Summary: This study found a significant positive correlation between CXCR7 expression and CAF activation markers in colonic tissues and transgenic mice. RNA sequencing revealed an increase in miR-146a-5p and miR-155-5p levels in CXCR7-overexpressing CRC cells and their exosomes. Furthermore, these CRC cell-derived miRNAs could be uptaken by CAFs via exosomes and promote their activation, leading to enhanced invasive capacity of CRC cells and pro-metastatic switch. The activation of CAFs by miR-146a-5p and miR-155-5p also facilitated tumor formation and lung metastasis of CRC in vivo. Therefore, these findings provide new insights into CXCR7-mediated CRC metastasis and suggest potential biomarkers and therapeutic targets for inhibiting CRC metastasis.

Summary: miR-34a and miR-34b/c, as direct targets of p53, play a crucial role in the tumor suppressive effects of p53. Inactivating miR-34a and/or miR-34b/c in colorectal cancer cell line HCT116 had significant effects on proliferation, migration, invasion, chemosensitivity, autophagy, and apoptosis. The concomitant deletion of miR-34a and miR-34b/c resulted in EMT signature enrichment, impaired gene repression by the p53-DREAM pathway, and elevated autophagy. A gene signature upregulated in cells with combined inactivation of miR-34a and miR-34b/c was associated with invasive colon cancer subtype CMS4, poor overall survival in CRC patients, and 5-FU resistance in CRC cell lines.

Summary: The incidence of colorectal cancer (CRC) is increasing globally. We identified SCNN1B as a down-regulated gene in CRC that acts as a tumor suppressor. SCNN1B mRNA and protein expression levels were decreased in primary CRC and CRC cells. In a cohort study, SCNN1B protein was an independent prognostic factor associated with favorable outcomes in CRC. Overexpression of SCNN1B in CRC cell lines inhibited cell proliferation, induced apoptosis and cell cycle arrest, and suppressed cell migration. Xenograft models confirmed the tumor suppressive function of SCNN1B in vivo. Mechanistically, SCNN1B suppressed the MAPK signaling pathway by inhibiting the activation of c-Raf, a downstream target of KRAS.
