If left unchecked, prediabetic hyperglycemia can progress to diabetes and often life-threatening attendant secondary complications. Central to the process of glucose homeostasis are pancreatic beta cells, which sense elevations in plasma glucose and additional dietary components and respond by releasing the appropriate quantity of insulin, ensuring the arrest of hepatic glucose output and glucose uptake in peripheral tissues. Given that b cell failure is associated with the transition from prediabetes to diabetes, improved b cell function ('compensation') has a central role in preventing type 2 diabetes mellitus (T2DM). Recent data have shown that both insulin secretion and b cell mass dynamics are regulated by the liver kinase B1-AMP-activated kinase (LKB1-AMPK) pathway and related kinases of the AMPK family; thus, an improved understanding of the biological roles of AMPK in the b cell is now of considerable interest.

Summary: This pilot study evaluated the neutralizing ability of plasma collected from Canadian blood donors during the first wave of the pandemic against later SARS-CoV-2 variants of concern. Results showed that six specimens with high neutralizing titers against wild-type SARS-CoV-2 had reduced neutralizing capacity against beta variants, and three of them also had reduced capacity against gamma variants.

Summary: This article reports the preclinical development of a lipid nanoparticle-formulated SARS-CoV-2 mRNA vaccine, PTX-COVID19-B. It was chosen among three candidates after showing the strongest immune response in mouse vaccination. Further tests in mice and hamsters demonstrated that PTX-COVID19-B induced robust immune responses and provided complete protection against SARS-CoV-2 infection.

Neutralization against Omicron variant in transplant recipients after three doses of mRNA vaccine

Summary: The Omicron variant of the SARS-CoV-2 virus has become the dominant circulating strain globally, and three doses of mRNA COVID-19 vaccine are recommended for transplant recipients. However, there is limited research on the immunogenicity of mRNA vaccines specifically against the Omicron variant. This study analyzed the neutralization effects of Omicron in transplant recipients who received three doses of mRNA-1273 vaccine, and the results showed poor neutralizing responses against the Omicron variant in these patients.

Summary: Limited information is available on the impact of immunosuppressants on COVID-19 vaccination in patients with immune-mediated inflammatory diseases. This observational cohort study investigated the immunogenicity of SARS-CoV-2 mRNA vaccines in adult patients with various inflammatory diseases. The study found that antibody and T cell responses to SARS-CoV-2 were lower in patients with immune-mediated inflammatory diseases compared to healthy controls, especially those treated with anti-TNF therapy. The findings support the administration of a third dose of the mRNA vaccine and continuous monitoring of immunity in these patient groups.

SARS-CoV-2 Virus-Like Particle Neutralizing Capacity in Blood Donors Depends on Serological Profile and Donor-Declared SARS-CoV-2 Vaccination History

Summary: This study aimed to investigate the levels of neutralizing titers and breadth of antibody protection against wild-type and variant SARS-CoV-2 in Canadian blood donors from January to March 2021. The results showed that during this period, a small proportion of Canadians had received a second dose of the vaccine, resulting in varying levels of humoral protection. The study found elevated levels of neutralizing capacity in vaccinated blood donors, but reduced neutralization against some SARS-CoV-2 strains. The research highlights the importance of vaccination in generating a broad neutralizing antibody response against SARS-CoV-2 variants.
