Bacterial resistance is a major clinical problem, which is compounded by both a lack of new antibiotics and emergence of multi- and extremely-drug resistant microbes. In this context, non-toxic nanoparticles could play an important role in conferring protection against bacterial infections and in this study we have made an attempt to show the usefulness of jacalin capped platinum nanoparticles in protecting zebrafish against multiple infections with Aeromonas hydrophila. Our results also indicate that use of nanoparticles promotes adaptive immune response against the pathogen, so much so that zebrafish is able to survive repetitive infection even after twenty one days of being treated with jacalincapped platinum nanoparticles. This is significant given that platinum salt is not antibacterial and jacalin is non-immunogenic. Our study for the first time reveals a novel mechanism of action of nanoparticles, which could form an alternate antibacterial strategy with minimal bacterial resistance.

Shrimp lectin-conjugated copper sulfide nanoparticles enhance immune response and gene expression in Etroplus suratensis infected with Aeromonas hydrophila

Preetham Elumalai, Abdul Salam Rubeena, Sreeja Lakshmi, Veerappan Anbazhagan, Jesu Arockiaraj, Mani Divya, Sekar Vijayakumar, Baskaralingam Vaseeharan

Summary: In this study, CuS nanoparticles were conjugated with shrimp lectin (Md-Lec) to enhance their antimicrobial activity against aquatic pathogens. The conjugation led to a four-fold reduction in minimum inhibitory concentration (MIC) and decreased hydrophobicity index and EPS production of the pathogens. Furthermore, treated fishes showed elevated non-specific immune parameters and upregulated immune gene expression, suggesting the potential of Md-Lec-pCuS NPs as an immunostimulant for disease-free fish production in aquaculture industry.

Metal Sulfide Nanoparticles Based Phytolectin Scaffolds Inhibit Vulvovaginal Candidiasis Causing Candida albicans

Summary: Vulvovaginal candidiasis is a common infection caused by Candida species, especially Candida albicans. Plant protein-based nano scaffolds were found to effectively mitigate fluconazole-resistant Candida albicans infections. Lectin-functionalized copper sulfide nanoparticles showed lower inhibitory concentrations and synergistic effects with antifungal drugs like micafungin.

Summary: A novel method involving the combination of butea monosperma seed lectin and antimicrobial lipid was proposed to overcome serum protein interference and demonstrated significant antimicrobial activity against uropathogenic Escherichia coli. The conjugates showed good biocompatibility and biosafety in a zebrafish model, exhibiting high antimicrobial intensity and low likelihood of resistance development.

Artocarpus integrifolia seed lectin enhances membrane damage, oxidative stress and biofilm inhibition activity of silver nanoparticles against Staphylococcus aureus

Summary: Functionalizing nanoparticles to target bacteria surfaces is an effective approach to combat drug-resistant pathogens. This study synthesized N-myristoyltaurine capped silver nanoparticles (AgNPs) and functionalized them with jacalin from Artocarpus integrifolia seeds. The functionalized AgNPs (JAgNPs) had a lower minimum inhibitory concentration and showed superior biofilm inhibition activity against S. aureus compared to AgNPs, suggesting a unique way to use nanosilver for treating infections associated with S. aureus biofilm.

Summary: Aggregation-induced emissive fluorophores have attracted attention due to their controllable solid-state fluorescence. This study explored the impact of interlinking, positional isomerism, and acceptor structure on the fluorescence tuning and switchability. Results revealed that changing the acceptor group led to fluorescence tuning between 534 and 614 nm, with closer spacing and tighter packing enhancing fluorescence efficiency.

Antimicrobial lipid capped copper sulfide nanoparticles display enhanced bactericidal effect against Carbapenem-Resistant Acinetobacter baumannii

Summary: In this study, cationic antimicrobial lipid was used as a stabilizing agent in the preparation of copper sulfide nano particles for the first time. The cN16E-CuS exhibited excellent antimicrobial activity against Carbapenem-Resistant Acinetobacter baumannii, with a two-fold lower MIC compared to pure cN16E. Mechanistic study revealed that cN16E-CuS produced excess reactive oxygen species, leading to superior antimicrobial activity at lower concentrations.

Summary: The study demonstrates that BcN16E is more effective than cN16E in impairing biofilm-associated virulence factors and exerting antimicrobial activity. This finding emphasizes the potential of phytolectin in enhancing anti-virulence strategies against cross-kingdom biofilm-related infections.

Excellent synergistic activity of a designed membrane acting pyridinium containing antimicrobial cationic N-acylethanolamine with isoniazid against mycobacterium

Summary: The introduction of new cationic N-acylethanolamines showed promising antimicrobial activity against Mycobacterium smegmatis, with the minimum inhibitory concentration decreasing as the N-acyl chain length increased. Additionally, cNAEs in combination with isoniazid exhibited synergistic activity by disrupting bacterial membrane, generating reactive oxygen species, and depolarizing the membrane. Hemolytic study confirmed that cN16E is non-toxic to human red blood cells.
