Do CAR-T and Allogeneic Stem Cell Transplant Both Have a Place in Lymphoid Neoplasms?

Summary: Allogeneic stem cell transplantation (allo-SCT) is the first immunotherapy for hematologic malignancies, being considered as a cure rather than a life-extending approach. The success of allo-SCT is attributed to intensive chemoradiotherapy and graft-versus-leukemia effects. However, challenges such as disease relapse, graft-versus-host disease, infections, and toxicities remain. Furthermore, the introduction of CAR-T cell therapy has revolutionized the treatment of lymphoid neoplasms. This review discusses the current standards, shifting paradigms, and the role of CAR-T in lymphoid neoplasms.

Machine Learning Approaches in Diagnosis, Prognosis and Treatment Selection of Cardiac Amyloidosis

Summary: Cardiac amyloidosis is a rare restrictive cardiomyopathy caused by abnormal deposition of amyloid protein, leading to impaired organ function. Due to similar clinical symptoms with more common hypertrophic diseases, early diagnosis of cardiac amyloidosis is often delayed. Accurate differentiation of different types of amyloidosis is necessary for appropriate treatment. The underdiagnosis of cardiac amyloidosis delays necessary therapeutic procedures, hinders quality of life, and affects clinical prognosis. Diagnostic work-up includes identification of clinical features, electrocardiographic and imaging findings, and histological demonstration of amyloid deposition. Automated diagnostic algorithms using machine learning techniques can help overcome the difficulty of early diagnosis. This review aims to evaluate various diagnostic approaches and artificial intelligence computational techniques in detecting cardiac amyloidosis.

Dipeptide Nitrile CD34 with Curcumin: A New Improved Combination Strategy to Synergistically Inhibit Rhodesain of Trypanosoma brucei rhodesiense

Summary: In this study, a synthetic inhibitor CD34 was found to have nanomolar binding affinity towards Rhodesain and showed synergistic action when used in combination with Curcumin, effectively inhibiting the activity of Trypanosoma brucei rhodesiense. Therefore, the combination of CD34 and Curcumin holds promise as a potential therapeutic strategy.

This review aims to compile information on novel biomarkers for immune thrombocytopenia to enhance patient management.

The role of High-mobility group box-1 and Psoriasin in multiple myeloma: Analysis of a population affected by monoclonal gammopathies and review of the literature

Summary: This study compared the plasma levels of HMGB-1 and psoriasin in patients with MM and MGUS, as well as in healthy controls. The results showed that MGUS patients had higher levels of HMGB-1 compared to the control group (p < 0.001), and MM patients had a significant difference in HMGB-1 levels compared to controls (p = 0.001). There was no difference in psoriasin levels among the three groups. Additionally, the study evaluated the existing knowledge in the literature about the potential mechanisms of action for these molecules in the onset and development of these disorders.

Summary: MicroRNAs are small molecules that play important roles in both healthy and pathological cells. They are influenced by genetic factors and epigenetic mechanisms, such as genomic imprinting and X chromosome inactivation in females. MiRNAs have been shown to be correlated with sex and cancer in both solid tumors and hematological malignancies. Understanding the role of miRNAs in cancerogenesis, autophagy, and apoptosis of different types of tumors can have implications for cancer treatment and prognosis.

Identification and Targeting of Mutant Neoantigens in Multiple Myeloma Treatment

Summary: Multiple myeloma (MM) is a difficult-to-treat malignant disease characterized by the proliferation of plasma cells in the bone marrow. This review analyzes the presence of neoantigens in MM patients and evaluates their potential as prognostic factors or therapeutic targets. The use of neoantigen treatment strategies and bispecific, trispecific, and conjugated antibodies for MM treatment are discussed. The utilization of CAR-T in relapsed and refractory patients is also examined.

Monoclonal Antibodies: The Greatest Resource to Treat Multiple Myeloma

Summary: Multiple myeloma is an incurable hematologic cancer characterized by immunological alterations in myeloid cells and lymphocytes. The current first-line therapy involves chemotherapy, but there is a high relapse rate and refractory MM cases. New monoclonal antibodies (Mab) including daratumumab, isatuximab, and elotuzumab, along with bispecific antibodies and CAR T cell therapy, have shown promise in immunotherapy for MM. CD38 (daratumumab and isatuximab), SLAM7 (elotuzumab) and BCMA (belantamab mafodotin) are the main antibody targets for MM treatment. Although MM is still incurable, combining the available drugs offers hope for improving outcomes.
