Summary: This article explores the requirements for an optimal grading system for non-muscle-invasive bladder cancer through expert opinion. It suggests splitting the WHO 2004 high-grade category into WHO 1973 grade 2 and 3 subsets, providing more detailed histological criteria. These changes may lead to better treatment decisions for patients with bladder cancer.

Summary: There is evidence that cribriform morphology is associated with a worse prognosis for prostatic adenocarcinoma. This study aimed to investigate the reproducibility of cribriform cancer diagnosis in prostate needle biopsies. A panel of nine prostate pathology experts independently reviewed 304 digitised biopsies and achieved a 2/3 consensus diagnosis of cribriform and noncribriform cancer in 90% of the cases. Strict diagnostic criteria were used to identify cribriform patterns and generate a set of consensus cases for standardization.

Summary: Aberrant activation of the Ras/Raf/ERK-MAPK pathway is involved in cancer progression, but the negative regulation by Spred2 may counter this progression in non-invasive bladder carcinoma. The loss of Spred2 regulation is associated with increased ERK activation and Ki67 index in more malignant cancers. Spred2 appears to be a key regulator in the progression of non-invasive bladder carcinoma.

Summary: This study aimed to develop a novel histologic grading system for gastrointestinal graft-versus-host disease (GVHD) by incorporating independent evaluation of apoptotic counts and crypt destruction. However, due to lack of reproducibility, widespread adoption of this system is not recommended.

Summary: The study evaluated interobserver variability among a multi-institutional group of breast pathologists using digital whole slide imaging for breast cancer grading, showing moderate concordance overall, with variations in observer agreement for different grades and components. Discrepancies in grading infrequently led to clinically meaningful changes in prognostic stage, with tubule formation showing the best observer agreement.

Summary: The study surveyed the preferences and practices of members of the European Association of Urology and International Society of Urological Pathology regarding the grading of non-muscle-invasive bladder cancer (NMIBC). Both the WHO1973 and WHO2004 classification systems are still widely used, but there is limited support for continuing to use them in their current formats. A hybrid grading system composed of both WHO1973 and WHO2004 may be a promising alternative.

Summary: This review critically analyzes the application of 3D bioprinting-based tissue engineering approaches for urinary bladder and urethra reconstruction, discussing the materials and biomechanical characteristics of scaffolds, emphasizing the need to mimic the biomechanical properties of native urological tissues, and suggesting that bioprinting with biomolecular hydrogel bioink may provide a solution for creating patient-specific grafts.

Summary: Bladder cancer can be classified into basal and luminal molecular subtypes, with basal tumors responding better to immune checkpoint inhibitors but having worse prognosis, while luminal tumors respond better to FGFR3 and HER2. Tumors with squamous differentiation show better response to EGFR-targeting agents.

Summary: Enormous progress in diagnostic testing allows for more accurate diagnosis and improved clinical outcomes. However, the increasing volume and diversity of test results can overwhelm clinicians, leading to incorrect or delayed diagnoses. Integrative diagnostics, combining diagnostic and clinical data through informatics tools, has the potential to enhance treatment selection, modification, and termination, ultimately improving patient outcomes and reducing costs.

Summary: This study validates how psychological stress affects the paracellular permeability of bladder mucosa, with its non-invasive assay using MRI. Under water avoidance stress conditions, urothelial denudation, edema, and ZO-1 downregulation were observed.

Summary: Acute abdomen caused by spontaneous rupture of the inflammatory myofibroblastic tumor of the urinary bladder (IMTUB) is a rare clinical emergency. This article reports a case of emergency laparotomy and partial cystectomy for the spontaneous rupture of IMTUB, highlighting the importance of preoperative examination and postoperative pathology in determining the nature of the tumor and implementing personalized treatment.

Summary: The research assessed the concordance between biopsy and radical prostatectomy specimens using different grading systems, showing that the 2014 ISUP modified system improves overall concordance between the two types of specimens. This improvement allows for more accurate prognostication and management in high-grade disease, independent of other factors.

Summary: This study evaluated the prognostic value of the IASLC histologic grading system in a large Japanese cohort and found that it has similar predictive ability for recurrence and death compared to the Arch system and the Sica's system. The IASLC grading system is considered to provide physicians with better information for postsurgery treatment.

Summary: A man in his 70s, previously diagnosed with prostate adenocarcinoma, received radiotherapy 11 years ago. He developed urinary symptoms 10 years later, and a bladder neck tumor was found. Pathology revealed colorectal adenocarcinoma. Despite unremarkable colonoscopy, imaging showed bladder and prostate involvement. The patient refused surgery and underwent chemoradiation. Reirradiation was performed on a recurrent lymph node, with a positive response.
