Rab GTPases are key regulators of eukaryotic membrane traffic, and their functions and activities are limited to particular intracellular transport steps and their membrane localization is by and large restricted. Some Rabs do participate in more than one transport steps, but broadly speaking, there is a clear demarcation between exocytic and endocytic Rabs. One Rab protein, Rab10, however, appears to be anomalous in this regard and has a diverse array of functions and subcellular localizations. Rab10 has been implicated in a myriad of activities ranging from polarized exocytosis and endosomal sorting in polarized cells, insulin-dependent Glut4 transport in adipocytes, axonal growth in neurons, and endo-phagocytic processes in macrophages. It's reported subcellular localizations include the endoplasmic reticulum (ER), Golgi/TGN, the endosomes/phagosomes and the primary cilia. In this review, we summarize and discuss the multitude of known roles of Rab10 in cellular membrane transport and the molecular players and mechanisms associated with these roles.

A research misconduct severity matrix that could serve to harmonize adjudication of findings

Nicole Shu Ling Yeo-Teh, Bor Luen Tang

Summary: Investigations into research misconduct are typically guided by ethics and integrity policies, but the lack of specific severity criteria can result in varying conclusions and impact the standardization of sanctions. Proposed research misconduct severity matrix aims to address this issue by considering multiple ethical elements with different weights to determine the severity of misconduct more objectively.

Defects in early secretory pathway transport machinery components and neurodevelopmental disorders

Bor Luen Tang

Summary: The early secretory pathway is crucial for a constant supply of secretory and plasma membrane materials in mammalian cells. Neurons have a high demand for membrane dynamics and protein/lipid trafficking, and mutations in genes encoding early secretory pathway components can lead to neurodevelopmental or neurological disorders.

Wilfully submitting to and publishing in predatory journals-a covert form of research misconduct

Nicole Shu Ling Yeo-Teh, Bor Luen Tang

Summary: Predatory journals masquerade as genuine academic publications without rigorous peer review, focusing on financial profit over scientific dissemination. Such practices can flood scientific literature with unsound research, potentially misleading policy makers and damaging the reputation of science. Wilfully submitting to predatory journals may be considered an act of avoidance of peer review and scientific misconduct, which could be effectively discouraged through institutional and funding rules.

Cholesterol synthesis inhibition or depletion in axon regeneration

Bor Luen Tang

Summary: Cholesterol plays important structural and regulatory roles in animal cells, and is crucial for post-injury axon regrowth. Depletion and inhibition of cholesterol synthesis can enhance axon regeneration, possibly by interfering with axon growth inhibitory signals and enhancing neuronal survival signaling processes.

WNK3 Maintains the GABAergic Inhibitory Tone, Synaptic Excitation and Neuronal Excitability via Regulation of KCC2 Cotransporter in Mature Neurons

Wee Meng Lim, Eunice W. M. Chin, Bor Luen Tang, Tingting Chen, Eyleen L. K. Goh

Summary: WNK3 plays a crucial role in maintaining the GABAergic inhibitory tone in mature neurons, influencing neuronal development and excitability. Its knockdown results in hyperpolarization of E-GABA in mature neurons, without affecting immature neurons, suggesting a role in the developmental switch of GABA function. This study highlights the importance of WNK3 in regulating GABA signaling and neuronal physiology.

Research data mismanagement - from questionable research practice to research misconduct

Nicole Shu Ling Yeo-Teh, Bor Luen Tang

Summary: Good record keeping practice and research data management are essential for responsible research conduct and reproducibility of findings. Inadequate research data management often accompanies research misconduct. Research data mismanagement (RDMM) can be considered a questionable research practice and in severe cases, an act of research misconduct. This article analyzes the contexts and circumstances under which RDMM can be viewed as misrepresentation or fabrication of research data, and discusses how it can be adjudicated as research misconduct based on intent and consequences.
