Metabolomics analysis of developing Garcinia mangostana eseed reveals modulated levels of sugars, organic acids and phenylpropanoid compounds

Mangosteen seeds are categorized as recalcitrant as they are sensitive to desiccation and low temperature. The seeds also do not go through the final developmental phase of maturation drying, hence, they are actively metabolic when they are shed. As such, metabolomics analysis could be useful to further understand mangosteen seed development. In this study, we used liquid chromatography-mass spectrometry and gas chromatography mass spectrometry approaches to analyze the profiles of polar compounds of mangosteen seeds harvested at four different stages of development (i.e. six, eight, 10 and 12 weeks after anthesis) and in mature seeds at 14 weeks. Principal component analysis (PCA) suggested temporal regulation of these metabolites during mangosteen seed development. Sugar levels such as glucose and fructose gradually increased throughout development, suggesting active metabolism until the mature stage. Organic acids such as citrate and malate as well as glycerol transiently increased during the seed growth phase which may imply their active energy and storage reserve production, respectively. High level of growth-related metabolites such as gamma-aminobutyric acid found at discrete developmental stages infer their roles in the seed development. Furthermore, accumulation of secondary metabolites was active between 10 and 12 weeks after anthesis particularly metabolites related to phenylpropanoid pathway. The elevated levels of gambiriin Al, rutin, chlorogenic acid, 3-O-feruloylquinic acid, thymo1-beta-D-glucopyranoside and guaico-beta-D-glucopyranoside may enhance seed protection and defense. In conclusion, mangosteen seed displays recalcitrant seed characteristics, with metabolism geared to be germination-ready in terms of resources and seed defense.

Discrimination and prediction of the chemical composition and the phytotoxic activity of Wedelia trilobata essential oil (EO) using metabolomics and chemometrics

Summary: This study used metabolomics and bioassay experiments to understand the chemical differences and inhibitory effects of W. trilobata essential oil (EO) grown in different conditions. It was found that EO grown in the field contained large amounts of monoterpenes, while EO grown in controlled conditions was rich in sesquiterpene hydrocarbons. The field-grown EO showed strong inhibitory effects on the growth of weedy rice and lettuce, while certain compounds like alpha-phellandrene, terpinen-4-ol, trans-pinocarveol, and pinocarvone were found to be responsible for the inhibitory activity.

Dose-Dependent Effect of Wedelia trilobata Essential Oil (EO) on Lettuce (Lactuca sativa L.) with Multivariate Analysis

Summary: This study investigated the metabolite changes in lettuce after exposure to W. trilobata essential oil using a GC/MS metabolomics approach. The results showed that the shoots and roots of lettuce responded differently to the essential oil, with changes in important metabolic pathways such as glycolysis and fatty acid metabolism. These findings suggest that lettuce may increase the levels of certain metabolites to enhance tolerance against the essential oil.

Bioinformatics analysis revealed the involvement of these amino acids in immune-related pathways.

Summary: This study investigated the secondary metabolite composition of Mitragyna speciosa leaves in different maturity stages using LC-ESI-TOF-MS. The results identified 86 putatively annotated metabolite features, including alkaloids, flavonoids, terpenoids, phenylpropanoids, carboxylic acids, glucosides, phenols, and phenolic aldehydes. The multivariate analysis showed a significant difference in metabolite levels between young and mature leaves. Several new alkaloids were tentatively identified, highlighting the diverse biological activities of M. speciosa. These findings contribute to a better understanding of its pharmaceutical potential.

Summary: Mangosteen exhibits unique ripening behavior and transcriptomic analysis reveals differentially expressed transcripts at different ripening stages. The fruit conserves carbon skeleton rather than energy production at the early ripening stage and upregulates transcripts related to anthocyanin and xanthone biosynthesis. This study provides insights into the regulation of ripening in mangosteen and could contribute to its commercial utilization and preservation.

Optimization of Ultrasound-Assisted Extraction for Antioxidant Activity in Relation to Rhoifolin Content of Fortunella polyandra using Response Surface Methodology (RSM)

Summary: This study aims to optimize the conditions of ultrasound-assisted extraction (UAE) for antioxidant activities and rhoifolin contents in Fortunella polyandra leaves extract. The results showed that the optimal extraction conditions for antioxidant activity were a 5-minute extraction time, a temperature of 30 degrees C, and a solvent ratio of 70:30 ethanol:water. The rhoifolin content and ferric reducing antioxidant potential (FRAP) assay shared the same optimum conditions at a 5-minute extraction time, a temperature of 30 degrees C, and a solvent ratio of 30:70 ethanol:water. Correlation study revealed no relationship between rhoifolin content and antioxidant activities in the extract.

Expression Analysis of OsWRKY11 and OsNAC2 Genes in New Malaysian Drought- tolerance Rice Cultivar MR303

Summary: In this study, we investigated the relative expression of OsWRKY11 and OsNAC2 genes in the local drought-tolerant rice variety MR303 and their effects on plant morphology and drought tolerance. The results showed that OsWRKY11 gene was highly expressed under drought stress, consistent with previous findings in Oryza sativa spp. Japonica. However, contrary to previous studies, OsNAC2 gene was also highly expressed instead of downregulated under drought stress in MR303.

Effects of paper pouches of different light transmittance on the phenolic synthesis and related gene expression in peach fruit

Summary: Bagging is a crucial cultivation measure that affects the quality of peach fruit by regulating exposure to light. This study investigated the effects of bagging on the content of phenolic compounds in peach fruit and identified the most sensitive compounds to bagging treatment. The study also explored the regulatory role of bagging on phenolic synthetic-related genes. The results showed that bagging affected the accumulation of different phenolics in different peach cultivars and light intensity played a significant role in the accumulation of anthocyanins and other phenolic substances in peach fruit. Additionally, the expression of structural genes and transcription factors related to phenolic synthesis was regulated by bagging and light conditions. These findings provide a foundation for understanding the regulation mechanism of light on the synthesis of phenolic compounds in peach fruit.

Estimation of above ground biomass, biophysical and quality parameters of spinach (Spinacia Oleracea L.) using Sentinel-2 to support the supply chain

Interactive effects of drought and cadmium stress on adzuki bean seedling growth, DNA damage repair, and Cd accumulation

Genotypic differences in water deficit effects on leaf and crown traits in mature field-grown cocoa

Summary: This study investigated the effects of water stress on cocoa trees and found that they are highly sensitive to water stress, with different genotypes exhibiting varied responses. Potassium application did not mitigate the negative effects of water stress. There was significant variation among genotypes in terms of leaf and crown traits, and positive interactions between genotype and irrigation were observed. These findings provide insights into the acclimation strategies of cocoa trees and can be useful for selecting drought-tolerant genotypes.
