Summary: Accurate reconstruction of metabolic pathways is crucial for understanding metabolomics changes and biological processes in diseases. A tracer-based metabolomics strategy using stable isotope labeled precursors can trace pathways by measuring the transformation of metabolites. By quantifying labeled metabolite substructures, a new method achieves simultaneous isotopic labeling information at the intact metabolite and moiety level. This method was applied to trace the fate of labeled atoms in human-induced pluripotent stem cell-derived neurons, revealing the pathway reconstruction of de novo glutathione synthesis and its alteration under oxidative stress and neurodegeneration.

Summary: A set of QA/QC criteria for nontargeted measurement of pesticide exposure markers in human urine has been proposed and applied in the HBM4EU project. The criteria include monitoring signal intensities, mass accuracy, and retention times of selected markers. The results show high repeatability and accuracy of the data. This is the first time that harmonized QA/QC criteria have been used to assess the quality of nontargeted analysis of human samples across multiple laboratories.

Summary: In this study, a method called 2D-LC/TQMS was developed for simultaneous analysis of hydrophilic and hydrophobic metabolites using the combination of HILIC and RPLC. The method can detect 417 metabolites involved in various metabolic pathways with good reproducibility, linearity, and wide linear range. It was tested using different biological samples and applied to a bladder cancer case/control study, identifying 30 differential metabolites related to carbohydrate and amino acid metabolism.

The use of C-13-labelled P. pastoris extract as internal standards resulted in significant reduction in lipid variations compared to other normalization methods. Summary: The study identified SNORA73 as a protective factor against lipotoxicity and oxidative stress, showing that its deficiency leads to metabolic reprogramming and protection against steatohepatitis in mice. Specifically, disruption of SNORA73 causes resistance to lipid-induced cell death and oxidative stress in cultured cells, with knockdown of SNORA73 in vivo protecting against hepatic steatosis and lipid-induced oxidative stress and inflammation. This demonstrates a role for SNORA73 in regulating metabolism and lipotoxicity.

It was found that challenges in the cytosol affected NADPH fluxes in the cytosol, while challenges in the mitochondria affected NADPH fluxes in the mitochondria. This work highlights the value of using proline labeling as a reporter system for studying compartmentalized metabolism and reveals independent regulation of NADPH homeostasis in the cytosol and mitochondria, without evidence for NADPH shuttle activity.

The integration of mass spectrometry imaging (MSI), stable isotope labeling, and isotopologue spectral analysis enables the mapping of metabolite distributions and metabolic fluxes in the brains of mice with GL261 glioma. This approach reveals alterations in anabolic pathways, specifically an increase in de novo fatty acid synthesis and elongation fluxes in the glioma compared to surrounding healthy tissue.

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

Summary: This study employed network pharmacology-based analysis and reverse molecular docking to investigate the molecular targets and pathways associated with gynecological disorders and the potential therapeutic effects of fenugreek constituents. The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
