Fecal Galectin-3: A New Promising Biomarker for Severity and Progression of Colorectal Carcinoma

Background and Objectives. The aim of the study was to determine systemic and fecal values of galectin-3 and pro-and anti-inflammatory cytokines in patients with CRC and the relationship with clinicopathological aspects. Methods. Concentrations of galectin-3, TNF-alpha, TGF-beta, IL-10, and IL-1 beta were analyzed in samples of blood and stool of 60 patients with CRC. Results. Systemic concentration of TNF-alpha was significantly lower in patients with severe diseases (advanced TNM stage, nuclear grade, and poor histological differentiation) as in patients with more progressive CRC (lymph and blood vessel invasion, presence of metastasis). Fecal values of anti-inflammatory cytokines TGF-beta and IL-10 were increased in patients with severe stadium of CRC. Fecal concentration of Gal-3 was enhanced in CRC patients with higher nuclear grade, poor tumor tissue differentiation, advanced TNM stage, and metastatic disease. Gal-3/TNF-alpha ratio in sera and feces had a higher trend in patients with severe and advanced diseases. Positive correlation between fecal Gal-3 and disease severity, tumor progression, and biomarkers AFP and CEA, respectively, was also observed. Conclusions. Predomination of Gal-3 in patients with advanced diseases may implicate on its role in limiting ongoing proinflammatory processes. The fecal values of Gal-3 can be used as a valuable marker for CRC severity and progression.

The influence of vitamin D supplementation on the expression of mediators of inflammation in knee osteoarthritis

Summary: This trial suggests that vitamin D3 has potential therapeutic and immunomodulatory effects in patients with knee osteoarthritis. Oral supplementation of vitamin D3 can significantly reduce pain and improve the quality of life in knee osteoarthritis patients. Vitamin D3 supplementation also increases systemic values of IL-33.

Galectin-3 as an important prognostic marker for COVID-19 severity

Summary: Gal-3 is associated with COVID-19 severity and immune response. Patients in critical stage have higher levels of Gal-3, cytokines, and CCR5 expression, indicating more severe inflammation. Gal-3 also correlates with clinical and radiographic findings, as well as peripheral blood immune cell composition.

The Role of IL-17 in the Pathogenesis of Oral Squamous Cell Carcinoma

Summary: Elucidating the inflammatory mechanisms underlying formation and progression of oral squamous cell carcinoma (OSCC) is crucial for discovering new targeted therapeutics. The proinflammatory cytokine IL-17 has proven roles in tumor formation, growth, and metastasis. The presence of IL-17 is demonstrated in both in vitro and in vivo models, and in OSCC patients, is mostly accompanied by enhanced proliferation and invasiveness of cancer cells. Here we review the known facts regarding the role of IL-17 in OSCC pathogenesis, namely the IL-17 mediated production of proinflammatory mediators that mobilize and activate myeloid cells with suppressive and proangiogenic activities and proliferative signals that directly induce proliferation of cancer cells and stem cells. The possibility of a potential IL-17 blockade in OSCC therapy is also discussed.

The expression of mammaglobin in tumoral and peritumoral tissues and its circulating levels in the blood will be thoroughly examined, providing insights into its possible function in cancer progression and metastasis. Furthermore, the potential application of mammaglobin as a non-invasive diagnostic tool and a target for personalized treatment strategies will be discussed.

Summary: Four new ligands and their corresponding palladium(II) complexes were synthesized and characterized. The interaction between the complexes and human serum albumin (HSA) was studied using fluorescence spectroscopy. The most active and selective compound, ligand L4, showed potential for future in vivo testing and induced cell death mainly by apoptosis.

Myeloid-derived suppressor-like cells - a potential biomarker for prognosis of colorectal cancer?

Summary: The aim of this study was to examine the potential prognostic role of peripheral blood MDSC counts in colorectal cancer (CRC) patients. The results showed that percentages and absolute numbers of CD16(low) granulocytes, as well as the absolute numbers of M-MDSC like cells, the CD16(high)/CD16(low) granulocytes ratio, monocyte/M-MDSC like cells ratio, and NLR ratio, may be reliable indicators of overall survival in CRC patients.

Myeloid-derived suppressor-like cells - does their frequency change in patients with different stages of CRC?

Summary: This study analyzed the frequency and significance of MDSC-like cells in different stages of colorectal cancer patients. The results showed a significant increase in the number and proportion of PMN-MDSC-like cells in all patients with CRC compared to the healthy control group. The absolute and relative numbers of PMN-MDSC-like cells significantly correlated with the stages of CRC disease and the number of organs affected by metastases.

Summary: The aim of this study was to investigate the relationship between biochemical, haemostatic, and immune parameters of atherosclerosis and ulcerative colitis patients and platelet aggregation. The results showed higher levels of inflammatory markers and platelet aggregation in ulcerative colitis patients, as well as lower levels of total cholesterol and LDL. Inflammatory markers and platelet aggregation could be potential discrimination markers for ulcerative colitis patients with a higher risk of atherosclerosis.

Increased Pro Th1 And Th17 Transcriptional Activity In Patients With Severe COVID-19

Summary: This study aimed to determine the transcriptional activity and functional phenotype of T cells and the relationship between pro-and anti-inflammatory cytokines and clinical parameters of COVID-19 severity. The results showed a significant elevation of IL-12, IFN-gamma, IL-17, and IL-23 in stage IV of the disease, with a strong intercorrelation between these cytokines. CD4+ T cells displayed increased transcriptional activity and an increased percentage of IFN-gamma and IL-17 producing T cells.
