU assay, and cell apoptosis was examined using flow cytometry and TUNEL assay. The expression of the relevant proteins was detected using Western blot. Results: Treatment with a-mangostin (3-10 mu g/mL) inhibited the viability of both BGC-823 and SGC-7901 cells in dose- and time-manners. Furthermore, a-mangostin (7 mu g/mL) time-dependently increased the apoptosis index of the cancer cells, reduced the mitochondrial membrane potential of the cancer cells, and significantly increased the release of cytochrome c and AIF into cytoplasm. Moreover, the a-mangostin treatment markedly suppressed the constitutive Stat3 protein activation, and Stat3-regulated Bcl-xL and Mcl-1 protein levels in the cancer cells. Conclusion: The anti-tumor effects of alpha-mangostin against human gastric adenocarcinoma cells in vitro can be partly attributed to blockade of Stat3 signaling pathway.

p53 upregulated by HIF-1 α promotes gastric mucosal epithelial cells apoptosis in portal hypertensive gastropathy

Summary: This study investigated the role of hypoxia-inducible factor-1 alpha (HIF-1 alpha) and p53 in mucosal apoptosis in portal hypertensive gastropathy (PHG). The results showed that HIF-1 alpha and p53 were significantly upregulated in the gastric mucosa of PHG patients and animal models, and inhibition of these molecules could attenuate apoptosis. It was also found that HIF-1 alpha regulated the p53-induced mucosal epithelial apoptotic signaling pathway. Therefore, HIF-1 alpha and p53 may be potential therapeutic targets for PHG.

Summary: This study investigated the impact of sex on outcomes following transcatheter aortic valve replacement (TAVR) in patients with bicuspid aortic valves (BAVs). The results showed that women with BAVs had fewer comorbidities, while men had a higher proportion of type 1 BAV, more calcification, and larger aortic roots. Men had better in-hospital outcomes, except for the need for second valve implantation, but 1-year survival was comparable between sexes.

Effect of endotracheal tube size on airway resistance and dynamic lung compliance

Summary: This study investigated the effect of endotracheal tube (ETT) size on airway resistance (RAW) and dynamic lung compliance (COMPL) in patients undergoing laparoscopic colorectal surgery. The results showed that in the non-pneumoperitoneal state, a smaller ETT internal diameter resulted in higher RAW and lower COMPL. In the pneumoperitoneal state, the ETT size had no effect on COMPL, but the RAW was higher with an ID7.0 ETT compared to an ID8.0 ETT within a certain range.

Semi-synthesis of biotin-bearing activity-based ubiquitin probes through sequential enzymatic ligation, N-S acyl transfer and aminolysis reaction

Summary: Activity-based ubiquitin probes (Ub-ABPs) are effective tools for studying the molecular mechanism and profiling of deubiquitinating enzymes (DUBs). However, the synthesis of commonly used biotin-bearing Ub-ABPs is technically challenging. In this study, a one-pot semi-synthetic strategy was developed to obtain Ub-ABPs carrying a biotin tag through enzymatic ligation, N-S acyl transfer, and aminolysis reaction without purification steps. These probes enable the capture of different DUBs families for enrichment and immunoblotting using the attached biotin tag.

Liver injury in COVID-19: Clinical features, potential mechanisms, risk factors and clinical treatments

Summary: The COVID-19 pandemic has been a global health threat for nearly 3 years and it often causes liver injury in addition to pulmonary complications. The incidence of liver injury is significantly increased in severe cases and understanding its causes is urgently needed. Clinical features of liver injury include abnormal liver function and imaging changes, which can be evaluated through tests and scans. Risk factors for liver injury in COVID-19 patients include male sex, preexisting liver disease, diabetes, obesity, and hypertension. The mechanism of liver injury caused by COVID-19 is not fully understood, but it is believed to be related to inflammatory response, hypoxic damage, ischemia-reperfusion injury, and drug side effects. This review summarizesthe existing literature on liver injury caused by COVID-19, including clinical features, underlying mechanisms, and potential risk factors. Clinical management and recommendations for patients with liver injury are also discussed.

Summary: In this study, a deep learning algorithm was used to achieve rapid classification and diagnosis of coronary artery disease from CCTA images. The deep learning model can automatically segment the coronary artery quickly and accurately, delivering a diagnosis of >= 50% coronary artery stenosis. Artificial intelligence methods such as deep learning have the potential to significantly improve efficiency in CCTA image analysis.
