The immunomodulatory effects of adipose-derived mesenchymal stem cells and mesenchymal stem cells-conditioned medium in chronic colitis

Inflammatory bowel disease (IBD) as a chronic recurrent disorder is characterized by mucosal immune response dysregulation, which is more prevalent in the youth. Adipose-derived mesenchymal stem cells (ADMSCs) are the multipotent cells that can be effective in immune response regulation via cell-cell interaction and their secretions. In this study, the effects of ADMSCs and mesenchymal stem cell-conditioned medium (MSC-CM) were evaluated on dextran sulfate sodium (DSS)-induced colitis in mice. Chronic colitis was induced in female C57BL/6 mice using 2% DSS in drinking water for three cycles; there were 4 days of DSS-water administration that was followed by 7 days of DSS-free water, in a cycle. ADMSCs, 10(6) cells per mouse, were injected intraperitoneally (IP), whereas the MSC-CM injection was also performed six times from the last day of DSS in Cycle 1. Clinical symptoms were recorded daily. The colon pathological changes, cytokine levels, and regulatory T (Treg) cell percentages were then analyzed. After receiving ADMSCs and MSC-CM in colitis mice, the clinical symptoms and disease activity index were improved and the survival rate was increased. The histopathological examination also showed tissue healing in comparison with the nontreated group. In addition, the increased level of transforming growth factor beta, increased percentage of Treg cells, increased level of interleukin (IL)-10, and decreased level of IL-17 were observed after the treatment. This study showed the regulatory effects of ADMSCs and MSC-CM on inflammatory responses. Therefore, the use of ADMSCs and MSC-CM can be introduced as a new and effective therapeutic approach for patients with colitis.

The effect of poly I:C or LPS priming on the therapeutic efficacy of mesenchymal stem cells in an adjuvant-induced arthritis rat model

Summary: In this study, the effect of priming adipose-derived mesenchymal stem cells (ADMSCs) with lipopolysaccharide (LPS) or polyinosinic:polycytidylic acid (poly I:C) on their immunomodulatory function was investigated. The results showed that priming with poly I:C significantly reduced splenocyte proliferation mediated by ADMSCs and alleviated the clinical and histopathological severity of arthritis. Furthermore, poly I:C-primed ADMSCs also decreased the secretion of inflammatory cytokines and increased the levels of anti-inflammatory cytokines. In contrast, priming with LPS reduced the therapeutic effect of ADMSCs in the arthritis model.

Summary: Encapsulating chemotherapeutic molecules in nanocarriers is a justifiable solution in decreasing the risk of their side effects and boosting the efficiency of treatment. This study developed a new method for treating colorectal cancer using AS1411 aptamer-functionalized exosomes to deliver chemotherapy drugs. The results demonstrated that this method effectively suppressed tumor growth and has potential clinical applications.

Summary: The molecular mechanisms of senescence in dental pulp stem cells (hDPSCs) stimulated with lipopolysaccharide (LPS) from oral pathogenic bacteria were investigated in this study. The mRNA and protein expression of four senescence-related genes were significantly upregulated in a time-dependent manner following stimulation with different LPSs. Furthermore, the highest expression of certain genes was observed at different time points depending on the type of LPS.

RNAa-mediated epigenetic attenuation of the cell senescence via locus specific induction of endogenous SIRT1

Summary: This study demonstrates that upregulation of SIRT1 can be achieved by knocking down the natural antisense transcript SIRT1-NAT, leading to improved therapeutic potentials of human MSCs. Knockdown of SIRT1-NAT enhances the proliferation and differentiation abilities of MSCs, reduces senescence-associated activity, and reverses senescence-associated molecular changes.

Mechanisms behind therapeutic potentials of mesenchymal stem cell mitochondria transfer/delivery

Summary: Mesenchymal stromal/stem cells (MSCs) exert their therapeutic effects through various mechanisms, including differentiation, production of growth factors, immunomodulatory factors, and extracellular vesicles. Recent studies have also highlighted the therapeutic potential of MSCs through mitochondrial transfer, with different methods of transfer being investigated. Pathological conditions can cause mitochondrial damage in cells, leading to decreased ATP production and cell death. Restoring mitochondrial content, function, and homeostasis through MSCs mitochondrial transfer can have significant effects on cell attributes and behavior, crucial for therapeutic purposes.

Effects of Lipopolysaccharide from Porphyromonas gingivalis and Escherichia coli on Gene Expression Levels of Toll-like Receptors and Inflammatory Cytokines in Human Dental Pulp Stem Cells

Summary: This study evaluated the effects of P. gingivalis and E. coli lipopolysaccharide (LPS) on the gene expression of TLRs and inflammatory cytokines in human dental pulp stem cells. The results showed that both types of LPS stimulate inflammation through increased TLR4 expression, with P. gingivalis LPS showing stronger stimulation within the first 24 hours. However, with prolonged exposure time, both LPS appeared to have anti-inflammatory properties.
