Two novel periodic mesoporous organosilica materials were synthesized with a neutral phenylenebridged ligand, 1,4-bis(trimethoxysilylethyl)benzene, one of them using tetraethyl orthosilicate as additional silica source (PMO-TMSEB-1 and PMO-TMSEB-2). A third material was also synthesized with 1,4-bis(triethoxysilyl)benzene ligand (PMO-TESB-1) which use has scarcely been reported. The three materials were evaluated as solid-phase extraction (SPE) sorbents for the off-line extraction of a mixture of seven drugs of different nature (duloxetine, terbutaline, econazole, propranolol, verapamil, metoprolol, and betaxolol) from water samples. Subsequent simultaneous enantiomeric analysis by CE, using sulfated-beta-cyclodextrin (2% w/v) dissolved in a 25 mM phosphate buffer (pH 3.0) and a voltage of -20 kV (negative polarity) was carried out. Enantiomeric resolutions ranging from 2.4 to 8.5 were obtained in an analysis time of 16 min. After optimization of SPE parameters, it was shown that using just 100 mg of PMO-TESB-1 as sorbent, a preconcentration factor of 400 with 200 mL solution was achieved, allowing recoveries between 80.5 and 103.1% (except for terbutaline), with good repeatability (% RSD = 2-8 %, n = 5). Analytical characteristics of the method were evaluated in terms of precision, linearity and accuracy with method quantitation limits between 5.6 and 21.9 mu g/L. The developed method was applied to the analysis of spiked wastewater samples collected in different treatment plants, with recoveries between 73.9 and 102.9% except for econazole with recovery values ranging between 58.5 and 72.4%. Sonia Morante-Zarcero, Alba Endrino, Natalia Casado, Damian Perez-Quintanilla, Isabel Sierra

Summary: In this study, mesostructured silicas with wormhole-like pore arrangement were synthesized and evaluated as carriers for bioactive flavonoids. The results showed that these silicas can effectively load and release quercetin and naringin, with potential applications in food and biomedical fields. Furthermore, the antioxidant activity of the encapsulates was determined, confirming the suitability of wormhole-like mesostructured silicas as carriers for enhancing stability and bioavailability of flavonoids.

Nanomaterials-modified electrochemical sensors for sensitive determination of alkaloids: Recent trends in the application to biological, pharmaceutical and agri-food samples

Judith Ganan, Gonzalo Martinez-Garcia, Sonia Morante-Zarcero, Damian Perez-Quintanilla, Isabel Sierra

Summary: Alkaloids are widely found secondary metabolites in plants, many of which have significant biological activity and pharmacological applications. However, the potential adverse effects of alkaloids present in food, such as acute hepatotoxicity, genotoxicity, and neurological damage, raise concerns among consumers. Therefore, developing sensitive, selective, and convenient sensors for alkaloids screening is crucial. Electrochemical detection platforms, particularly those based on nanomaterials, have shown great potential as a cost-effective, efficient, eco-friendly, and easy-to-operate analytical approach for alkaloids detection in various applications.

Insight into the Impact of Food Processing and Culinary Preparations on the Stability and Content of Plant Alkaloids Considered as Natural Food Contaminants

Natalia Casado, Gema Casado-Hidalgo, Lorena Gonzalez-Gomez, Sonia Morante-Zarcero, Isabel Sierra

Summary: Pyrrolizidine alkaloids, tropane alkaloids, and opium alkaloids are natural plant toxins that are of great concern in food safety due to their presence in various foods and feeds. Despite recent legislation, the concentration levels of these toxins often exceed the maximum limits established by authorities. Moreover, the regulations primarily focus on raw materials and do not consider processed products. Therefore, it is crucial to assess the health risk associated with these alkaloids through diet. This review aims to provide insight into these alkaloids and their influence on content and stability during food processing and culinary preparation, highlighting the need for further research to understand their transformation and determine the exact risk exposure to the population.

Evaluation of the thermal stability and transfer rate of pyrrolizidine alkaloids during the brewing of herbal infusions contaminated with Echium vulgare and Senecio vulgaris weeds

Begona Fernandez-Pintor, Natalia Casado, Sonia Morante-Zarcero, Isabel Sierra

Summary: Pyrrolizidine alkaloids (PAs) have attracted attention in food safety due to their potential health risk. Tea and herbal tea, as they can be contaminated with PAs-producing weeds, have been particularly highlighted. This study simulated the cross-contamination of chamomile tea samples with weeds to evaluate the effect of brewing on PA content. The results showed that PAs are stable compounds and the initial PA concentration strongly affects their thermal stability. Not all PAs are effectively transferred from dry material to the infusion during brewing, and the extraction efficiency may be influenced by the type of weed. The study emphasizes the importance of analyzing infusion samples to correctly assess the risk exposure of consumers to PAs.

Influence of fermentation and storage on the content of opium alkaloids in poppy seed yoghurt

Gema Casado-Hidalgo, Sonia Morante-Zarcero, Damian Perez-Quintanilla, Isabel Sierra

Summary: There is a current trend to add seeds such as poppy seeds in yoghurts to enhance their nutrition. However, these seeds may be contaminated with opium alkaloids (OAs) which can be harmful to health. In this study, an analytical methodology was developed to detect OAs in commercial yoghurts and showed their presence but below the limit of quantification. The study also found that lactic fermentation and storage can lead to degradation of OAs in yoghurts.

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.
