Peroxidase-like activity of 2′,7′-difluorofluorescein and its application for galactose detection

The peroxidase-like activity of 2',7'-difluorofluorescein (DFF), was investigated using 3,3',5,5'-tetramethylbenzidine (TMB) as a chromogenic substrate in the presence of H2O2. DFF could catalyze oxidization of TMB by H2O2 to produce a blue colored oxidation product. Effect of various reaction conditions, such as pH, temperature, H2O2 concentration and reaction time on the catalytic activity of DFF was studied. The peroxidase-like activity of DFF was found to follow Michaelis-Menten kinetics, and its catalysis accorded with ping-pong mechanism. The calculated kinetic parameters (K-cat) of DFF catalysis showed higher peroxidase-like activity than fluorescein and 2',7'-dichlorofluorescein (DCF). According to the radical capture and electron spin resonance (ESR) spectroscopy results, we confirmed that hydroxyl radical (center dot OH) is the active specie of catalytic process. It is known that the oxidation of galactose by galactose oxidase (GAOx) enzyme leads to the formation of H2O2, the H2O2 released in this reaction was consequently quantified using DFF as peroxides mimics and TMB as the chromogen. Thus, a combination of above two reactions was exploited to establish a method for galactose detection. Under the optimum conditions, the linear range of this method was from 10 mu M to 20 mM with the detection limit down to 3 mu M. Moreover, the developed method was applied to detect galactose in urine samples. Our work will facilitate the utilization of DFF intrinsic peroxidase-like activity in medical diagnostics and biotechnology.

Summary: A series of DL-serine covalently modified multinuclear lanthanide implanted arsenotungstates were synthesized and exhibited rapid photochromic behaviors. These compounds contain a basic polyoxoan-ion structure and form one-dimensional linear chainlike structures. The colors of the compounds changed quickly under UV irradiation and gradually recovered in the dark.

Summary: A large case series revealed that over 80% of newborns with prolonged jaundice had genetic disorders. The study aimed to investigate the distribution of causes and outcomes in neonates with prolonged neonatal jaundice. Genetic disorders were the leading cause in neonates with prolonged conjugated hyperbilirubinaemia, while a small percentage of neonates with prolonged unconjugated hyperbilirubinaemia had a genetic cause. Genetic data can modify the clinical diagnosis and guide disease management, potentially improving outcomes for these infants.

Sequencing of the ZMYND15 gene in a cohort of infertile Chinese men reveals novel mutations in patients with teratozoospermia

Summary: A study was conducted to investigate the relationship between ZMYND15 variants and male infertility. Whole exome sequencing and other methods were used to identify potential pathogenic variations of ZMYND15 in infertile men. The study found several ZMYND15 variants in the infertile patients, and three deleterious biallelic variants were detected in affected individuals with oligoasthenoteratozoospermia. The study also revealed the potential signaling pathway of ZMYND15 in regulating spermatogenesis.

Peer relationship and adolescents' smartphone addiction: the mediating role of alienation and the moderating role of sex

Furthermore, sex moderates both the direct and indirect associations between peer relationships and smartphone addiction.

Summary: This study evaluated the weed control effect of four pyroligneous acid mixtures and found that its concentration significantly affected the efficacy. Both greenhouse and field trials showed that the pyroligneous acid had good control effects on the tested plants and weeds. The results suggest that pyroligneous acid has the potential to be developed as an organic herbicide.

Summary: Using data from e+e- annihilation at a center-of-mass energy of 3.773 GeV with the BESIII detector, the branching fractions of D0 -> pi+pi+pi-X and D+ -> pi+ pi+ pi-X inclusive decays have been measured for the first time. The pions from K0S decays in the pi+pi+pi- system were excluded and X represents any possible combination of particles. The branching fractions of D0(D+) -> pi+pi+pi-X are determined to be B(D0 -> pi+pi+pi-X) = (17.60 +/- 0.11 +/- 0.22)% and B(D+ ->pi+pi+pi-X) = (15.25 +/- 0.09 +/- 0.18)%, with the first uncertainties being statistical and the second systematic.

Summary: This study investigated the hepatoprotective effects of peptides from Antarctic krill (AKP) on carbon tetrachloride (CCl4)-induced acute liver injury (ALI) in mice and found that AKP pretreatment significantly attenuated CCl4-induced liver injury. This protective effect was associated with the activation of the Nrf2/HO-1 pathway and the improvement of redox balance.

Flexible control of an ultrastable levitated orbital micro-gyroscope through orbital-translational coupling

Summary: Introducing rotational degree of control into conventional optical tweezers offers new possibilities in physics, optical manipulation, and life science. Previous rotational schemes relied on microsphere anisotropy, limiting their applications. This study demonstrates the first experimental realization of orbiting a homogeneous microsphere using angular momentum in a transversely rotating optical trap. This method enables the exploration of orbital-translational coupling and the creation of an ultra-stable micro-gyroscope.

Pseudo-LiDAR for Visual Odometry

Summary: In this study, a fluorescent biosensing system for high sensitivity detection of MT was established using a new Ag+-stabilized DNA triplex probe (Ag+-SDTP) combined with catalytic hairpin assembly (CHA) and CRISPR/Cas12a signal enhancements. The sensing method is able to selectively detect MT from other non-specific molecules and can achieve low level detection of MT in diluted human serums, demonstrating its potential for monitoring disease-specific MT biomarker at trace levels.
