Summary: The renin-angiotensin system (RAS) is crucial for regulating arterial pressure and fluid balance, and the angiotensin type 2 receptor (AT2R) plays a protective role in pathophysiological conditions. This study investigated the impact of single nucleotide polymorphisms (SNPs) on AT2R and its interactions with agonist and antagonist molecules. Computational analysis and molecular dynamics simulations revealed that certain mutations affect the protein conformation and ligand binding. These findings have implications for the development of better medications and future in vitro and in vivo studies.

Summary: This study demonstrates that the combination therapy of neprilysin inhibitor sacubitril and angiotensin II type 2 receptor agonist C21 provides superior renal protection compared to the combination therapy of sacubitril and valsartan in obese rats fed a high salt diet. The sacubitril/C21 treatment significantly decreases proteinuria, albuminuria, the expression of nephrin, and kidney weight, independent of hyperfiltration, and completely prevents HSD-induced increases in renal angiotensin II.

The renin-angiotensin-aldosterone system plays a critical role in maternal-fetal health and the development of pregnancy complications, and understanding its programming during pregnancy is crucial for identifying at-risk individuals and developing effective therapies. Preclinical and translational research should adhere to best practices in accurately measuring components of this system to ensure reliable results.

The renin-angiotensin system (RAS) plays a role in hypertension and emotional disorders, and elevated RAS activity is associated with depression and anxiety through neuroinflammation, stress, and oxidative stress. Blocking the RAS could potentially be a future treatment approach for anxiety and depression. The positive effects of RAS blockers on these disorders are reviewed, aiming to provide a promising target for novel medications and to improve the efficacy of current treatments independent of blood pressure management.

Summary: This study evaluated the role of serum progesterone sulfates in the etiology of gestational diabetes mellitus (GDM). The results showed that concentrations of epiallopregnanolone sulfate (PM5S) were reduced in GDM and in women with higher fasting plasma glucose. PM5S increased glucose-stimulated insulin secretion (GSIS) and this effect was mediated by TRPM3. These findings highlight the importance of progesterone sulfates in the pathogenesis of GDM.

Summary: This study examines risk factors for anxiety and depression among pregnant women during the COVID-19 pandemic. The results indicate that pregnant women in high-income economies experience higher levels of anxiety and depression. Factors such as the need for psychiatric treatment before pregnancy, restrictions in social distancing and access to leisure activities, pregnancy-related complications, fear of adverse outcomes in children related to COVID-19, and financial burden are associated with increased risk of anxiety and depression during pregnancy.

Summary: This sequential, prospective meta-analysis aimed to identify risk factors among pregnant and postpartum women with COVID-19 for adverse outcomes related to disease severity, maternal morbidities, neonatal mortality and morbidity, and adverse birth outcomes. Data from 21 participating studies were included, and risk factors and outcomes were analyzed. The study found that women with comorbidities, HIV infection, prepregnancy underweight, and anemia were at increased risk for severe COVID-19-related outcomes and adverse birth outcomes.

Summary: This study aimed to examine the relationship between the English index of multiple deprivation (IMD) and the incidence of stillbirth and evaluate whether IMD contributes to the prediction of stillbirth provided by maternal demographic characteristics and elements of medical history. The study found that women living in the most deprived areas in South East England had higher rates of stillbirth, particularly placental dysfunction-related stillbirth. However, including IMD in stillbirth screening did not improve the prediction provided by race, other maternal characteristics, and elements of medical history.

Summary: Despite heterogeneity in study design and quality, a sequential, prospective meta-analysis found that SARS-CoV-2 infection during pregnancy increases the risk of maternal mortality, severe maternal morbidity, and neonatal morbidity. However, there is no association with stillbirth or intrauterine growth restriction. The findings highlight the importance of preventing and managing SARS-CoV-2 infection in pregnant women. Overall, the study is rated 8 out of 10 in terms of importance.

Summary: The aim of this study was to assess whether intrapartum ultrasound (ITU) measurements in maternal lateral posture are comparable to ITU measurements in semi-recumbent position. The results showed that the angle of progression (AOP) and head-perineum distance (HPD) were lower in the lateral posture compared to semi-recumbent position, while they were higher with contractions. Therefore, monitoring the second stage of labor with ITU in lateral maternal posture is feasible.

Summary: This retrospective study analyzed 683 patients with uterine sarcoma to identify and evaluate the impact of different prognostic factors on overall survival and disease-free survival. The results showed variations in 5-year overall survival and disease-free survival rates among different subtypes of uterine sarcoma. The presence of residual disease after surgery was identified as the most significant factor affecting overall survival.

Summary: This study aimed to assess the diagnostic accuracy of placental growth factor and placental growth factor-based models in predicting the development of preeclampsia in asymptomatic women. The findings suggest that placental growth factor combined with maternal factors has the highest predictive performance for early preeclampsia, while placental growth factor-based models perform similarly to soluble fms-like tyrosine kinase-1-placental growth factor ratio for any-onset preeclampsia in the third trimester. Standardized research using consistent models is needed to accurately predict preeclampsia.
