Nicotine Fast Dissolving Films Made of Maltodextrins: A Feasibility Study

This work aimed to develop a fast-dissolving film made of low dextrose equivalent maltodextrins (MDX) containing nicotine hydrogen tartrate salt (NHT). Particular attention was given to the selection of the suitable taste-masking agent (TMA) and the characterisation of the ductility and flexibility under different mechanical stresses. MDX with two different dextrose equivalents (DEs), namely DE 6 and DE 12, were selected in order to evaluate the effect of polymer molecular weight on film tensile properties. The bitterness and astringency intensity of NHT and the suppression effect of several TMA were evaluated by a Taste-Sensing System. The films were characterised in term of NHT content, tensile properties, disintegration time and drug dissolution test. As expected, placebo films made of MDX DE 6 appeared stiffer and less ductile than film prepared using MDX DE 12. The films disintegrated within 10 s. Among the tested TMA, the milk and mint flavours resulted particularly suitable to mask the taste of NHT. The addition of NHT and taste-masking agents affected film tensile properties; however, the effect of the addition of these components can be counterweighted by modulating the glycerine content and/or the MDX molecular weight. The feasibility of NHT loaded fast-dissolving films was demonstrated.

Printing of cutaneous patches loaded with propranolol for the treatment of infantile haemangiomas

Summary: Topical propranolol has been used for treating cutaneous infantile haemangiomas, but frequent applications are needed. This study developed propranolol patches using hot-melt ram extrusion printing, with formulations releasing the drug in a reasonable time and exhibiting suitable adhesive properties. The patch made of Eudragit RL and TBC with 1% propranolol hydrochloride is the most promising for superficial haemangiomas, while the patch made of Eudragit RL and ATEC with 1% propranolol base can be used for deep haemangiomas.

Design of Liposomal Lidocaine/Cannabidiol Fixed Combinations for Local Neuropathic Pain Treatment

Summary: The administration of drug fixed combinations by nanocarriers is an attractive approach to enhance the skin penetration and synergistic effects of drugs. This study investigated the use of deformable liposomes to deliver the combination of lidocaine and cannabidiol in the deeper layers of the skin. Results showed that the DiMiL formulation had higher skin permeation and dermal retention compared to the G-liposomes formulation, and both were more effective than control formulations carrying free drugs.

Ultrasound-Assisted Extraction of Cannabinoids from Cannabis Sativa for Medicinal Purpose

Summary: In the past 20 years, the interest in medicinal cannabis oily extracts has grown, leading to the need for high quality preparations. This study systematically investigated the use of an ultrasonic homogenizer as an alternative extraction method and found that it can provide equivalent content in CBD and THC compared to the traditional hot maceration method, with the advantage of a faster process. The qualitative characteristics of the oil remained constant for both treatments.

Micelles-in-Liposome Systems Obtained by Proliposomal Approach for Cannabidiol Delivery: Structural Features and Skin Penetration

Summary: This study investigated the advantages of combining proliposomes and DiMiL as drug carriers to enhance the skin penetration of CBD. Sucrose and trehalose were found to be the best carriers for spray-dried and slurried proliposomes, respectively. Cryo-EM images showed the presence of micelles in the aqueous core of lipid vesicles in the DiMiL system, and the presence of sugars did not alter the structural organization. CBD permeation through human epidermis was significantly improved when carried by DiMiL systems, and trehalose further increased the flux.

3D printed mucoadhesive orodispersible films manufactured by direct powder extrusion for personalized clobetasol propionate based paediatric therapies

Summary: The aim of this study is to develop and produce novel oral mucoadhesive films delivering Clobetasol propionate (CBS) through Direct Powder Extrusion (DPE) 3D printing technique for paediatric treatment of Oral Lichen Planus (OLP). Different polymeric materials, such as hydroxypropylmethylcellulose or polyethylene oxide blended with chitosan (CS), were tested to obtain suitable mucoadhesive films. Hydroxypropyl-& beta;-cyclodextrin was added to enhance CBS solubility. The DPE-printed films showed strong adhesion, improved drug characteristics, and significant drug exposure time on the mucosa.

Feeding Next-Generation Nanomedicines to Europe: Regulatory and Quality Challenges

Summary: New and innovative nanomedicines have revolutionized the prognosis of many human diseases. However, their peculiar physicochemical properties pose new challenges in regulatory approval. This article critically discusses the current open issues regarding regulatory qualification and physicochemical characterization of nanosystems, considering the current European regulatory framework on nanomedicine products. Practicable paths for improving their quality assurance and predicting their fate in vivo are also proposed.

Comparative Evaluation of Metformin and Metronidazole Release from Oral Lyophilisates with Different Methods

Summary: The aim of this study was to compare three different dissolution methods for drug release assessment in oral lyophilisates. Interpolyelectrolyte complexes (IPECs) were prepared using two different polymers and two model drugs. The drug release kinetics were evaluated using artificial saliva and three different set-ups. The results showed that the choice of set-up significantly influenced the drug release profiles.
