Blood Serum From Head and Neck Squamous Cell Carcinoma Patients Induces Altered MicroRNA and Target Gene Expression Profile in Treated Cells

The head and neck squamous cell carcinoma (HNSCC) represents one of the most common cancers in humans. Close to 600,000 new diagnoses are made every year worldwide and over half of diagnosed patients will not survive. In view of this low survival rate, the development of novel cell-based assays for HNSCC will allow more mechanistic approaches for specific diagnostics for each individual patient. The cell-based assays will provide more informative data predicting cellular processes in treated patient, which in effect would improve patient follow up. More importantly, it will increase the specificity and effectiveness of therapeutic approaches. In this study, we investigated the role of serum from HNSCC patients on the regulation of microRNA (miRNA) expression in exposed cells in vitro. Next-generation sequencing of miRNA revealed that serum from HNSCC patients induced a different miRNA expression profile than the serum from healthy individuals. Out of 377 miRNA detected, we found that 16 miRNAs were differentially expressed when comparing cells exposed to serum from HNSCC or healthy individuals. The analysis of gene ontologies and pathway analysis revealed that these miRNA target genes were involved in biological cancer-related processes, including cell cycle and apoptosis. The real-time PCR analysis revealed that serum from HNSCC patients downregulate the expression level of five genes involved in carcinogenesis and two of these genes-P53 and SLC2A1-are direct targets of detected miRNAs. These novel findings provide new insight into how cancer-associated factors in circulation regulate the expression of genes and regulatory elements in distal cells in favor of tumorigenesis. This has the potential for new therapeutic approaches and more specific diagnostics with tumor-specific cell lines or single-cell in vitro assays for personalized treatment and early detection of primary tumors or metastasis.

Extracellular vesicle proteomics and phosphoproteomics identify pathways for increased risk in patients hospitalized with COVID-19 and type 2 diabetes mellitus

Summary: Recent studies indicate that extracellular vesicles (EVs) are involved in the development of SARS-CoV-2 infection and the severity of COVID-19. However, the connection between COVID-19 and type 2 diabetes (T2D) in relation to EVs has not been explored. This study examined the proteomic and phosphoproteomic profiles of circulating EVs in hospitalized patients with and without T2D, who had either COVID-19 or non-COVID-19 acute respiratory illness (RSP). Differential expression of certain proteins and phosphoproteins was observed, with a central cluster of immunomodulatory and coagulation proteins (C1QA, C1QB, and C1QC) identified in both COVID-19 and T2D networks. The study also revealed the involvement of cellular kinases and EV-shuttled kinases in severe COVID-19, particularly in patients with comorbid diabetes.

Detectable Lipidomes and Metabolomes by Different Plasma Exosome Isolation Methods in Healthy Controls and Patients with Advanced Prostate and Lung Cancer

Summary: Circulating exosomes in the blood can be used as biomarkers for cancer. This study evaluated the effects of different methods of isolating plasma exosomes on the detection of multi-omic profiles in patients with lung cancer, prostate cancer, and healthy controls. The results showed that different isolation methods enriched distinct exosome cargos, and the size exclusion method captured the most diverse exosome populations. These findings suggest that the choice of isolation method can significantly impact cancer biomarker discovery.

Lipopolysaccharide effects on activation and apoptosis of primordial ovarian follicles in heifers

Summary: The objective of this study was to evaluate the effect of lipopolysaccharide (LPS) administration on activation and apoptosis of primordial follicles. The results showed no difference in the total number of follicles and different types of follicles. Furthermore, LPS challenge did not modulate the expression of genes related to ovarian reserve, oocyte survival, activation rate, and follicular abnormalities.

microRNA-449a reduces growth hormone-stimulated senescent cell burden through PI3K-mTOR signaling

Summary: Cellular senescence, implicated in various age-related disorders, can be attenuated by investigating methods to reduce the accumulation of senescent cells. miR-449a is down-regulated with age in normal mice but maintained in long-living GH-deficient df/df mice, showing potential as a serotherapeutic. GH downregulates miR-449a expression and accelerates senescence, while miR-449a upregulation reduces senescence through targeted reduction of senescence-associated genes and signaling pathways.

Summary: The aim of this study was to assess the morphological characteristics of ovarian follicles and cortex, as well as senescence markers, in goats of different ages. The results showed that the percentage of morphologically normal follicles decreased with age, but was not related to cellular senescence. Young females had higher follicular density compared to adult females, and the presence of lipofuscin was higher in adults than in young goats. Beta-galactosidase activity and gamma H2AX immunolabeling did not differ between groups.

Chronological and reproductive aging-associated changes in resistance to oxidative stress in post-reproductive female mice

Summary: This study aimed to investigate the role of young and old ovarian tissues in the response to oxidative stress. The results showed that the greatest changes in oxidative stress response proteins occurred during the period of reproductive failure. Transplanted mice exhibited different protein changes compared to control mice, suggesting an ovarian influence on aging-associated changes in oxidative stress response that is independent of germ cells and estradiol levels.
