Diffusion kurtosis imaging allows the early detection and longitudinal follow-up of amyloid-β-induced pathology

Background: Alzheimer's disease (AD) is a progressive neurodegenerative disorder and the most common cause of dementia in the elderly population. In this study, we used the APP/PS1 transgenic mouse model to explore the feasibility of using diffusion kurtosis imaging (DKI) as a tool for the early detection of microstructural changes in the brain due to amyloid-beta (A beta) plaque deposition. Methods: We longitudinally acquired DKI data of wild-type (WT) and APP/PS1 mice at 2, 4, 6 and 8 months of age, after which these mice were sacrificed for histological examination. Three additional cohorts of mice were also included at 2, 4 and 6 months of age to allow voxel-based co-registration between diffusion tensor and diffusion kurtosis metrics and immunohistochemistry. Results: Changes were observed in diffusion tensor (DT) and diffusion kurtosis (DK) metrics in many of the 23 regions of interest that were analysed. Mean and axial kurtosis were greatly increased owing to A beta-induced pathological changes in the motor cortex of APP/PS1 mice at 4, 6 and 8 months of age. Additionally, fractional anisotropy (FA) was decreased in APP/PS1 mice at these respective ages. Linear discriminant analysis of the motor cortex data indicated that combining diffusion tensor and diffusion kurtosis metrics permits improved separation of WT from APP/PS1 mice compared with either diffusion tensor or diffusion kurtosis metrics alone. We observed that mean kurtosis and FA are the critical metrics for a correct genotype classification. Furthermore, using a newly developed platform to co-register the in vivo diffusion-weighted magnetic resonance imaging with multiple 3D histological stacks, we found high correlations between DK metrics and anti-A beta (clone 4G8) antibody, glial fibrillary acidic protein, ionised calcium-binding adapter molecule 1 and myelin basic protein immunohistochemistry. Finally, we observed reduced FA in the septal nuclei of APP/PS1 mice at all ages investigated. The latter was at least partially also observed by voxel-based statistical parametric mapping, which showed significantly reduced FA in the septal nuclei, as well as in the corpus callosum, of 8-month-old APP/PS1 mice compared with WT mice. Conclusions: Our results indicate that DKI metrics hold tremendous potential for the early detection and longitudinal follow-up of A beta-induced pathology.

Summary: In this study, the suitability of particle systems for segmenting the mitral valve from 3D TOE images was evaluated. The results showed accurate representation of the mitral valve and indicated a correlation between particle strength and leaflet thickness. This research contributes to the development of treatment strategies for mitral valve patients.

Tabu-DART: a dynamic update strategy for efficient discrete algebraic reconstruction

Summary: In X-ray computed tomography, we introduce an update strategy framework that allows for dynamic rules and increases control for divide-and-conquer methods for discrete tomography (DT). Through simulated and real data reconstruction experiments, we show that our approach yields similar or improved reconstruction quality compared to traditional methods, with substantially lower computational complexity.

ADEPT: Accurate Diffusion Echo-Planar imaging with multi-contrast shoTs

Summary: This article introduces a novel imaging and parameter estimation framework for accurate multi-shot diffusion MRI. The proposed framework, called ADEPT, enables fast diffusion MRI by allowing diffusion contrast settings to change between shots in a multi-shot EPI acquisition. ADEPT estimates diffusion parameter maps directly from the acquired k-space data and simultaneously accounts for shot-to-shot phase inconsistencies. Simulation and real-data experiments demonstrate that ADEPT provides more accurate and precise estimates of diffusion metrics compared to reference methods.

Optimal experimental design and estimation for q-space trajectory imaging

Summary: Tensor-valued diffusion encoding allows disentangling variations in diffusivity from microscopic anisotropy, orientation dispersion, and mixtures of multiple isotropic diffusivities. Two precision-optimized acquisition schemes are created to estimate the diffusion tensor distribution (DTD) parameters accurately and precisely. The weighted linear least squares (WLLS) estimator with the squared reciprocal of the predicted signal as weights outperforms conventional estimators in terms of accuracy and precision when appropriate constraints are used.

cuPARE: parametric reconstruction of curved fibres from glass fibre-reinforced composites

Summary: A new framework is proposed for reconstructing curved fibres from X-ray computed tomography data of glass fibre-reinforced composites. It can detect fibres in a low-dose, low-resolution CT scan, and estimate their parameters directly from a few high-resolution 2D projection images. The framework is validated on both simulated and real data, showing robustness to noise and requiring less than 10 high-resolution projections for accurate estimates. It can also improve existing estimation frameworks based on full 3D scans.

Systematic review of reconstruction techniques for accelerated quantitative MRI

Summary: This article systematically reviews the techniques for addressing undersampling artifacts in accelerated quantitative magnetic resonance imaging (qMRI). A literature search was conducted to identify studies proposing reconstruction techniques for accelerated qMRI published before July 2022. The reviewed studies are categorized based on methodology, and a technical overview of each category is provided. The increasing number of articles proposing new techniques for accelerated qMRI reconstruction indicates the importance of acceleration in qMRI.

Summary: Since the outbreak of SARS-CoV-2, several rapidly spreading variants of concern (VOC) have emerged. A single dose of Ad26.COV2.S has shown to protect against the Gamma and Delta variants in naive hamsters, supporting its efficacy in humans against these VOC. However, an Omicron BA.1-based booster does not improve immunogenicity and efficacy against Omicron BA.2 compared to an Ad26.COV2.S booster in hamsters with pre-existing immunity to SARS-CoV-2.

A preparation pulse for fast steady state approach in Actual Flip angle Imaging

Fiber Orientation Estimation from X-ray Dark Field Images of Fiber Reinforced Polymers Using Constrained Spherical Deconvolution

Summary: This article discusses the challenges of studying fiber reinforced polymer properties using X-ray computed tomography (XCT) and proposes anisotropic dark field tomography via grating based interferometry (GBI) and edge illumination phase contrast imaging as solutions. It is shown through Monte Carlo simulation experiments that constrained spherical deconvolution (CSD) outperforms the traditional Funk-Radon transform (FRT) in accurately estimating fiber orientations.

Summary: The stability and manufacturability of a subunit vaccine antigen were further optimized, and the potency with different adjuvants was evaluated. It was found that a vaccine formulation with aluminum hydroxide or specific ratio of SLA-LSQ induced broad neutralizing antibodies after the second immunization. The study provided insights into potential adjuvant formulations for an H1 stem-based vaccine.
