Summary: Ulcerated cutaneous melanoma has a poor prognosis, and the underlying biology driving its aggressive behavior remains largely unexplored. This study aimed to investigate the unique miRNA profile in ulcerated melanoma and its correlation with biologically important target genes. The expression of miRNAs and mRNAs in ulcerated and non-ulcerated melanomas was assessed, and pathway enrichment and functional annotations were determined. The results revealed differential expression of miRNAs and mRNAs in ulcerated melanoma, providing further insight into its molecular characteristics.

Summary: This study aims to elucidate the regulatory role of miRNAs in the mRNA signature of atrial fibrillation (AF) and identify potential targets for future therapies. Through combined miRNA-mRNA sequencing, differentially expressed miRNAs and their target genes were discovered in atrial tissues of patients with and without AF. Downregulated miRNAs were associated with upregulated gene expression involved in cell migration and glycosaminoglycan biosynthesis. In vitro inhibition experiments confirmed the effects of miRNAs on predicted target genes. These identified miRNAs can be explored as potential targets for anti-fibrotic therapies in AF.

Summary: This study investigated the molecular mechanisms of enhanced purine metabolism in mammals under high oxidative stress. Through analyzing the evolution of 117 purine metabolism-related genes, it was found that positively selected genes, convergent changes, and nonparallel amino acid substitutions may be associated with adaptation to oxidative stress in mammals. In particular, the evolution of convergent genes with cAMP and cGMP regulation roles may protect mammals from oxidative damage. Additionally, key purine salvage enzymes were identified as under positive selection in cetaceans, and functional adaptations in cetacean xanthine dehydrogenase were found to reduce radical oxygen species production during diving.
