Summary: Inflammation and oxidative stress have been implicated in the development of depression. This study investigated the antidepressant effects and mechanisms of action of Edaravone (EDA), a free radical scavenger. The results demonstrated that EDA alleviated depressive and anxiety-like behaviors in a chronic social defeat stress (CSDS) depression model. Furthermore, EDA reduced neuronal loss, oxidative stress damage, and activation of inflammatory cytokines in the hippocampus and medial prefrontal cortex. The antidepressant effects of EDA were mediated through the Sirt1/Nrf2/HO-1/Gpx4 signaling pathway and Gpx4-mediated ferroptosis.

Summary: Deficiency of GRIM-19 correlates with poor survival outcomes in GC patients, and DNA hypermethylation is associated with downregulation of GRIM-19. GRIM-19 deficiency promotes GC metastasis through aberrant oxidative stress and ROS-dependent activation of the NRF2-HO-1 axis.

Summary: Kainate receptors (KARs) are ionotropic glutamate receptors that play important physiological roles in neuronal circuits, regulating EPSCs as post-synaptic receptors and controlling ion channels through non-canonical metabotropic actions. KARs, activated by endogenous glutamate, modulate neuronal circuit activity and plasticity under normal physiological conditions.

Summary: Chronic kidney disease (CKD) is a common chronic disease that can lead to kidney failure and death. Renal fibrosis, caused by oxidative stress and inflammation, is a key characteristic of CKD. The activation of Nrf2/HO-1 signal pathway has shown promising results in protecting the kidneys against fibrosis induced by oxidative stress and inflammation. This review summarizes studies on Nrf2/HO-1 and explores potential bioactive compounds that may attenuate renal fibrosis through HO-1 activation.

Summary: This study aimed to investigate the effects and mechanism of Edaravone dexborneol on cerebral ischemia reperfusion injury (CIRI). The results showed that Edaravone dexborneol improved learning and memory, reduced neuronal damage, and enhanced antioxidant, anti-inflammatory, and anti-apoptotic effects in CIRI mice. Furthermore, Edaravone dexborneol exerted its effects by activating the Nrf2/HO-1 signaling pathway and inhibiting NOX2 expression.

Summary: Oxidative stresses play a pivotal role in various pathophysiological conditions. The Keap1/Nrf2 pathway is a dual-edged sword with beneficial effects on both normal and cancer cells by regulating endogenous antioxidant enzymes. Targeting the Nrf2/HO-1 axis is considered a novel strategy for cancer treatment, especially in lung cancer.

Summary: The study found that DPP-4i may accelerate murine breast cancer metastasis by promoting an oncogenic ROS-NRF2-HO-1 axis, inducing aberrant oxidative stress through ROS overproduction. Therefore, this research not only reveals the oncogenic role of DPP-4i in breast cancer progression but also provides a new strategy to alleviate the adverse effects of DPP-4i by targeting HO-1.

Summary: Temporal lobe epilepsy (TLE) is a common form of epilepsy in adults, characterized by aberrant epileptogenic network formed by mossy fiber sprouting. Kainate receptors (KARs) play a prominent role in this network by facilitating the recruitment of neurons into coactive cell assemblies, contributing to the occurrence of epileptiform network events.

Summary: This study found that Maackiain, a natural compound from Sophora flavescens, protects against sepsis by reducing organ injury, inflammation, and oxidative stress. Maackiain activates the AMPK/Nrf2/HO-1 pathway, leading to its anti-inflammatory and antioxidant effects. Inhibition of this pathway abolishes the protective effects of Maackiain.

Summary: This study investigates the anti-inflammatory effects of Stevioside (STE) on osteoarthritis (OA) and its underlying mechanism. The findings suggest that STE can ameliorate OA development by inhibiting inflammation, potentially through the Nrf2/HO-1/NF-Kappa B signaling pathway. Treatment with STE also significantly reduces degeneration in the mouse destabilization of the medial meniscus (DMM) model.

Summary: Our study demonstrates the role of interleukin-33 (IL-33) in a demyelinating mouse model induced by cuprizone (CPZ), showing that IL-33 can alleviate the reduction of APC+ OLs and the decline of IL-33 levels in the corpus callosum, and promote the expression of myelin basic protein (MBP).
