Summary: Pancreatic cancer is a highly aggressive malignant tumor with poor prognosis and high mortality rates. Despite advancements in diagnosis and treatment, the current therapeutic strategies for pancreatic cancer have limited efficacy, necessitating urgent exploration of alternative options. Mesenchymal stromal cells (MSCs) have gained attention as a potential therapy for pancreatic cancer due to their tumor-homing properties, but the specific antitumor effect of MSCs remains controversial. Therefore, this study aims to focus on the potential of the MSC-based approach for anti-cancer treatment and summarize the current challenges in the clinical application of MSCs for pancreatic cancer.

Summary: Prostate cancer is a common cancer in men and a major cause of cancer-related deaths. In this study, we investigated the cell-type-specific signaling networks initiated by intercellular communication between prostate epithelial cells and patient-derived cancer-associated fibroblasts (CAFs) when grown together. Significant differences in proteomic profiles were identified, and the upregulation of transglutaminase-2 (TGM2) was found to promote migration and proliferation of epithelial cells under co-culture conditions. These findings provide insights into intercellular communication in prostate cancer that may improve patient management.

Summary: This study analyzed hyaluronan (HA) deposition in ovarian cancer tissues and found that pericellular tumor-cell-associated HA deposition was significantly associated with malignancy and represented an independent unfavorable prognostic marker for overall survival. Additionally, high mRNA levels of CD44, HAS2, and HAS3 were significantly associated with pericellular HA deposition in cancer cells. In contrast, stromal hyaluronan deposition had no impact on ovarian cancer prognosis.

Summary: Extracellular vesicles (EVs) are microscopic particles released naturally in biofluids and can serve as promising liquid biomarkers for human diseases. Flow cytometry is a popular method for detecting, characterizing, and determining the concentration of EVs. However, the limited awareness and utilization of standardization and calibration methods in the scientific community pose challenges for data reproducibility and inter-laboratory comparison. This study presents guidelines and best practices for flow cytometry in EV research, providing a systematic approach to evaluate performance and improve detection sensitivity for EVs.

Summary: Narirutin, a compound found in lemon, orange, passion fruit, bergamot, and grapefruit, has potential as an anti-allergic, cardioprotective, neuroprotective, and hepatoprotective agent. It can suppress the growth of prostate cancer cells, but its chemopreventive potential against hormone-refractory prostate cancer cells (PC-3) and its mode of action are not yet known. In this study, the chemopreventive possibility of narirutin was explored in PC-3 cells, and it was found that narirutin reduced cell viability, induced G0/G1 phase arrest, and increased the generation of reactive oxygen species (ROS). Narirutin also inhibited the activity of hyaluronidase and showed a strong binding interaction with the enzyme. Molecular dynamics simulation analysis supported the idea that narirutin binding enhanced the compactness and stability of the enzyme. Additionally, narirutin was predicted to be non-toxic and non-CYPs inhibitor. These results suggest that narirutin may be a potential chemopreventive agent for hormone-resistant prostate cancer cells and provide data supporting the use of diet-based nutraceutical agents to prevent prostate cancer.

Summary: VEGF-C secreted by EMT breast cancer cells can promote proliferation, migration, and invasion of epithelial breast cancer cells through non-canonical activation of GLI signaling. Inhibiting VEGF-C in EMT cells or knocking down NRP2 in neighboring epithelial cancer cells disrupts the aggressive phenotypes induced by EMT cells, suggesting a novel therapeutic target for metastasis in breast cancer.

Summary: Prostate cancer (PCa) is the second most lethal cancer for men in western countries, with androgen receptor (AR) playing a central role in its initiation and progression. The interactions between stromal cells and PCa epithelial cells, mediated by AR signaling, impact the growth and invasiveness of PCa cells. Androgen deprivation therapy (ADT) treatment can enhance the reciprocal interaction between stromal and epithelial cells, promoting cell invasion of PCa cells.

Summary: Prostate cancer is the most commonly diagnosed male malignancy in the Western world. Metabolic reprogramming and epigenetic remodeling play important roles in the progression and therapy resistance of prostate cancer, and there is crosstalk between metabolism and epigenetics. A high-fat diet has been associated with prostate cancer progression. Understanding these alterations and mechanisms can help identify prognostic and predictive biomarkers, as well as targetable vulnerabilities.
