In vitro studies of deferasirox derivatives as potential organelle-targeting traceable anti-cancer therapeutics

Summary: In this study, strategic functionalization of the FDA approved chelator deferasirox was conducted to produce organelle-targeting iron chelators with enhanced activity against A549 lung cancer cells. Derivative 8 exhibited improved antiproliferative activity compared to the original compound 1, and was shown to preferentially localize within the lysosome through fluorescent cell imaging.

The new mitochondrial uncoupler BAM15 induces ROS production for treatment of acute myeloid leukemia

Summary: This study found that the expression of FABP5 promotes cell viability, inhibits apoptosis, and maintains triglyceride production in acute myeloid leukemia (AML) cells. FABP5 expression is upregulated in AML patients and is associated with poor overall survival. FABP5 can be used as a biomarker to distinguish between normal and high-risk AML patients. It also predicts drug resistance in AML patients.

8-Hydroxydaidzein Induces Apoptosis and Inhibits AML-Associated Gene Expression in U-937 Cells: Potential Phytochemical for AML Treatment

Summary: The compound 8-hydroxydaidzein (8-OHD) has been found to inhibit cell proliferation and induce apoptosis in acute myeloid leukemia (AML) cells. It downregulates the expression of AML-associated genes and interacts with key proteins involved in cell proliferation. Furthermore, a synergistic effect is observed when 8-OHD is used in combination with cytarabine (Ara-C), a standard chemotherapy drug for AML.

Targeting Iron Metabolism and Ferroptosis as Novel Therapeutic Approaches in Cardiovascular Diseases

Summary: Iron plays a crucial role in the cardiovascular system, and dysregulation of iron metabolism and ferroptosis are involved in the pathogenesis of various cardiovascular diseases. Iron chelators and lipophilic antioxidants have been shown to inhibit ferroptosis and lipid peroxidation in atherosclerosis, cardiomyopathy, and hypertension. Inhibition of ferroptosis can alleviate cardiac impairments and fibrosis in hypertension by enhancing GPX4 signaling. Novel small molecules targeting ferroptosis may be effective in the treatment of cardiovascular diseases. This article summarizes the regulatory roles and underlying mechanisms of iron metabolism dysregulation and ferroptosis in cardiovascular diseases and suggests targeting iron metabolism and ferroptosis as potential therapeutic strategies.

Identification of highly efficacious PROTACs targeting BRD4 against acute myeloid leukemia: Design, synthesis, and biological evaluations

Summary: The abnormal activation of BRD4 accelerates the progression of acute myeloid leukemia (AML). A range of small-molecule PROTACs were rationally designed to degrade BRD4 mediated by the E3 ubiquitin ligase CRBN. Among them, B24 exhibited remarkable BRD4 degradation and excellent anti-proliferative activities in MV4-11 cells, surpassing the effect of the BRD4 inhibitor ( +)-JQ-1. B24 also induced cell apoptosis and inhibited cell proliferation by down-regulating c-Myc and up-regulating Bax.

Downregulation of SET8 is one of the mechanisms by which LukS-PV induces apoptosis in AML cells, suggesting SET8 as a potential therapeutic target for AML. LukS-PV may serve as a drug candidate for AML treatment targeting epigenetic modifications.

A bavachinin analog, D36, induces cell death by targeting both autophagy and apoptosis pathway in acute myeloid leukemia cells

Summary: This study suggests that D36, a compound isolated from Psoralea corylifolia L., exhibits potential in treating acute myeloid leukemia (AML). D36 can induce cell death in AML cells through the activation of apoptosis and autophagy, as well as by blocking the cell cycle progression. Furthermore, D36 shows promising anti-tumor activity in a mouse model of AML without significant side effects.

MiR-376a-3p increases cell apoptosis in acute myeloid leukemia by targeting MT1X

Summary: Metallothionein1X (MT1X) is highly expressed in acute myeloid leukemia (AML) and acts as a candidate prognostic indicator. Inhibiting MT1X can suppress AML cell proliferation, induce apoptosis, and inhibit nuclear factor-kappa B (NF-kappa B) signaling. MiR-376a-3p negatively regulates MT1X, suppressing AML cell proliferation and inducing apoptosis. These findings suggest that elevated MT1X expression may contribute to AML progression and indicate that the miR-376a/MT1X axis could be a potential therapeutic target for AML patients.

Prognostic outcomes of cytomegalovirus reactivation after autologous stem cell transplantation

Summary: This study analyzed the association between CMV reactivation and survival outcomes in patients undergoing auto-SCT and developed a predictive model for late CMV reactivation. The study found that early CMV reactivation was significantly associated with better overall survival in multiple myeloma patients, while late CMV reactivation, high lactate dehydrogenase level, and lymphoma diagnosis were independent risk factors for poor overall survival. The predictive risk model showed good discrimination in identifying high-risk patients for late CMV reactivation.

Implications of the 5th Edition of the World Health Organization Classification and International Consensus Classification of Myeloid Neoplasm in Myelodysplastic Syndrome With Excess Blasts and Acute Myeloid Leukemia

Summary: The WHO and ICC have recently published updated classifications of myeloid neoplasms. A study was conducted to examine the changes in diagnosis distribution among MDS-EB and AML patients using these new classifications. The results showed that approximately 31.9% of the patients were reclassified based on the 2022 classifications, primarily due to changes in mutation-based diagnostic criteria. The detection of multi-hit TP53 alterations at diagnosis was found to be important for prognosis in MDS-EB patients. Implementation of these classifications may lead to the identification of different subtypes based on genetic characteristics.

Summary: This study evaluated the prophylactic effectiveness of pegfilgrastim in reducing neutropenia and febrile neutropenia in CLL patients undergoing R-FC chemotherapy. The results showed that prophylactic pegfilgrastim significantly reduced the incidence of neutropenia and febrile neutropenia, as well as the risk of treatment interruption and treatment-related mortality.

Identification and overcoming rituximab resistance in diffuse large B-cell lymphoma using next-generation sequencing

Summary: A study compared the response of sensitive and resistant cell lines to the chemotherapy drug rituximab and found that the resistant cells had increased activation of the p38 MAPK pathway.
