Summary: Triple-negative breast cancer (TNBC) has poor prognosis and limited treatment options. Abnormal overexpression of circular RNA KIF4A (circKIF4A) in TNBC is associated with poor survival rate. This study aims to examine the functional role and underlying mechanism of circKIF4A. The results showed that circKIF4A is significantly upregulated in TNBC and its knockdown suppresses TNBC cell proliferation, migration, and invasion. CircKIF4A directly interacts with EIF4A3, which in turn interacts with SDC1. Knockdown of circKIF4A reduces the interaction between EIF4A3 and SDC1 as well as the stability of SDC1 mRNA. SDC1 activates the c-src/FAK signaling pathways and promotes TNBC progression. In vivo mouse models confirmed that circKIF4A induces TNBC progression through SDC1. CircKIF4A interacts with EIF4A3 to stabilize SDC1 mRNA, activating the c-src/FAK signaling pathways and promoting TNBC progression, potentially providing a therapeutic target for TNBC treatment.

Summary: The projected increase in life expectancy is expected to lead to a rise in age-related diseases, including cancer. This report critically reviews the literature on managing older patients with head and neck cancer (HNC). Older adults, especially those who are frail or have comorbidities, are more prone to complications and toxicities from HNC treatment. Screening older patients for predispositions and managing comorbidities is essential. Age alone should not be a determining factor for less intensive treatment, and older patients should be treated according to the best standard of care to avoid increased treatment failure rates and mortality. A multidisciplinary tumor board should establish the treatment plan, involving shared decision-making with patients and family, and treatment modifications should be considered for patients with severe comorbidities, frailty, or who refuse recommendations.

Summary: Treatment of head and neck squamous cell carcinomas (HNSCC) remains challenging. Dysregulation of miRNA is closely linked to tumorigenesis and tumor progression, making it a suitable target for cancer treatment. In this study, miR-301a was found to be overexpressed in HNSCC tumors and was shown to play a role in promoting tumor growth and invasion. The PI3K/PTEN/AKT and MEK/ERK pathways were identified as central signaling nodes responsible for mediating the oncogenic activity of miR-301a.

Summary: Osteosarcoma remains a challenging malignancy with limited immunotherapy options. However, this study demonstrates that chimeric antigen receptor (CAR) T cells targeting the ALPL-1 isoform of alkaline phosphatase show high efficiency and specificity in preclinical models of primary and metastatic osteosarcoma. These CAR-T cells display potent cytotoxicity against ALPL-positive cells without unexpected toxicities, providing a promising path for clinical translation of CAR-T immunotherapy in osteosarcoma.

Summary: Osteosarcoma is a malignant bone cancer with poor prognosis, partly due to drug-resistant cancer stem cells. Cold atmospheric plasmas and plasma-treated liquids (PTL) have shown selectivity against cancer cells, but their effects on cancer stem cells and in vivo tumor growth are not well understood. This study demonstrates that low doses of PTL promote stemness factors and self-renewal ability of osteosarcoma cells, leading to enhanced tumor growth in vivo. However, combining PTL with a STAT3 inhibitor can suppress tumor growth effectively, suggesting a potential treatment for osteosarcoma.

Summary: In non-well differentiated thyroid cancer, the rate of nodal involvement varies depending on the histology of the tumor. Treatment for anaplastic carcinoma, medullary carcinoma, and poorly differentiated carcinoma of the thyroid is well-established. However, treatment for other rare tumors needs to be individualized based on histology and anatomical distribution. The rate of lymph node involvement in non-well differentiated thyroid cancer is variable. Different types of thyroid cancer require different approaches to neck management.

This review provides current knowledge on the incidence, management, and outcomes of recurrent head and neck squamous cell carcinoma after salvage surgery.
