Summary: High-grade tumor budding is associated with a more aggressive tumor phenotype, and when combined with MMP/TIMP expression by stromal cells at the invasive front of the tumor, it helps identify patients with poor prognosis. The optimal cut-off for predicting both relapse-free and overall survival was found to be a tumor budding count >= 4. The study also showed that high-grade tumor budding was linked to MMP/TIMP expression by cancer-associated fibroblasts.

Summary: This study aimed to explore the prognostic factors of pembrolizumab in patients with advanced biliary tract cancer (BTC) and provide treatment guidelines for patients who are refractory to gemcitabine-based chemotherapy. The results showed that high albumin levels and lymphocyte-to-monocyte ratio (LMR), as well as low alkaline phosphatase (ALP) levels and total bilirubin (TB), were significantly associated with better overall survival in patients treated with pembrolizumab.

Summary: The study evaluated the diagnostic efficacy and prognostic value of vimentin-positive circulating tumor cells (V-CTCs) in patients with biliary tract cancer (BTC). Results showed significant differences in V-CTC count and ratio between BTC and benign biliary disease (BBD), and combining these parameters improved accuracy. V-CTC count > 50/mL blood was a significant factor affecting survival, suggesting its potential as a biomarker for early diagnosis and prognosis prediction in BTC patients.

The combination parameter including HALP was identified as a superior predictor and an independent factor for overall survival prediction. The developed nomogram based on HALP, tumor-node-metastasis staging, and operative outcome showed promising results in predicting overall survival.

A high density of FoxP3(-) CD4(+) helper T cells in the tumor margin may be associated with better clinical outcomes.

CTGF can activate SPARC expression in the tumor stroma, promoting tumor progression. Summary: The invasive tumor front is crucial for malignant cell progression and metastasis, with interactions between tumor cells and host cells determining the fate of the tumor. While similarities were found between uterine adenocarcinomas and leiomyosarcomas at the invasive front, differences were also observed in immune cell populations, gene expression, and methylation signatures. Further research on the molecular and architectural characterization of invasive front in uterine malignancies could provide additional prognostic information.
