Clonal/subclonal changes and accumulation of CTX-M-type β-lactamase genes in fluoroquinolone-resistant Escherichia coli ST131 and ST1193 strains isolated during the past 12 years, Japan

Yukari Fukushima, Toyotaka Sato, Naoyuki Tsukamoto, Chie Nakajima, Yasuhiko Suzuki, Satoshi Takahashi, Shin-ichi Yokota

Summary: The study found that the increased resistance of E. coli to fluoroquinolones and third-generation cephalosporins in Japan can be attributed to the accumulation of bla(CTX-M) in different subclades and ST1193.

Oligosaccharide Metabolism and Lipoteichoic Acid Production in Lactobacillus gasseri and Lactobacillus paragasseri

Tsukasa Shiraishi, Shintaro Maeno, Sayoko Kishi, Tadashi Fujii, Hiroki Tanno, Katsuaki Hirano, Takumi Tochio, Yasuhiro Tanizawa, Masanori Arita, Shin-ichi Yokota, Akihito Endo

Summary: The growth and lipoteichoic acid production of two lactobacilli strains varied significantly in different oligosaccharide culture media, indicating that oligosaccharide metabolism can influence their probiotic properties. Transcription analysis suggested that differences in LTA size among the carbohydrates tested were partly due to variations in alanylation levels of LTA. This study highlights the importance of available oligosaccharides on the LTA production of industrially important lactobacilli.

Design, synthesis and biological evaluation of simplified analogues of MraY inhibitory natural product with rigid scaffold

Kazuhiro Okamoto, Aoi Ishikawa, Ryotaro Okawa, Kazuki Yamamoto, Toyotaka Sato, Shin-ichi Yokota, Kazuhiro Chiba, Satoshi Ichikawa

Summary: This study successfully simplified the chemical structures of Muraymycins and caprazamycins through structure based drug design, leading to the discovery of a novel compound scaffold with sub-micromolar MraY inhibitory activity. This scaffold shows potential for further optimization of antibacterial activities in the future.

Reduction of susceptibility to azoles and 5-fluorocytosine and growth acceleration in Candida albicans in glucosuria

Yoshiki Hiyama, Toyotaka Sato, Satoshi Takahashi, Soh Yamamoto, Noriko Ogasawara, Naoya Masumori, Shin-ichi Yokota

Summary: Research shows that the growth and antifungal susceptibilities of Candida species are significantly enhanced in urine with added glucose. This provides valuable insights into the clinical treatment of Candida infections in patients with diabetes mellitus and hyperglucosuria.

Role of Lipoteichoic Acid from the Genus Apilactobacillus in Inducing a Strong IgA Response

Chiaki Matsuzaki, Tsukasa Shiraishi, Tai-Ying Chiou, Yukari Nakashima, Yasuki Higashimura, Shin-ichi Yokota, Kenji Yamamoto, Tomoya Takahashi

Summary: Components of lactic acid bacteria with immunostimulatory effects are being increasingly studied for their therapeutic and prophylactic potential as alternatives to antibiotics, cognitive enhancements, and vaccine adjuvants. This study found that lactic acid bacteria belonging to the genus Apilactobacillus possess significantly higher activity in inducing immunoglobulin A (IgA) compared to other species. The differences in the structure of lipoteichoic acids (LTAs) may be related to this distinction in IgA-inducing activity.

A hydroxypropyl methylcellulose plaque assay for human respiratory syncytial virus

Summary: Quantifying proliferative virus particles is crucial in virology. Newly developed cellulose derivatives, such as hydroxypropyl methylcellulose (HPMC), have been found to be the optimal overlay material for plaque-forming assays. HPMC maintains cell proliferation and virus infectivity for human respiratory syncytial virus (RSV), making it a suitable material for plaque assays.

Solid-Phase Total Synthesis of Plusbacin A3

Kazuki Takashina, Akira Katsuyama, Rintaro Kaguchi, Kazuki Yamamoto, Toyotaka Sato, Satoshi Takahashi, Motohiro Horiuchi, Shin-ichi Yokota, Satoshi Ichikawa

Summary: The total synthesis of the depsipeptide natural product plusbacin A3 (1) was achieved through solid-phase peptide synthesis (SPPS). A specific three-component reaction and hydrolysis were used to prepare a key intermediate compatible with Fmoc SPPS. The peptide was then elongated on a solid support, cleaved from the resin, and subjected to macrolactamization and global deprotection to obtain plusbacin A3 (1).

Investigating the role of heat shock protein 47 in fibrosis in Crohn's disease

Hiroki Kurumi, Tomoaki Takata, Tsutomu Kanda, Takaaki Sugihara, Tomoyuki Kakugawa, Shin-ichi Yokota, Tomohito Morisaki, Taro Akashi, Hajime Isomoto

Summary: This study found that heat shock protein 47 (HSP47) is highly expressed in patients with Crohn's disease (CD) and is associated with fibrosis. Serum HSP47 levels and anti-HSP47 antibody titers can serve as biomarkers to differentiate fibrotic from non-fibrotic forms of CD. In addition, HSP47 inhibition can suppress collagen production in fibroblasts.

Colistin Susceptibility in Companion Animal-Derived Escherichia coli, Klebsiella spp., and Enterobacter spp. in Japan: Frequent Isolation of Colistin-Resistant Enterobacter cloacae Complex

Toyotaka Sato, Kazuki Harada, Masaru Usui, Shin-ichi Yokota, Motohiro Horiuchi

Summary: This study investigated the colistin susceptibility of Enterobacterales isolated from companion animals in Japan and found that most E. coli were susceptible to colistin, while a small proportion of Klebsiella spp. and Enterobacter spp. were resistant. Moreover, some colistin-resistant Enterobacter spp. belonged to lineages associated with humans and exhibited resistance to multiple classes of antibiotics.

Wall teichoic acid-dependent phagocytosis of intact cell walls of Lactiplantibacillus plantarum elicits IL-12 secretion from macrophages

Naoya Kojima, Shohei Kojima, Shin Hosokawa, Yoshiki Oda, Daisuke Zenke, Yuta Toura, Emi Onohara, Shin-ichi Yokota, Masato Nagaoka, Yasuhiro Kuroda

Summary: Selected lactic acid bacteria can stimulate macrophages and dendritic cells to secrete IL-12, and this study found that the cell wall teichoic acids (WTAs) displayed on the intact cell walls (ICWs) of L. plantarum play a key role in activating mouse macrophages to secrete IL-12. The study also showed that the size and shape of the cell walls impact actin remodeling and subsequent IL-12 production.
