Summary: The study demonstrated that lotus honey has higher physicochemical properties and antioxidant activity, and the combination of honey and ciprofloxacin showed better antibacterial effects against an E. coli mutant. A synergistic interaction was observed when using a triplex complex of ciprofloxacin, honey, and plant extract. The triplex complex significantly reduced gene expression levels and decreased the mutant's oxidative stress response.

Summary: This study found that certain antidepressants can induce bacterial resistance to disinfectants and activate certain genes to increase bacterial efflux of disinfectants, thereby increasing resistance. This provides novel insights into the potential source of water-borne disinfectant-resistant pathogens.

Summary: Our research has demonstrated that the loss of AcrB function in E. coli and S. Typhimurium leads to different biological effects despite conferring drug resistance to the same antibiotics. Additionally, we identified potential natural substrates of AcrB and discovered metabolite classes implicated in the virulence of S. Typhimurium. This information can aid in the development of new efflux inhibitors and potentiate the activity of antibiotics.

Summary: Three novel matrine-derived alkaloids were isolated from the seeds of Sophora alopecuroides, showing anti-proliferative activities against human lung cancer cells by inducing apoptosis and cell-cycle arrest. The compounds appeared to exert their effects through a mitochondria-mediated apoptotic pathway, as indicated by various cellular and molecular assays.

Summary: A study on the seeds of Sophora alopecuroides uncovered fourteen matrine-based alkaloids, with eight being new compounds. Among them, alopecuroide F (1) was identified as the first dimeric matrine-type skeleton assembled via a unique connection. Biological evaluations showed that alopecuroide F (1) could dose-dependently inhibit the production of two pro-inflammatory cytokines.

Summary: A phytochemical investigation on the alkaloids from the water-soluble part of Sophora alopecuroides led to the discovery of forty matrine-type alkaloids, including eighteen new compounds. Among them, eight belong to rare bis-amide matrine-type alkaloids. Most isolates showed anti-neuroinflammatory activities, with compound 39 significantly suppressing TNF-α and IL-6 expression in a dose-dependent manner. Further mechanistic study revealed that compound 39 regulates the NF-κB signaling pathway by inhibiting the phosphorylation of I kappa B alpha and p65 subunit.

Summary: Efflux pumps are mechanisms of resistance in bacteria that can expel antibiotics to confer resistance. Overexpression of efflux pumps can lead to multi-drug resistance. By studying the gene regulation network associated with efflux, a mathematical model is presented and analyzed using time-dependent asymptotic analysis.

Summary: Antibiotic resistance is an increasingly urgent threat to human health, and multidrug efflux pumps are potential targets for combating this issue. The compound NSC-33353 has been identified as a potent inhibitor of efflux pumps, enhancing antibacterial activity in wild-type cells. Structural modifications of NSC-33353 have led to the development of compounds with improved inhibitory and substrate properties, offering insights into the duality of efflux substrate/inhibitor features and facilitating the development of new efflux pump inhibitors.

Summary: This study investigates the impact of internal ionic interactions on the antibiotic resistance of E. coli by studying a representative MFS efflux pump called MdfA. The researchers found that extending the salt bridge on the cytoplasmic side of MdfA resulted in elevated antibiotic resistance, which was further enhanced by combining it with an artificial mutation. By analyzing molecular dynamics simulations, they discovered that enhanced interactions led to a larger surface for MdfA's protonation, indicating a higher possibility for its activation.

Summary: Vanillin improves the organic solvent tolerance of Escherichia coli by enhancing the expression of AcrAB-TolC efflux pump, which is regulated by marA transcription factor. The DacrB mutant is more sensitive to vanillin, but its tolerance can be restored by introducing the acrB gene.

Summary: Seven new matrine-based alkaloids, Sophaloseedlines A-G (1-7), along with two known analogues, were isolated from Sophora alopecuroides seeds. The new structures were determined using spectroscopic data, electronic circular dichroism calculations, and X-ray crystallography. Notably, sophaloseedline A (1) features a novel rearranged alkaloid with unprecedented highly constructed tetracyclic fused ring skeleton. Additionally, new alkaloids 1 and 2 exhibited more potent antiviral activities against hepatitis B virus compared to matrine and positive control.

Summary: This study reveals that the activity of AcrR, the main transcriptional regulator of the multidrug efflux pump AcrAB-TolC, is controlled by the antimicrobial ethidium bromide and polyamines produced by E. coli. AcrR regulates the expression of multiple genes involved in efflux and detoxification.

Summary: This study investigated the antibacterial activity of honey, total alkaloids of Sophora alopecuroides (TASA) and matrine alone and in combination with antibiotics against multidrug-resistant Pseudomonas aeruginosa strains. The results showed a synergistic effect of matrine and TASA-honey in combination with ciprofloxacin against all tested isolates. These natural products can be used against overactivating MexAB-OprM but not MBL-producing P. aeruginosa isolates.
