Summary: CMTM4 overexpression in HNSCC is correlated with advanced tumor progression and poor prognosis, and knockdown of CMTM4 can inhibit EMT process, tumor cell invasion, CSC markers, and PD-L1 expression. Positive correlation was found between CMTM4 expression and CD8(+) and PD-1(+) cell density in the stroma.

Summary: The expression of PD-L1 by tumor cells is primarily associated with its immunosuppressive effect, but there is evidence of additional cell-intrinsic functions. PD-L1's function and expression vary depending on factors such as cell cycle, differentiation status, and microenvironment. Understanding these variations may provide insights into differential response to PD-1/PD-L1 antibody therapy and optimize antibody-based immunotherapy.

Summary: This study investigated the expression and role of SEC11A in head and neck squamous cell carcinoma (HNSCC). The results showed that SEC11A expression was significantly elevated in HNSCC tissues compared to adjacent normal tissues. Functional assays demonstrated that SEC11A knockdown reduced cell proliferation, migration, and invasion in vitro, and inhibited tumor growth in vivo. These findings suggest that SEC11A plays a crucial role in HNSCC proliferation and progression, and may serve as a potential therapeutic target.

Summary: This study reveals that the levels of m6A modification and METTL3 expression are significantly increased in head and neck squamous cell carcinoma (HNSCC). METTL3 overexpression promotes HNSCC cell proliferation, migration, invasion, and angiogenesis, and is associated with poor prognosis in HNSCC patients.

Summary: HNSCC, the 9th most common malignant tumor worldwide, can be divided into HPV-related and HPV-unrelated subtypes based on etiology. HPV-positive HNSCC is more sensitive to treatment with better survival rates, requiring individualized therapy. EGFR amplification and PI3K/AKT/mTOR pathway aberrant activation are common in HPV-positive HNSCC.

Summary: This study investigated the cancer-promoting effect of DDIT4 in Head and Neck Squamous Cell Carcinoma (HNSCC) and its mechanisms. DDIT4 overexpression was found to be associated with worse tumor characteristics, while DDIT4 knockdown inhibited the proliferation, invasion, and migration of HNSCC cells, as well as the expression of HIF-1a, VEGF, and vimentin. Additionally, DDIT4 expression was negatively correlated with immune infiltrations and played a crucial role in the differentiation of CAFs and T cells.

Summary: This study demonstrates that the combination therapy of CDK4/6 and panEGFR inhibitors can significantly reduce tumor growth, induce cellular senescence, and regulate metabolic pathways in HNSCC, suggesting that dual targeting may be a critical therapeutic strategy in blocking tumor progression.

Summary: Head and Neck Squamous Cell Carcinoma (HNSCC) is the sixth most common cancer worldwide, accounting for approximately 6% of all cases and around 2% of all cancer deaths. Despite advancements in treatment, survival rates remain low. Epigenetic modifications have associations with biological processes and cancer, suggesting a need for a more systematic approach to identify potential targets and effective therapies.

Summary: Head and neck squamous cell carcinoma (HNSCC) originates from different parts of the head and neck, and the causes and molecular basis can vary. The tumor microenvironment (TME) consisting of various cells and molecules is crucial for the survival and spread of cancer cells. Understanding the interaction between tumor cells and the TME is essential for developing effective anti-cancer therapies.

Summary: This study validated a protocol using microfluidic enrichment and flow cytometry to characterize EMT CTCs in HNSCC. CTCs were associated with advanced disease, but not with progression-free or overall survival. Furthermore, there was no correlation between tumor and CTC EMT profiles, suggesting that CTCs may undergo an adaptive EMT in response to stimuli within the circulation.

Summary: This study demonstrates that SQLE plays a vital role in the progression of head and neck squamous cell carcinoma (HNSCC). High expression of SQLE is associated with aggressive tumor features and poor prognosis. Additionally, overexpression of SQLE enhances the proliferation, migration, and invasion abilities of HNSCC cells. The PI3K/Akt signaling pathway is implicated in the SQLE-mediated progression of HNSCC.

Summary: A new scoring system based on EMT and hypoxia, named CIEH, has been developed for predicting prognosis and guiding treatment planning in HNSCC patients. The CIEH model, containing 14 characteristic factors, shows potential as biomarkers for HNSCC and is correlated with tumor immune infiltration.

Summary: In this study, researchers found that Lon Peptidase 2, Peroxisomal (LONP2) was upregulated in head and neck squamous cell carcinoma (HNSCC) and its overexpression promoted cell growth and migration. Silencing of LONP2 inhibited tumor growth and improved apoptosis rate in HNSCC cells. Knockdown of LONP2 also downregulated the expression of plastin 3 (PLS3) and inhibited cell proliferation induced by LONP2 overexpression, suggesting that LONP2 exerts its oncogenic effect in HNSCC cells by positively regulating PLS3 expression.
