Summary: We analyzed a large cohort of lung adenocarcinomas to study the clinicopathological and genomic features associated with metastasis and organotropism. We found that patients with metastasis were younger, male, and had specific histological subtypes and genomic alterations. Certain genes were correlated with a shorter time to metastasis in specific sites. Our findings also revealed common and unique genetic alterations between primary tumors and metastases. Only a small percentage of metastases had therapeutically actionable alterations not found in the primary tumors. Overall, our analysis highlights the complexity of clinicopathological features and tumor genomics in lung adenocarcinoma organotropism.

Summary: The study discovered that 16% of stage I lung adenocarcinoma cases had tumor necrosis, which was significantly associated with poorer prognosis. The ratio of necrotic area did not impact prognosis. Tumor necrosis, invasive component size, vascular invasion were independently linked to a higher risk of recurrence. Gene expression analysis revealed differentially expressed genes and biological processes in cases with necrosis. Cases with tumor necrosis showed more single nucleotide variants, especially in smokers.

Summary: Ossified cartilage is more susceptible to cancer infiltration, and the relationship between ossification pattern and cancer infiltration was investigated in this study. The results showed that there was no association between ossification pattern and cancer infiltration, but the distance between the ossified area and cancer cells was greater in the usual pattern compared to the unusual pattern. These findings suggest the possibility of an active ossification due to tumor progression.
