Canine hepatocellular carcinoma (HCC) is the most common primary hepatic tumour in dogs. MicroRNA (miRNA) dysregulation has been reported in human HCC and shown to have diagnostic and prognostic value; however, there are no data on miRNA expression in canine HCC. The aim of the present study was to investigate differentially expressed miRNAs in canine HCC. Analysis of miRNA expression in canine HCC tissues and cell lines by quantitative reverse transcription PCR showed that miR-1, miR-122, let-7a, and let-7g were downregulated, whereas miR-10b and miR-21 were upregulated in canine HCC. MET is one of the target genes of miR-1. MET was upregulated in canine HCC at the gene and protein levels, and a significant correlation between the concomitant downregulation of miR-1 and upregulation of MET was observed. Fast/intermediate-proliferating canine HCC cell lines had higher MET gene and protein expression levels than the slow-proliferating cell line. These findings suggest that miRNAs are differentially expressed in canine HCC, and that the miR-1/MET pathway may be associated with canine HCC cell proliferation.

Identification of melanoma-specific exosomal miRNAs as the potential biomarker for canine oral melanoma

Al Asmaul Husna, Md Mahfuzur Rahman, Yu-Chang Lai, Hui-Wen Chen, Md Nazmul Hasan, Takayuki Nakagawa, Naoki Miura

Summary: This study identified several differentially expressed miRNAs in exosomes of canine melanoma cells, with certain miRNAs significantly increased in plasma exosomes of metastatic melanoma patients. These miRNAs could serve as potential biomarkers for identifying metastatic melanoma and improving prognosis, with sensitivity and specificity over 85% for distinguishing between non-metastatic and metastatic patients.

Characteristics and outcome of suspected cerebrovascular disease in dogs: 66 cases (2009-2016)

T. Ozawa, N. Miura, H. Hasegawa, T. Uemura, Y. Nakamoto, M. Tsujio, T. Takeuchi, M. Shiraishi

Summary: This study characterized the clinical signs of suspected cerebrovascular disease in dogs by analyzing medical records from November 2009 to December 2016. Seizure was identified as an important clinical sign, and a significant seasonal variation in the number of diagnosed cases was observed in Japan. The findings highlight the need for additional research in this area to further understand and manage cerebrovascular disease in dogs.

Micro RNA differential expression profile in canine mammary gland tumor by next generation sequencing

Hui-Wen Chen, Yu-Chang Lai, Md Mahfuzur Rahman, Al Asmaul Husna, Md Nazmul Hasan, Naoki Miura

Summary: This study identified four differentially expressed miRNAs in canine mammary gland tumors using NGS and qPCR, and analyzed their potential target genes and pathways using gene ontology and KEGG database. The results suggest that these four miRNAs could be promising biomarker candidates for canine mammary gland tumors.

Long non-coding RNA and transfer RNA-derived small fragments in exosomes are potential biomarkers for canine oral melanoma

Al Asmaul Husna, Md Mahfuzur Rahman, Hui-Wen Chen, Md Nazmul Hasan, Takayuki Nakagawa, Naoki Miura

Summary: Novel small non-coding RNAs (sRNAs) have diverse regulatory and functional roles in human and canine oncology. In our study on canine oral melanoma, we identified differentially expressed novel sRNAs in exosomes and validated their potential as biomarkers for accurate diagnosis of this disease.

Protective Effect on Pancreatic Acinar Cell by Maintaining Cardiac Output in Canine Heart Failure Model With Decreased Pancreatic Blood Flow

Aritada Yoshimura, Takahiro Ohmori, Daiki Hirao, Miori Kishimoto, Tomoko Iwanaga, Naoki Miura, Kazuhiko Suzuki, Ryuji Fukushima

Summary: This study investigated the relationship between cardiac function, pancreatic blood flow (PBF), and histopathological changes in pancreatic acinar cells. The findings showed that the decrease in PBF caused by cardiac dysfunction directly led to pancreatic acinar cell atrophy. Maintaining PBF is clinically important for treating dogs with heart failure, and these findings provide a reliable basis for developing new therapeutic strategies, such as pancreatic protection, for heart failure in dogs.

microRNAs Are Abundant and Stable in Platelet-Rich Fibrin and Other Autologous Blood Products of Canines

Indre Jasineviciute, Md Nazmul Hasan, Juozas Grigas, Arnoldas Pautienius, Arunas Stankevicius, Judita Zymantiene, Naoki Miura

Summary: This study aimed to investigate the existence of microRNAs (miRNAs) in platelet-rich fibrin (PRF) and the stability of canine autologous blood products under different storage conditions. Total RNAs were isolated using newly adapted protocols from PRF and other autologous blood products, and quantitative real-time polymerase chain reaction analysis was performed to detect miRNAs. miR-16, miR-21, miR-155, and miR-146a were found to be abundant in PRF and other autologous blood products of canines. Furthermore, these miRNAs were shown to maintain stability under extended freezing temperatures of -30 degrees C for at least one month. Our findings suggest that PRF may serve as a stable resource for various miRNAs in canines.

YRNA and tRNA fragments can differentiate benign from malignant canine mammary gland tumors

M. D. Nazmul Hasan, Md Mahfuzur Rahman, Al Asmaul Husna, Nobuhiro Nozaki, Osamu Yamato, Naoki Miura

Summary: This study investigated the expression of ncRNAs other than miRNAs in different histologic subtypes of canine mammary gland tumors (MGT). Three aberrantly expressed ncRNAs were identified as potential biomarkers for differentiating MGT subtypes. YRNA and tRFs expression levels were found to be decreased in metastatic compared to primary MGT cell lines.
