Genome-wide identification of urinary cell-free microRNAs for non-invasive detection of bladder cancer

Urinary microRNAs (miRNAs) are emerging as clinically useful tool for early and non-invasive detection of various types of cancer including bladder cancer (BCA). In this study, 205 patients with BCA and 99 healthy controls were prospectively enrolled. Expression profiles of urinary miRNAs were obtained using Affymetrix miRNA microarrays (2578 miRNAs) and candidate miRNAs further validated in independent cohorts using qRT-PCR. Whole-genome profiling identified 76 miRNAs with significantly different concentrations in urine of BCA compared to controls (P < 0.01). In the training and independent validation phase of the study, miR-31-5p, miR-93-5p and miR-191-5p were confirmed to have significantly higher levels in urine of patients with BCA in comparison with controls (P < 0.01). We further established 2-miRNA-based urinary DxScore (miR-93-5p, miR-31-5p) enabling sensitive BCA detection with AUC being 0.84 and 0.81 in the training and validation phase, respectively. Moreover, DxScore significantly differed in the various histopathological subgroups of BCA and decreased post-operatively. In conclusion, we identified and independently validated cell-free urinary miRNAs as promising biomarkers enabling non-invasive detection of BCA.

Oncological outcomes of surgery for isolated retroperitoneal recurrence in renal cancer patients after radical nephrectomy

Summary: Aggressive surgical therapy is feasible for isolated retroperitoneal recurrence in renal cancer patients after radical nephrectomy, with more than half of the patients achieving long-term survival. Prognostic factors of poor outcome include an interval from nephrectomy to recurrence of less than 12 months, tumor grade 3-4, and female sex. Metastasis-directed therapy may prolong the time to initiation of systemic treatment in cases of disease progression.

Use of Hypolipidemic Drugs and the Risk of Second Primary Malignancy in Colorectal Cancer Patients

Summary: The association between hypolipidemic drugs and the occurrence of a second primary malignancy in colorectal cancer survivors is not clear. Subgroup analysis shows a lower incidence of secondary neoplasia in statin users, but colorectal cancer patients using hypolipidemic drugs are more likely to develop gastric and bladder cancer. Patients with early-stage colorectal carcinoma who suffered a second cancer have significantly worse survival outcomes if treated with hypolipidemic drugs. Therefore, screening for second primary malignancies in colorectal cancer survivors treated with hypolipidemic drugs should be done more cautiously, especially for gastric and bladder cancer.

Furthermore, the research indicates that the progression of HNSCC may be influenced by the support of CAFs in the tumor microenvironment, and the heterogeneity of this disease may be attributed to the level of cooperation between CAFs and epithelial cancer cells.

Chromosomal damage in occupationally exposed health professionals assessed by two cytogenetic methods

Summary: This study investigates the perioperative kinetics of D-lactate and ischemia-modified albumin (IMA) and their predictive value in early diagnosis of acute intestinal ischemia. The results suggest that serum levels of IMA and D-lactate can serve as influential predictive markers for postoperative intestinal ischemia, especially after 3 hours from complete declamping of vascular reconstruction.

Summary: This study aims to explore the processes and principles of embedding reference centers (RCs) for rare cancers in national health systems. The results show heterogeneity in the formalization of RCs' status and their integration in different health systems, with two models (center-based and network-based) envisioned at the national level. Expert clinicians in some countries cooperate in a structured way using network mechanisms and provide a referral system to facilitate access to treatment for patients from non-expert centers.

Involvement of Small Non-Coding RNA and Cell Antigens in Pathogenesis of Extramedullary Multiple Myeloma

Summary: In this study, antigens and miRNAs involved in the pathogenesis of EMD were identified using flow cytometry and next-generation sequencing. MiR-26a-5p and miR-30e-5p were found to be down-regulated in EMD. Aberrant miRNAs associated with cell cycle regulation, proteolysis, and infection or neurological disorder-related signaling pathways were observed. Additionally, clinicopathological characteristics and CD antigens associated with MM and EMD prognosis were identified.
