Early IgG/IgA response in hospitalized COVID-19 patients is associated with a less severe disease

Giorgio Fedele, Gianluca Russo, Ilaria Schiavoni, Pasqualina Leone, Eleonora Olivetta, Valentina Perri, Maria Antonella Zingaropoli, Maria Rosa Ciardi, Patrizia Pasculli, Claudio Maria Mastroianni, Paola Stefanelli

Summary: The kinetics of anti-SARS-CoV-2 antibody response were determined in fifteen hospitalized COVID-19 patients, showing that patients with milder symptoms were more prone to an early IgG/IgA response. Neutralization of Spike binding to ACE2 correlated with anti-Spike IgG and IgA, and the IgG and IgA antibody response persisted at the 6 months follow-up.

HMGB1 in Pediatric COVID-19 Infection and MIS-C: A Pilot Study

Laura Petrarca, Valeria Manganelli, Raffaella Nenna, Antonella Frassanito, Shira Ben David, Enrica Mancino, Tina Garofalo, Maurizio Sorice, Roberta Misasi, Fabio Midulla

Summary: This study described the clinical picture of MIS-C patients and compared their HMGB1 serum levels with patients with previous SARS-CoV-2 infection and healthy children. The results showed that the HMGB1 levels in MIS-C patients were significantly higher than those in COVID-19 patients and the control group, suggesting its involvement in inflammatory manifestations and suggesting HMGB1 as a potential biomarker and therapeutic target in patients with severe illness.

Serum NGF and BDNF in Long-COVID-19 Adolescents: A Pilot Study

Carla Petrella, Raffaella Nenna, Laura Petrarca, Francesca Tarani, Roberto Paparella, Enrica Mancino, Greta Di Mattia, Maria Giulia Conti, Luigi Matera, Enea Bonci, Flavio Maria Ceci, Giampiero Ferraguti, Francesca Gabanella, Christian Barbato, Maria Grazia Di Certo, Luca Cavalcanti, Antonio Minni, Fabio Midulla, Luigi Tarani, Marco Fiore

Summary: COVID-19 is an infectious disease caused by the SARS-CoV-2 virus. This study aimed to investigate the changes in NGF and BDNF levels in infected adolescents and their relationship with inflammatory biomarkers. The results showed that NGF serum content was lower in post-infected-COVID-19 individuals, and BDNF levels were higher in symptomatic and future long-COVID-19 girls. Inflammatory biomarkers and oxidative stress index remained unchanged in male and female adolescents.

T-Cell Mediated Response after Primary and Booster SARS-CoV-2 Messenger RNA Vaccination in Nursing Home Residents

Ilaria Schiavoni, Annapina Palmieri, Eleonora Olivetta, Pasqualina Leone, Anna Di Lonardo, Alessandra Mazzoli, Carmine Cafariello, Alba Malara, Anna Teresa Palamara, Raffaele Antonelli Incalzi, Graziano Onder, Paola Stefanelli, Giorgio Fedele

Summary: This study aimed to monitor mRNA vaccine-induced T-cell responses in Italian nursing home residents and assess the interaction between T-cell responses and humoral immunity. The study found that before the booster vaccination, 31 out of 34 residents had a positive cell-mediated immunity response, which increased to 28 out of 34 residents after the booster dose. This suggests that the booster vaccine dose can restore spike-specific T-cell responses in SARS-CoV-2 naive residents who responded poorly to the initial vaccination.

Summary: Contradictory results have been reported regarding the production of interferon (IFN) lambda and IFN gamma in COVID-19 patients. This study aimed to investigate the expression of IFN lambda 1-3 and IFN gamma mRNA in peripheral blood mononuclear cells (PBMCs) and bronchoalveolar lavages (BALs) of COVID-19 patients. The results showed lower levels of IFN lambda 1-3 and IFN gamma in PBMCs and BALs of severely ill patients compared to healthy donors. Secondary bacterial infections were associated with decreased IFN lambda in PBMCs but increased IFN lambda 3 in BALs. Dysregulation in IFN gamma, IFN lambda, and Toll-like receptors (TLRs) was observed in severe COVID-19 cases.

NRF2 Antioxidant Response and Interferon-Stimulated Genes Are Differentially Expressed in Respiratory-Syncytial-Virus- and Rhinovirus-Infected Hospitalized Children

Leonardo Sorrentino, Walter Toscanelli, Matteo Fracella, Marta De Angelis, Federica Frasca, Carolina Scagnolari, Laura Petrarca, Raffaella Nenna, Fabio Midulla, Anna Teresa Palamara, Lucia Nencioni, Alessandra Pierangeli

Summary: Respiratory syncytial virus (RSV) and human rhinovirus (HRV) are common causes of hospitalization in children, with RSV being more severe. The study investigated the interplay between interferon (IFN) responses and nuclear factor erythroid 2-related factor 2 (NRF2) antioxidant activity in RSV and HRV infections. The findings suggest that reduced NRF2 activation in RSV-infected infants may contribute to the severity of bronchiolitis.

Genetic diversity and its impact on disease severity in respiratory syncytial virus subtype-A and -B bronchiolitis before and after pandemic restrictions in Rome

Summary: This study aims to analyze the differences in respiratory syncytial virus (RSV) strains and their impact on bronchiolitis in children before and after the COVID-19 pandemic. The results show that RSV-A was more predominant in the 2021-2022 season compared to other seasons, while RSV-B was more predominant in the 2022-2023 season. The RSV-A strains in 2021-2022 were genetically distinct from pre-pandemic strains, while the RSV-B strains in 2022-2023 showed clear divergence from those in 2021-2022. RSV-A cases in the 2021-2022 season had a lower need for O2 therapy and intensive care compared to other seasons, while infants infected with RSV-B in the 2022-2023 season were more frequently admitted to intensive care units and needed O2 therapy.

No effects of COVID-19 on the development of type 1 diabetes autoimmunity and no evidence of an increased frequency of SARS-CoV-2 antibodies in newly diagnosed type 1 diabetes patients relative to healthy subjects

Tiberti Claudio, Nenna Raffaella, Tromba Valeria, Filardi Tiziana, Petrarca Laura, Silvestri Francesca, Fassino Valeria, Montuori Monica, Mancino Enrica, Lenzi Andrea, Midulla Fabio, Costantino Francesco, Morano Susanna

Summary: The study aimed to assess the relationship between SARS-CoV-2 infection and autoimmunity in type 1 diabetes (T1D) and the frequency of SARS-CoV-2 antibodies in newly diagnosed T1D patients. The results showed that none of the patients who contracted SARS-CoV-2 during the pandemic period tested positive for T1D autoantibodies. The frequency of SARS-CoV-2 antibodies in newly diagnosed T1D patients was not significantly different from healthy individuals.
