A simple, precise and sensitive method in which, a nano optical sensor binuclear Pt-2-aminobenzimidazole-bipyridine, Pt(abi)(bpy) complex doped in sol gel is used for the early diagnosis of liver cancer. The idea depends on the assessment of the concentration of alpha fetoprotein (AFP) in the serum samples of different liver patients. The nano binuclear Pt(abi)(bpy) has strong emission spectrum upon excitation at 380 nm in distilled water. The assessment of alpha fetoprotein (AFP) depends on the quenching of the emission spectrum of the optical sensor at 610 nm in water by the alpha fetoprotein (AFP). The calibration plot was achieved over the concentration 5.0 - 350 U L-1 with a correlation coefficient of 0.997 and a detection limit of 3.0 U L-1. The method was used satisfactorily for the diagnosis of liver cancer in a number of serum samples collected from various patients and health state; healthy (<= 30 U L-1), Virus B (42.2-69.5 U L-1), Virus C (75.7-98.4 U L-1), Cirrhosis (112-147 U L-1) and HCC (185.2-349.6 U L-1). Furthermore, the assessment of the alpha-fetoprotein by the proposed method increases its sensitivity (92.88%) and specificity (91.41%) for early diagnosis of HCC.

Rituximab alleviates increased disomic sperm in DBA/1J mouse models of rheumatoid arthritis via restoration of redox imbalance

Sabry M. Attia, Mohammed A. Al-Hamamah, Mohamed S. M. Attia, Abdulrazaq Alanazi, Sheikh F. Ahmad, Mushtaq A. Ansari, Ahmed Nadeem, Saleh A. Bakheet, Gamaleldin I. Harisa

Summary: Compared to the general population, arthritis patients have a higher risk of fertility abnormalities. Rituximab, a commonly used treatment, has not been fully studied in terms of its effect on germ cells. This study aimed to investigate the impact of rituximab on aneuploidy and redox imbalance in arthritic mouse sperm. Results showed that rituximab did not induce aneuploidy, while arthritis significantly increased the frequency of disomy. Rituximab treatment reduced the levels of disomic sperm and restored markers of oxidative stress. Furthermore, arthritis severity was reduced after rituximab treatment.

Aflatoxin B1 Exacerbates Genomic Instability and Apoptosis in the BTBR Autism Mouse Model via Dysregulating DNA Repair Pathway

Ali A. Alshamrani, Mohammad Y. Alwetaid, Mohammed A. Al-Hamamah, Mohamed S. M. Attia, Sheikh F. Ahmad, Majed A. Algonaiah, Ahmed Nadeem, Mushtaq A. Ansari, Saleh A. Bakheet, Sabry M. Attia

Summary: The pathophysiology of autism is influenced by a combination of environmental and genetic factors. In addition, individuals with autism have a higher risk of developing cancer, possibly due to Aflatoxin B1 (AFB1) exposure. This study investigated the effects of AFB1 on genomic instability and apoptosis in a mouse model of autism. The findings showed that AFB1 exposure increased micronuclei generation, oxidative DNA strand breaks, and apoptosis in the mice, indicating a disturbed redox balance and dysregulation in the DNA damage/repair pathway. These results suggest that AFB1-related genomic instability may accelerate cancer development and that mitigating redox imbalance and DNA damage/repair dysregulation could be potential therapeutic approaches.

Summary: Alzheimer's disease is a major cause of senile dementia and is characterized by the aggregation of neurofibrillary tangles and amyloid plaques in the brain. The A beta 1-42 peptide is considered the primary cause of neurotoxicity due to its insolubility and subsequent aggregation. A beta 1-42 and A beta 1-40 have been identified as relevant biomarkers for AD diagnosis, and SurfaceAssisted Laser Desorption/Ionization Mass Spectrometry (SALDI-MS) was used to monitor the A beta 1-42/A beta 1-40 ratio without prior sample treatment.
