The genus Mycobacterium includes human pathogens (Mycobacterium tuberculosis and Mycobacterium leprae) and environmental organisms known as non-tuberculous mycobacteria (NTM) that, when associated with biomaterials and chronic disease, can cause human infections. A common pathogenic factor of mycobacteria is the formation of biofilms. Various molecules are involved in this process, including glycopeptidolipids, shorter-chain mycolic acids, and GroEL1 chaperone. Nutrients, ions, and carbon sources influence bacterial behavior and have a regulatory role in biofilm formation. The ultrastructure of mycobacterial biofilms can be studied by confocal laser scanning microscopy, a technique that reveals different phenotypic characteristics. Cording is associated with NTM pathogenicity, and is also considered an important property of M. tuberculosis strains. Mycobacterial biofilms are more resistant to environmental aggressions and disinfectants than the planktonic form. Biofilm-forming mycobacteria have been reported in many environmental studies, especially in water systems. NTM cause respiratory disease in patients with underlying diseases, such as old tuberculosis scars, bronchiectasis, and cystic fibrosis. Pathogens can be either slowly growing mycobacteria, such as Mycobacterium avium complex, or rapidly growing species, such asMycobacteriumabscessus. Another important biofilm-related group of infections are those associated with biomaterials, and in this setting the most frequently isolated organisms are rapidly growingmycobacteria. M. tuberculosis can develop a biofilmwhich plays a role in the process of casseous necrosis and cavity formation in lung tissue. M. tuberculosis also develops biofilms on clinical biomaterials. Biofilm development is an important factor for antimicrobial resistance, as it affords protection against antibiotics that are normally active against the same bacteria in the planktonic state. This antibiotic resistance of biofilm-forming microorganisms may result in treatment failure, and biofilms have to be physically eradicated to resolve the infection. New strategies with potential antibiofilm molecules that improve treatment efficacy have been developed. A novel antibiofilm approach focuses on Methylobacterium sp. An understanding of biofilm is essential for the appropriate management of patients with many NTM diseases, while the recent discovery of M. tuberculosis biofilms opens a new research field.

Atypical Mutational Spectrum of SARS-CoV-2 Replicating in the Presence of Ribavirin

Summary: We report that ribavirin has an inhibitory and mutagenic activity on SARS-CoV-2-infected Vero cells, with a therapeutic index higher than 10. Deep sequencing analysis shows an increase in mutations, but not deletions, and modification of diversity indices in the presence of ribavirin. The major mutation types enhanced by ribavirin are A -> G and U -> C transitions, opposite to the dominant G -> A and C -> U transitions observed in most RNA viruses. The implications of ribavirin's inhibitory activity and atypical mutational bias on SARS-CoV-2 for the search for synergistic anti-COVID-19 lethal mutagen combinations are discussed.

Summary: This study analyzed the demographic and microbiological variables of acute osteosynthesis infection after ankle fracture fixation and identified the distinctive features of patients with E cloacae infection. The results showed a high incidence of E cloacae infections, which mainly occurred in older patients and required a higher use of negative-pressure therapy after debridement.

Differences in In Vitro Bacterial Adherence between Ti6Al4V and CoCrMo Alloys

Summary: Prosthetic joint infection is a rare but costly problem. This study evaluated the adherence of different bacteria to two types of alloys commonly used in joint prostheses. The results showed that the interaction between bacteria and the alloys differed significantly, with the CoCrMo alloy having an aggregating effect on all bacteria and additional anti-adherent activity against Pseudomonas aeruginosa.

Summary: This study aimed to translate clinical research outcomes based on metallodrugs into clinical practice for the treatment of MRSA. The novel metallodrug 2P-Pt showed antibacterial effect against MRSA and inhibited biofilm formation. These findings suggest that 2P-Pt may be a promising therapeutic alternative for MRSA infections.

Subspecies Distribution and Antimicrobial Susceptibility Testing of Mycobacterium abscessus Clinical Isolates in Madrid, Spain: a Retrospective Multicenter Study

Summary: The study aimed to determine the subspecies distribution of Mycobacterium abscessus (MABS) and its correlation with antibiotic profiles. The clinical isolates included 50 (52.1%) MABS subsp. abscessus, 33 (34.4%) MABS subsp. massiliense, and 13 (13.5%) MABS subsp. bolletii. Resistance rates varied among different drugs, with amikacin, cefoxitin, linezolid, and imipenem showing high activity against MABS. The GenoType NTM-DR assay demonstrated a high level of agreement with the traditional drug susceptibility testing.

Bacteriophage-Antibiotic Combination Therapy against Pseudomonas aeruginosa

Pseudomonas aeruginosa, a pathogen that commonly infects immunocompromised individuals, can be effectively treated using bacteriophages alone or in combination with antibiotics. This review summarizes recent studies highlighting the synergistic effects of phage-antibiotic combinations and successful treatments.

Antibacterial Properties of Mesoporous Silica Nanoparticles Modified with Fluoroquinolones and Copper or Silver Species

Summary: Antibiotic resistance is a global problem, and this study aimed to synthesize and characterize seven materials based on silica mesoporous nanoparticles functionalized with fluoroquinolones, Cu2+ or Ag+ species. The antibacterial properties of these materials were evaluated against different strains including Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, and Pseudomonas aeruginosa. The Ag+ materials exhibited a broader spectrum of antibacterial activity, inhibiting even multi-drug resistant strains, and the material with phenytoin sodium showed potential in reducing biofilm development and inhibiting bacterial growth.

Molecular Diagnosis of Osteoarticular Implant-Associated Infection: Available Techniques and How We Can Use Them

Summary: The genus Mycobacterium includes deadly pathogens like tuberculosis and leprosy, but it also includes environmental microorganisms. Some of these environmental species can be human pathogens, and the study of their biofilms has increased in recent decades. The importance of glycopeptidolipids and lipidic molecules in the extracellular matrix of mycobacterial biofilms has been well established. Mycobacterial biofilms have clinical importance in chronic diseases, especially lung diseases and implant-related ones. The increasing resistance of mycobacteria in biofilm form has been proven, and new tools or approaches are needed to treat these patients.
