Exposure to 1.8 GHz electromagnetic fields affects morphology, DNA-related Raman spectra and mitochondrial functions in human lympho-monocytes

Blood is a fluid connective tissue of human body, where it plays vital functions for the nutrition, defense and well-being of the organism. When circulating in peripheral districts, it is exposed to some physical stresses coming from outside the human body, as electromagnetic fields (EMFs) which can cross the skin. Such fields may interact with biomolecules possibly inducing non thermal-mediated biological effects at the cellular level. In this study, the occurrence of biochemical/biological modifications in human peripheral blood lympho-monocytes exposed in a reverberation chamber for times ranging from 1 to 20 h to EMFs at 1.8 GHz frequency and 200 V/m electric field strength was investigated. Morphological analysis of adherent cells unveiled, in some of these, appearance of an enlarged and deformed shape after EMFs exposure. Raman spectra of the nuclear compartment of cells exposed to EMFs revealed the onset of biochemical modifications, mainly consisting in the reduction of the DNA backbone-linked vibrational modes. Respirometric measurements of mitochondrial activity in intact lympho-monocytes resulted in increase of the resting oxygen consumption rate after 20 h of exposure, which was coupled to a significant increase of the FoF1-ATP synthase-related oxygen consumption. Notably, at lower time-intervals of EMFs exposure (i.e. 5 and 12 h) a large increase of the proton leak-related respiration was observed which, however, recovered at control levels after 20 h exposure. Confocal microscopy analysis of the mitochondrial membrane potential supported the respiratory activities whereas no significant variations in the mitochondrial mass/morphology was observed in EMFs-exposed lympho-monocytes. Finally, altered redox homeostasis was shown in EMFs-exposed lympho-monocytes, which progressed differently in nucleated cellular subsets. This results suggest the occurrence of adaptive mechanisms put in action, likely via redox signaling, to compensate for early impairments of the oxidative phosphorylation system caused by exposure to EMFs. Overall the data presented warn for health safety of people involved in long-term exposure to electromagnetic fields, although further studies are required to pinpoint the leukocyte cellular subset(s) selectively targeted by the EMFs action and the mechanisms by which it is achieved.

Summary: Fourier transform infrared micro-spectroscopy is a valuable tool for studying changes in human cells after exposure to ionizing radiation, and recent attention has been focused on its use in cells exposed to proton beams. The use of low-e-substrates slides for cell culture has been proposed to simplify and speed up the irradiation and measurements phases. Analyzing spectra obtained in transflection geometry reveals significant changes in DNA, lipids, and protein components of cells.

Comparison of FTIR spectra of different breast cell lines to detect spectral biomarkers of pathology

Summary: This study compares the FTIR spectra of different breast cell lines and finds significant differences in absorption spectra, particularly in the amide I and II bands and the intensity of nucleic acid bands. The ratio of certain peaks at specific wavenumbers is able to differentiate between non-malignant, malignant, and metastatic cells. These findings suggest that FTIR microspectroscopy has potential as a tool for cytological diagnosis of breast cancer.

Keratinocyte cellular damage induced by pesticide doses below the cytotoxic level evidenced by electrical impedance and broadband dielectric spectroscopy

Summary: The cellular response of a normal human keratinocyte cell line to non-cytotoxic doses of a deltamethrin-based pesticide was investigated using two electrical impedance spectroscopy techniques. The results showed that the membrane capacity and the impedance of the extracellular matrix increased with pesticide concentration, affecting electric current flow. Additionally, dielectric permittivity changes and an increase in neutral membrane lipids were observed in the cellular samples. Fluorescence measurements indicated a time increase in emission intensity, suggesting a membrane lipid response to repair cell damage caused by pesticide exposure. These spectroscopic techniques provide potential means for early diagnosis of cellular modifications related to pesticide exposure.

Classification of Healthy and Cancer Colon Cells Grown on Glass Coverslip by Means of Fourier Transform Infrared Spectroscopy and Multivariate Methods

Summary: Fourier Transform Infrared (FTIR) microspectroscopy has shown great potential in cytological diagnostics as it provides rapid and label-free biochemical information about cell samples. The use of FTIR as a clinical tool has been hindered due to the incompatibility with glass slides commonly used in clinical practice. However, recent research has shown the possibility of obtaining diagnostic information from the IR absorption spectra in the 2700-3700 cm(-1) range. This study applies FTIR in this range to diagnose human colon cells and successfully distinguishes healthy cells from cancerous ones using principal component analysis (PCA) and classification models.
