Summary: Dilated cardiomyopathy (DCM) has a genetic cause in up to 40% of cases, and the presence of cardiac inflammation is associated with a younger disease onset in genetic DCM patients. However, cardiac inflammation does not increase the risk of mortality, heart failure hospitalization, or life-threatening arrhythmias. This suggests that myocarditis may be an exogeneous trigger for DCM in patients with a genetic susceptibility or that cardiac inflammation represents an early phase of the disease.

Summary: This study investigated the effect of PNPLA3-I148M on the activation of hepatic stellate cell line LX-2 and the progression of liver fibrosis. The results showed that PNPLA3-I148M significantly promoted intracellular free cholesterol aggregation in LX-2 cells by decreasing cholesterol efflux protein (ABCG1) expression, leading to mitochondrial dysfunction. This study provides the first evidence that PNPLA3-I148M causes mitochondrial dysfunction in LX-2 cells through the accumulation of free cholesterol, thereby promoting LX-2 cell activation and the development of liver fibrosis.

Upregulation of TRPM8 in HCC is associated with worse clinicopathological characteristics. Inhibition or downregulation of TRPM8 can ameliorate hepatocarcinogenesis, providing a potential therapeutic target for HCC.

Summary: Mitochondrial DNA (mtDNA) is a powerful activator of the innate immune system, and its specific immunostimulatory properties and detection by cytosolic nucleic acid sensors are still poorly understood. Our study demonstrates that mitochondrial genome instability leads to the accumulation of Z-form DNA. Z-DNA Binding Protein 1 (ZBP1) stabilizes Z-form mtDNA and forms a cytosolic complex with cGAS, RIPK1, and RIPK3, resulting in sustained phosphorylation of STAT1 and signaling of type I interferon (IFN-I). Increased levels of Z-form mtDNA, ZBP1 expression, and IFN-I signaling are observed in cardiomyocytes exposed to Doxorubicin, a commonly used chemotherapeutic drug that causes cardiotoxicity. Remarkably, mice lacking ZBP1 or IFN-I signaling are protected from Doxorubicin-induced cardiotoxicity. Our findings highlight ZBP1 as a potential therapeutic target in heart failure and other disorders where mtDNA stress contributes to interferon-related pathology.

Summary: Mitochondria, responsible for generating energy in the form of ATP, have their own genetic material called mtDNA, along with nuclear genome, that determines their structure and function. Pathogenic variants in mtDNA or nuclear genome can cause mitochondrial disease, especially affecting high energy-demanding tissues like the heart. Mitochondrial cardiomyopathy, a result of genetic defects in either nDNA or mtDNA, manifests as hypertrophic or dilated cardiomyopathy and cardiac conduction defects. The complex pathophysiology of mitochondrial cardiomyopathy involves various abnormal processes such as deficient oxidative phosphorylation, ATP depletion, activation of alternative metabolic pathways, accumulation of reactive oxygen species, dysfunctional mitochondrial dynamics, abnormal calcium homeostasis, and mitochondrial iron overload.
