Ibuprofen (IBF), 2-(4-isobutylphenyl) propionic acid, is a surface-active, common nonsteroidal anti-inflammatory drug (NSAID), and it possesses a high critical micelle concentration (cmc) compared to that of conventional surfactants. The interactions of this common NSAID with an anionic surfactant, sodium octyl sulfosuccinate, were studied by tensiometric, fluorimetric, and calorimetric measurements to investigate this system as a possible model drug delivery system for an NSAID like IBF, particularly in a high-dose regime for IBF. The interactions between the drug and the surfactant were modeled using a regular solution theory approach in the presence and absence of a model electrolyte (sodium iodide) and a novel nonaromatic, gemini hydrotrope, tetramethylene-1,4-bis(N,N-dimethyl-N-butylammonium)bromide (4-4-4). Both the simple and the hydrotropic electrolyte were shown to have an effect on the solution properties (aggregation parameters, interfacial properties, and thermodynamics of aggregate formation) of the drug surfactant mixtures and on the interaction between the drug and the surfactant. Surface charges of all self-assembled potential measurements, whereas density functional theory calculations showed the interaction energy comparison among all of the binary and ternary combinations. All of these results were interpreted in terms of how altering the subtle balance of hydrophobic and electrostatic forces can significantly improve the ability of these self-assembled systems to transport drug molecules.

The Interaction of Glymes with Surfactant Micelles

Nicholas P. Petropolis, Harrison M. Petropolis, Nicole E. MacNeil, Taylor M. Doucet, D. Gerrard Marangoni

Summary: Favorable interactions were found between sodium dodecyl sulfate micelles and glyme cosurfactants, while little favorable interactions were observed between dodecyltrimethylammonium bromide micelles and glymes. These results were determined through conductance experiments and C-13 nuclear magnetic resonance chemical shifts analysis.

Amphiphilic star block copolymer micelles in saline as effective vehicle for quercetin solubilization

Sanjay Tiwari, Kulbir Singh, D. Gerrard Marangoni, Pratap Bahadur

Summary: This study investigates the solubilization of quercetin, a hydrophobic flavonoid, in a saline solution of an amphiphilic copolymer. The results show that the copolymer solution has superior solubilization compared to linear counterparts and provides a protective environment for quercetin against oxidation.

Summary: Gemini surfactants are composed of two hydrocarbon tails with corresponding polar headgroups, linked via a covalent spacer. The improved synthetic approach of these symmetrical gemini surfactants has led to their characterization of crystalline packing for the first time ever using X-ray crystallographic analysis.

Thermodynamics of glyme-sodium dodecyl sulfate (SDS) mixed micelle formation via isoperibol solution calorimetry: Temperature, and concentration effects

Shannon MacLennan, D. Gerrard Marangoni

Summary: This study determined the enthalpies of mixed micelle formation (ΔmicH) for various SDS/glyme systems using isoperibol solution calorimetry, with the concentration of glycol ether and temperature as variables. The glycol ethers investigated were DEGDME, TEGDME, TetraEGDME, and DEGDEE. The CMC values of the SDS/glyme mixed micelles were determined directly based on the data from the calorimeter. The entropies and Gibbs energies of mixed micellization (ΔmicS and ΔmicG) were calculated using the charged pseudo-phase separation model. The results showed that the CMC values and mixed micelle formation enthalpies were dependent on the structure and concentration of the glyme in the mixed solvents, indicating the importance of alkyl chain interactions, hydrophobic effects, and electrostatic contributions in micelle thermodynamics.

Lactate shuttling as an allostatic means of thermoregulation in the brain

Daniel A. Kane, Alexander C. Y. Foo, Erin B. Noftall, Karen Brebner, D. Gerrard Marangoni

Summary: Lactate, as the end product of glycolysis, plays various physiological roles by traveling within and between cells. However, its application to physical bioenergetics has been underexplored. This study suggests that lactate shuttling, as an allostatic thermoregulatory strategy, mitigates the consequences of elevated metabolic heat by exchanging extracellular lactate between tissues. The results of experiments on rat cortical brain samples support this hypothesis by showing lower heat and respiratory oxygen consumption rates during lactate-linked respiration compared to pyruvate-linked respiration.

The partitioning of primary alcohols into the aggregates of gemini amphiphiles determined from diffusion NMR experiments

Brooke A. Morris, Ahmed A. Elgendy, Nicole E. MacNeil, Olivia M. Singer, Jacob G. Hoare, Robert D. Singer, D. Gerrard Marangoni

Summary: The partition constants of primary alcohols in symmetric gemini surfactant aggregates were calculated from diffusion coefficients obtained from NMR-diffusion experiments. The Gibbs energy of transfer for the alcohols from water to the gemini aggregate phase decreased linearly with an increase in the alcohol carbon length. The partition coefficients of the alcohols in the symmetric gemini aggregates showed little dependence on the spacer length of the gemini amphiphiles and were consistent as the length of the main surfactant chain increased.
